- Alkaline phosphatase activities and the release of orthophosphate from endogenous substrates by these enzymes were measured in waters from two commercial fishponds in the watershed area of Lake Kinneret (Northern Israel). These data were compared with results from the lake at seasons of adequate or limited phosphorus supply. In the fishponds, high Relative Phosphatase Activity ratios (>2) and relatively large amounts of orthophosphate extracted from plankton by autoclaving (average 27% of readily available phosphorus) indicated adequate, or even excess, levels of phosphorus availability despite elevated pond productivity (2 to 3 tons carp/ha/yr). Therefore, we suggest that decreasing routine phosphorus fertilization of these ponds would not affect overall productivity but would eventually lower the amounts of phosphorus reaching Lake Kinneret.
- In general, the R. P. A. ratio may be a useful index to evaluate phosphorus availability for a wide range of natural waters. Values for this ratio of <1 and >2 appear indicative of limited or adequate phosphorus availability respectively.
- Three sources of orthophosphate, (Pi), readily available to phytoplankton, are indicated: (1) enzymatically released Pi, (2) Pi in intracellular pools and (3) Pi initially present in the water. Although the first source is always important, relatively greater amounts of Pi are contributed by the other fractions in situations of plentiful phosphorus availability.
- Activity of free dissolved phosphatases was found in filtered samples of fishpond water. However, neither these enzymes or added phosphatases released significant amounts of Pi from the dissolved organic phosphorus compounds in the filtered water.

This paper reports the first Raman spectroscopic study of the potassium complex of the cation-specific antibiotic valinomycin. Complete Raman spectra (140 to 3600 cm?1) of crystalline valinomycin-KSCN and its CCl4, CHCl3 and C2H5OH solutions are presented and used to probe the structure of the complex in these environments. In all cases a single, narrow peak is observed in the ester CO stretch region (1750 to 1775 cm?1) which contrasts strongly with the broad bands observed in solutions of uncomplexed valinomycin. This is consistent with the presence of a single conformation in which all six ester CO groups co-ordinate an enclosed potassium ion. We find that although the ester CO stretch frequencies of the complex are similar in the solid state and in non-polar solution (~1770 cm?1) they are considerably different in the presence of polar solvents (~1756 cm?1); this may indicate that the complexed potassium ion is still free to interact with nearby solvent ions (and possibly its counterion) through gaps in the hydrophobic “shield” provided by the hydrocarbon residues of valinomycin. In contrast the amide CO frequencies of the complex (~1650 cm?1) are solvent-independent. Summary A proteolipidic toxin, prymnesin, when added to the aqueous solutions around thin lipid membranes causes a marked increase in membrane conductance. The toxin-treated membrane is cation-permselective. The extent of cation permselectivity is dependent upon ionic strength of the aqueous solutions in a fashion similar to the dependence of cation permselectivity of a cation exchanger containing about 100mm of fixed negative sites. Dose-response relationship studies reveal a linear relation between log prymnesin concentration and log membrane conductance. The slope of the curve is around 3 if the toxin is applied to one side of the membrane and is around 7 if the toxin is applied to both sides of the membrane. The membrane treated with toxin on one side only is clearly asymmetric in its properties. These characteristics are expressed by an asymmetric current-voltage relationship, and by asymmetric sensitivity of membrane conductance to pH and to salt concentration. The conductance of the toxin-treated membrane is inversely proportional to temperature. It is suggested that aggregates of toxin moieties assemble in the membrane to form negatively charged aqueous pores. There is roughly a good correlation between the increase in membrane conductance and the increase in membrane permeability to urea if both were attributed to the formation of aqueous channels in the membrane. 相似文献

The richness of biodiversity in the tropics compared to high‐latitude parts of the world forms one of the most globally conspicuous patterns in biology, and yet few hypotheses aim to explain this phenomenon in terms of explicit microevolutionary mechanisms of speciation and extinction. We link population genetic processes of selection and adaptation to speciation and extinction by way of their interaction with environmental factors to drive global scale macroecological patterns. High‐latitude regions are both cradle and grave with respect to species diversification. In particular, we point to a conceptual equivalence of “environmental harshness” and “hard selection” as eco‐evolutionary drivers of local adaptation and ecological speciation. By describing how ecological speciation likely occurs more readily at high latitudes, with such nascent species especially prone to extinction by fusion, we derive the ephemeral ecological speciation hypothesis as an integrative mechanistic explanation for latitudinal gradients in species turnover and the net accumulation of biodiversity. 相似文献

We have used UV resonance Raman spectroscopy to study the acid-induced denaturation of horse apomyoglobin (apoMb) between pH 7. 0 and 1.8. The 206.5 nm excited Raman spectra are dominated by amide vibrations, which are used to quantitatively determine the apoMb secondary structure. The 229 nm excited Raman spectra are dominated by the Tyr and Trp Raman bands, which are analyzed to examine changes of Tyr and Trp environments and solvent exposures. We observe two partially unfolded apoMb intermediates at pH 4 and pH 2, while we observe only one partially unfolded holoMb intermediate at 2, in which the G and H helices are mainly intact, while the rest of protein is unfolded. This partially unfolded holoMb intermediate at pH 2 is essentially identical to the pH 2 apoMb intermediate. The partially unfolded pH 4 apoMb intermediate is composed of the three folded A, G, and H helices and contains 38% helical structure. The changes in the Trp Raman cross sections during the acid-induced denaturation indicates that Trp 7 is likely to be fully exposed in the apoMb pH 4 intermediate and that the A helix melts with a pKa approximately 3.5. 相似文献
