Infection of hepatocytes by Plasmodium falciparum sporozoites requires the host tetraspanin CD81. CD81 is also predicted to be a coreceptor, along with scavenger receptor BI (SR-BI), for hepatitis C virus. Using SR-BI-knockout, SR-BI-hypomorphic and SR-BI-transgenic primary hepatocytes, as well as specific SR-BI-blocking antibodies, we demonstrate that SR-BI significantly boosts hepatocyte permissiveness to P. falciparum, P. yoelii, and P. berghei entry and promotes parasite development. We show that SR-BI, but not the low-density lipoprotein receptor, acts as a major cholesterol provider that enhances Plasmodium infection. SR-BI regulates the organization of CD81 at the plasma membrane, mediating an arrangement that is highly permissive to penetration by sporozoites. Concomitantly, SR-BI upregulates the expression of the liver fatty-acid carrier L-FABP, a protein implicated in Plasmodium liver-stage maturation. These findings establish the mechanistic basis of the CD81-dependent Plasmodium sporozoite invasion pathway.  相似文献

Helix aspersa hibernates in Brittany (western France), where it may experience subzero temperatures. Studies on cold hardiness, although scarce in land snails, have shown a seasonal variation in supercooling ability, associated with high temperatures of crystallization (Tc). In the present work, two key environmental factors, temperature and photoperiod, were studied to elucidate, how they may affect the enhancement of supercooling ability in the snails from the end of summer to winter. Nine groups of adult snails were acclimated to different combinations of photoperiod (LD-16:8, LD-12:12, and LD-8:16 h) and temperature (15, 10, and 5°C). Temperature of crystallization, hemolymph osmolality, and water content were measured. The results demonstrate a significant effect of the photoperiod on Tc, i.e., shorter photoperiods induce lower Tc (LD-16:8 h, mean Tc = −3.0°C, SD = 2.0; LD-12:12 h, mean Tc = −4.3°C, SD = 1.9; LD-8:16 h, mean Tc = −5.2°C, SD = 1.9; n = 90), whereas the acclimation temperature had no effect. Measurements of hemolymph osmolality and water content showed that osmolality is negatively correlated with water content. Mechanisms such as dehydration are involved in the decrease of Tc. A declining photoperiod triggers a lower Tc, long before the onset of winter conditions. This response may have an adaptive component, allowing individuals to cope with the mild winters typically observed in oceanic regions.  相似文献

In the same way that cry genes, coding for larvicidal delta endotoxins, constitute a large and diverse gene family, the cyt genes for hemolytic toxins seem to compose another set of highly related genes in Bacillus thuringiensis. Although the occurrence of Cyt hemolytic factors in B. thuringiensis has been typically associated with mosquitocidal strains, we have recently shown that cyt genes are also present in strains with different pathotypes; this is the case for the morrisoni subspecies, which includes strains biologically active against dipteran, lepidopteran, and coleopteran larvae. In addition, while one Cyt type of protein has been described in all of the mosquitocidal strains studied so far, the present study confirms that at least two Cyt toxins coexist in the more toxic antidipteran strains, such as B. thuringiensis subsp. israelensis and subsp. morrisoni PG14, and that this could also be the case for many others. In fact, PCR screening and Western blot analysis of 50 B. thuringiensis strains revealed that cyt2-related genes are present in all strains with known antidipteran activity, as well as in some others with different or unknown host ranges. Partial DNA sequences for several of these genes were determined, and protein sequence alignments revealed a high degree of conservation of the structural domains. These findings point to an important biological role for Cyt toxins in the final in vivo toxic activity of many B. thuringiensis strains.  相似文献

The interaction of two cytolytic toxins, Cyt1Ab from Bacillus thuringiensis subsp. medellin and Cyt2Ba from Bacillus thuringiensis subsp. israelensis, with Bacillus sphaericus was evaluated against susceptible and resistant Culex quinquefasciatus and the nonsensitive species Aedes aegypti. Mixtures of B. sphaericus with either cytolytic toxin were synergistic, and B. sphaericus resistance in C. quinquefasciatus was suppressed from >17,000- to 2-fold with a 3:1 mixture of B. sphaericus and Cyt1Ab. This trait may prove useful for combating insecticide resistance and for improving the activity of microbial insecticides.  相似文献

The cerebrospinal fluid (CSF) of multiple sclerosis (MS) patients contains a 17 kDa glycoproteic factor with gliotoxic properties in vitro. In order to study the physiopathological role of this gliotoxic factor in vivo, we have injected a partially purified preparation and appropriate controls in rat CSF and investigated whether it induces cell death in the rat central nervous system (CNS), 10 days and 3 months after injection. We used the TUNEL assay in association with specific immunohistochemistry to characterize dying cells in the gliotoxic factor- treated rat CNS. At 10 days post-injection, TUNEL-positive cells were observed in the whole rat CNS. They were particularly numerous in the choroid plexus, ependymal epithelium, cerebral white matter, cerebral vascular endothelium, arachnoid spaces and less frequent in the gray matter of brain and spinal cord. The predominant type of TUNEL-positive cells observed at 10 days post-injection was astrocytes, in white matter, gray matter, occasionnally around damaged endothelial cells in periventricular and subpial spaces. Other TUNEL-positive cells were identified as oligodendrocytes by an oligodendrocyte specific RIP immunostaining, at 10 days post-treatment with the gliotoxic factor. Interestingly, demyelination and death of oligodendrocytes were more important 3 months post-injection: TUNEL-RIP positive oligodendrocytes were generally associated with multifocal demyelinating areas. Clearly, the 17 kDa gliotoxic factor injection in rat CSF triggers demyelination and may be used as a new animal model for MS. Also, our results suggest a new possible scenario for MS pathogenesis: death of oligodendrocytes and astrocytes, stimulated by the MS gliotoxic factor causes the breakdown of the blood-brain barrier (BBB) and the demyelinating cascade.  相似文献

In heterogeneous habitats with limited resources, spacing behaviour will affect individual variation in breeding success and density of populations, and is thus of general interest to ecologists. We investigated how red squirrels Sciurus vulgaris adapt their social organisation to tine‐grained heterogeneity in habitat quality, studying spacing behaviour, habitat use and population dynamics in a forest in north Italy, characterised by a mosaic of high‐quality (chestnut‐pine) and poor‐quality patches. We compared the data with those from more homogeneous broadleaf and mixed woodlands with similar overall tree seed abundance (“stable” habitats). Squirrels lived at lower densities (pre‐breeding density 0.39‐0.58 ha‐1) than in “stable” habitats, although breeding rate was not reduced. Female breeding success was related to being primiparous as yearlings, and increased with body mass and proportion high‐quality habitat in the home range. Persistency rate of females was as in stable habitats, It was higher than male persistency, but immigration and recruitment rates were male biased, resulting in even sex‐ratio. All residents occupied high‐quality patches, and no subadults established a permanent home range in poor‐quality habitat. Home range and core‐area size was typically larger in males than in females and a male's core‐area overlapped those of other males and of females. Female core‐areas were overlapped by males but not. or very little, by other females (intrasexual territoriality). Home ranges, or core‐areas, were not smaller than in “stable” habitats, nor did we find a higher degree of core‐area overlap. We conclude that in patchy habitats dominant, resident red squirrels exclude dispersing animals from preferred, high‐quality habitat, producing a spacing pattern referred to as ideal despotic distribution, and that poor patches were only used temporarily by transient individuals, resulting in a reduction of density in comparison to populations in “stable” habitats.  相似文献
