Most of the studies carried out in the past on economically important fish species rely on single species approach. Ecosystem dynamics are characterized by complex interaction among species, sharing common habitat needs and thus forming characteristic assemblages. The analysis of spatio-temporal variability of fish community, coupled to the analysis of spatial indices, provides a synthetic view of the fish community status evidencing, if any, the way a community changes. Such considerations drive also to the development of ecosystem-based fishery management paradigm. In the present study changes in pelagic fish community structure in an upwelling ecosystem of the central Mediterranean Sea during the last 10 years was analysed, by focusing the attention on the five most abundant small pelagic species: Engraulis encrasicolus, Sardina pilchardus, Sardinella aurita, Trachurus trachurus and Boops boops. Our results evidenced a quite stable community structure, characterized by spatial occupation strongly driven by ecosystem characteristics and modulated according to specie-specific behaviour. Obtained results lead us to hypothesize that the observed stability of community could be linked to the presence of different environments leading to efficient space partitioning and resources utilization among species.  相似文献

Cryobanking somatic foetal cells acquire much relevance in endangered species for biodiversity conservation purposes. Such cells could be later used to reintroduce the lost genes into the breeding pool, by inducing pluripotency and/or nuclear transfer if necessary. Since requirements for preserving foetal cells are not always the same as for adult ones, we evaluated the cryosensitivity of foetal skin cells in comparison with adult ones from the critically endangered Iberian lynx. Responses to cryoinjury were analyzed in both thawed cell types by means of cell viability and functionality (by analyzing their membrane integrity, metabolic activity, glycosaminoglycan content and proliferative activity). Freezing media included the permeating cryoprotectant Me2SO, either alone or along with the non-permeating cryoprotectant sucrose at 0.1 or 0.2 M. When Me2SO was the only cryoprotectant, survival rate fell in thawed foetal cells to 54 ± 4% (against 89 ± 6% for thawed adult ones) and both proliferative and metabolic activities remained significantly lower than values for thawed adult cells. However, the combination of sucrose (both 0.1 as 0.2) and Me2SO in foetal cells significantly increased their survival rates (to 71 ± 4% and 73 ± 5%, respectively), proliferative activities (partially at day 7 and completely at day 14 after thawing) and metabolic activities.  相似文献

A proper assessment of the foraging habits of the Guadalupe fur seal (GFS; Arctocephalus townsendi) is a priority to better understand its recovery, in which the potential for intraspecific competition for prey and space resources is expected to lead to segregation. This study aimed to determine the foraging habits of different sex and age classes. A total of 146 GFS fur samples was collected at Guadalupe Island, Mexico (2014–2020) for stable isotopes (δ13C and δ15N) analysis. Isotopic areas were created (SIBER package in R). Significant isotopic differences were observed between classes. Male (3.6‰2) and female (3.0‰2) juveniles had the largest isotopic areas due to a greater foraging dispersion. Adult females showed the lowest mean δ15N value (16.1‰ ± 0.5‰) due to foraging trips that are mostly performed towards high latitudes. Except for pups, adult and subadult males presented the highest mean δ15N (17.4‰ ± 0.4‰) and δ13C values (−17.0‰ ± 0.8‰) due to a possible higher trophic level and coastal foraging habits, whereas pups presented the highest mean δ15N value (17.6‰ ± 0.3‰) because of lactation, which reflects their mothers δ15N signal plus their own enrichment. Our findings suggest a segregation explained by differences in life history, energy requirements, and a possible strategy to avoid competition.  相似文献

Understanding patterns and processes in biological diversity is a critical task given current and rapid environmental change. Such knowledge is even more essential when the taxa under consideration are important ecological and evolutionary models. One of these cases is the monogonont rotifer cryptic species complex Brachionus plicatilis, which is by far the most extensively studied group of rotifers, is widely used in aquaculture, and is known to host a large amount of unresolved diversity. Here we collate a dataset of previously available and newly generated sequences of COI and ITS1 for 1273 isolates of the B. plicatilis complex and apply three approaches in DNA taxonomy (i.e. ABGD, PTP, and GMYC) to identify and provide support for the existence of 15 species within the complex. We used these results to explore phylogenetic signal in morphometric and ecological traits, and to understand correlation among the traits using phylogenetic comparative models. Our results support niche conservatism for some traits (e.g. body length) and phylogenetic plasticity for others (e.g. genome size).  相似文献

We used a response-surface analysis to determine the importance of different factors affecting the resting routine metabolic rate (QO2) of the white shrimp Litopenaeus vannamei. The oxygen consumption rates were estimated using a multi-factorial design with 28 combinations of different salinities (15, 20, 25, 30, 35, 40 and 45 psu) and temperature (20, 25, 30 and 35 °C) values. The response-surface analysis produced a quadratic function showing that temperature more profoundly affects the oxygen consumption rate. Response-surface curves were generated to predict the optimal conditions resulting in oxygen consumption to better understand the successful growth of this species.  相似文献

Twenty species of shrubbyOxalis species native in South America were arranged in six groups according to their karyotype similarities. The first group includes more than half of the species studied, all with 2n = 12 relatively small metacentric chromosomes. The rest of the species from the other groups has different basic chromosome numbers, and exhibits various degrees of chromosome asymmetry and size increase. These latter groups apparently have specialized to habitats with low light intensity such as the forest undergrowth in contrast to the frist group whose members live in open areas (i.e., grasslands, savannas etc.).  相似文献

The fungus Cryptococcus neoformans possesses a polysaccharide capsule and can form biofilms on medical devices. We describe the characteristics of C. neoformans biofilm development using a microtiter plate model, microscopic examinations, and a colorimetric 2,3-bis(2-methoxy-4-nitro-5-sulfophenyl)-5-[(phenylamino) carbonyl]-2H-tetrazolium-hydroxide (XTT) reduction assay to observe the metabolic activity of cryptococci within a biofilm. A strong correlation between XTT and CFU assays was demonstrated. Chemical analysis of the exopolymeric material revealed sugar composition consisting predominantly of xylose, mannose, and glucose, indicating the presence of other polysaccharides in addition to glucurunoxylomannan. Biofilm formation was affected by surface support differences, conditioning films on the surface, characteristics of the medium, and properties of the microbial cell. A specific antibody to the capsular polysaccharide of this fungus was used to stain the extracellular polysaccharide matrix of the fungal biofilms using light and confocal microscopy. Additionally, the susceptibility of C. neoformans biofilms and planktonic cells to environmental stress was investigated using XTT reduction and CFU assays. Biofilms were less susceptible to heat, cold, and UV light exposition than their planktonic counterparts. Our findings demonstrate that fungal biofilm formation is dependent on support surface characteristics and that growth in the biofilm state makes fungal cells less susceptible to potential environmental stresses.  相似文献

Following incubation with ATP and a cAMP-dependent protein kinase under optimal conditions of lipid acceptor, phospholipid and metal ion requirements, the transfer activity of partially purified dolichol phosphate mannose synthase (DPMS) increased about 60% and this activation correlated with a 50% increase in V(max) with no alteration in the apparent K(m) for GDP-Manose. Phosphorylation with [gamma-(32)P]ATP resulted in the labeling of several polypeptides, one of which exhibited the molecular weight of the enzyme (30 kDa) and was also recognized using a specific anti-DPMS monoclonal antibody. This and the fact that the phosphate label could be removed by an alkaline phosphatase indicate that Candida DPMS may be regulated by phosphorylation-dephosphorylation, a mechanism that has been proposed for the enzyme in other organisms.  相似文献

For both pathogenic fungi and bacteria, extracellular vesicles have been shown to contain many microbial components associated with virulence, suggesting a role in pathogenesis. However, there are many unresolved issues regarding vesicle synthesis and stability, including the fact that vesicular packaging for extracellular factors involved in virulence must also have a mechanism for vesicle unloading. Consequently, we studied the kinetics of vesicle production and stability using [1-(14) C] palmitic acid metabolic labelling and dynamic light scattering techniques. Cryptococcus neoformans vesicles were produced throughout all stages of fungal culture growth and they were stable once isolated. Density gradient analysis revealed that only a portion of the vesicle population carried cryptococcal polysaccharide, implying heterogeneity in vesicular cargo. Vesicle incubation with macrophages resulted in rapid vesicle instability, a phenomenon that was ultimately associated with serum albumin. Additionally, albumin, along with mouse serum and murine immunoglobulin destabilized Bacillus anthracis vesicles, but the effect was not observed with ovalbumin or keyhole limpet haemocyanin, demonstrating that this phenomenon is neither host-, microbe- nor protein-specific. Our findings strongly suggest that cryptococcal vesicles are short-lived in vivo and vesicle destabilization is mediated by albumin. The ability of albumin to promote vesicular offload through destabilization indicates a new activity for this abundant serum protein.  相似文献
