Summary A microdensitometer system for point measurements in areas down to a few square micrometers is described. The detector system is connected to an integrating digital voltmeter, constituting a very sensitive system. The digital output signals and a digital identification are registered on a recorder and on a tape punch, thus facilitating computer analysis.The precision of the equipment varied from 1.1 per cent, for low signal levels, to 0.3 percent for the highest signal levels. Stray light, the only important source of systematic errors, was found to influence the output signal level. Concerning determinations of the densities, however, a variation of the order of 1 per cent was only obtained for small areas with densities differing from that of the background.  相似文献

Summary A new histochemically demonstrable thiaminepyrophosphatase activity in liver tissue is described. It appears to be localized solely to the cytosomes of the sinusoidal endothelium. The activity, which requires high concentration of lead ion for its demonstration, has a high degree of substrate specificity. It resists formaldehyde fixation, ethanol and many inhibitors. Summary Histochemically demonstrable Golgi-associated TDP-ase activity in liver cells from cat, chicken, rat and frog has been investigated.This activity is highly substrate-specific, insensitive to aldehyde fixation, ethanol, acetate, lead and most enzyme inhibitors. It is stimulated by divalent manganese, calcium, magnesium and cobalt and optimum pH is at pH 6 to 7.The characteristics are identical for all four species but significant differences exist at a comparison with bile canalicular activity, Golgi-associated activity in other cells and biochemical findings.This study was supported in part by a grant from the Swedish Medical Research Council.  相似文献

Summary By means of a histochemical method adapted for electron microscopy a cholinesterase in body muscle cells of plaice (Pleuronectes platessa) has been localized to the sarcolemma. The cholinesterase activity disappeared from the sarcolemma after the muscle tissue had been incubated with a bacterial enzyme, which had earlier been shown, by biochemical methods, to be able to liberate this cholinesterase activity from plaice muscle.The provision of live plaices from Kristineberg Zoological Station, Fiskebäckskil, Sweden, is gratefully acknowledged. A simple method for the production and preparation of hyaluronic acid (HA) is described. Three media were tested, which all supported relatively good yields of cells, but only in one of them were appreciable amounts of HA formed. In this medium, HA was produced during the logarithmic growth phase and showed a molecular weight of 7.3 × 105. HA prepared from 24-hr cultures of the same organism had a molecular weight of only 2.6 × 105. The kinetics of HA production in batch culture were studied, as well as of cell production and glucose consumption.  相似文献
