The influence of UV-light (240-390 nm) at a dose of 151 and 755 J/m2 on the functional properties of lymphocyte metabolism key enzymes from donors' human blood: lactate dehydrogenase (LDH), cytochrome c oxidase, succinate dehydrogenase (SDH), Ca2(+)-ATPase of plasma membranes has been investigated. It has been revealed that photoinactivation of enzymes immediately after UV-irradiation which leads to the decrease of the ATP content in lymphocytes is replaced by the increased activity of the enzymes under investigation during daily incubation of lymphocytes. As a result, the level of ATP in photo-modified lymphocytes does not differ from that in native cells before incubation. This indicates the normalization of biochemical processes in lymphocytes influenced by UV-light applied in autotransfusion of UV-irradiated blood.  相似文献

The purpose of the present work is to study the vacuum ultraviolet radiation action on the optical and chromatographic characteristics of human hemoglobin molecules and their components - haem and globin. Using the methods of spectrophotometry and thin layer chromatography (TLC), we have investigated into the structural changes of molecules of human hemoglobin, haem and globin, induced by the influence of vacuum UV light (gamma = 118-134 nm, dose - 1.2 kJ/m2). It has been shown that vacuum ultraviolet radiation induces an infringement of the higher types of the spatial organization ofglobin molecules, thus leading to the changes in the structural state of the albuminous globule.  相似文献

The kinetics of photoinactivation of cardiac (H4) and muscular (M4) isoforms of lactate dehydrogenase irradiated by UV light (240-390 nm) in the free form and in the presence of sodium azide, D-mannitol, and serotonin was studied. It was shown that the decrease in the catalytic activity of both enzymes can be described by the kinetics of the first-order monomolecular reaction. The inactivation rate constant of lactate dehydrogenase M4 is considerably higher than that of lactate dehydrogenase H4, indicating a greater photochemical lability of the isoform M4. It was shown that sodium azide has a different protective action on the proteins studied. The irradiation of the muscular isoform in the presence of serotonin and D-mannitol did not change the character of the "dose-effect" curve and only led to a decrease in the photoinactivation rate constant of the protein.  相似文献

The examination of cytogenetic characteristics (the mitotic activity, the time of passing of the mitotic stages by cells, the level and the spectrum of mitosis disturbances, the nucleoli characteristics) of seed progeny of common oak trees, exposed to the radiation exposure after the accident at the Chernobyl Nuclear Power Station and growing at the territories with different levels of anthropogenic contamination was held. In spite of the normalization of the radioactive level at the examined territories the changes of the cytogenetic characteristics of common oak seed progeny in comparison with the control were noticed. Obtained results are explained from the position of the radiation induced genomic instable and of the wave kinetic of the mutation process. The questions of the advisability and of the validity of using of the seed progeny of the common oak trees, exposed to the radiation exposure, in forest restoration measures are discussed.  相似文献

Photoinduced changes of human ceruloplasmin (hCp) under laser and ultraviolet (UV) radiation have been studied. A complex character of hCp spectral modifications points to a different degree of the protein molecule folding. After the influence of UV radiation (240-390 nm) in a dose range 906-4530 J/m2 the decrease of oxidase hCp activity was registered. These changes are thought to be connected with modification of the conformation of hCp active centre or copper reduction that was confirmed by the decrease of optical density at 610 nm. An inactivation constant (kf = 3.8 x 10(-4) 1/s) and a photoinactivation quantum yield (1.4 x 10(-4)) were calculated. Kinetic dependencies of o-phenilen-diamine oxidation by intact and UV-modified glycoprotein do not correspond to Michaelis-Menten equation. UV radiation in a dose range 151-4530 J/m2 induced increasing of ceruloplasmin superoxiddismutase activity. The influence of He-Ne laser radiation (1.3 mW; lambda = 632.8 nm, exposure time from 1 up to 30 min) leads to the raise of the hCp functional activity; superoxiddismutase activity changes more significantly. Possible photophysical and photochemical processes in hCp molecule, leading to photomodification and changes of protein functional activity have been represented on the scheme.  相似文献

The dynamics of structural and functional changes proceeding in a peroxidase molecule under the effect of temperature was studied. It was shown that peroxidase thermoinactivation proceeds in two consequent stages. Based on the analysis of enzyme peroxidase and oxidase activity and peroxidase spectral and buffer characteristics, it was established that at temperatures from 20 to 55 degrees C reversible conformation there occur changes of the hemoprotein molecule related to consequent unfolding and folding of the protein globule. The influence of temperature of 60 degrees C and above induces the protein globule unfolding and loosing of peroxidase activity.  相似文献

The work deals with the development of the rapid method of the identification of acute streptococcal infection on the basis of the coagglutination test. The rapid method of the extraction of group-specific polysaccharide antigen from the cell walls of group A streptococci is proposed. The data on the use of native sera and their fractions in the development of coagglutination diagnostica have been described and analyzed. The advantages of the new method of the diagnosis of acute streptococcal infection in comparison with the traditional microbiological method are shown.  相似文献

The influence of a wide range of doses of UV-light (240-390 nm) on the expression level of receptor complex molecules (CD3, CD4, CD8 markers) on the membrane surface of human blood T-lymphocytes has been studied using flow cytometry and enzyme linked immunosorbent assay. UV-light at small and medium doses (15 1, 453 and 906 J/m2) has been established to have a unidirectional (activating) effect on the expression level of receptor complex molecules, and at a high dose of 1359 J/m2 it can either increase (CD4 and CD8 markers) or reduce (CD3 complexes) the quantity of the analyzed molecules on the membrane surface of T-lymphocytes.  相似文献
