A genomic library of partially EcoRI-digested DNA from the lesser snow goose, Anser caerulescens caerulescens, was constructed in the phage vector Charon 4. Phage containing only unique sequences were identified by screening plaques with 32P-labeled genomic DNA. Restriction-fragment- length polymorphisms (RFLPs) were identified by probing DNA from 11-13 male birds from the breeding colony at La Perouse Bay. Of the 17 probes examined, all detected RFLPs with at least one of EcoRi, HindIII, Msp1, and Taq1. Several of them identified highly variable regions with multiple alleles. These RFLPs are valuable DNA markers that can be used for (1) the examination of DNA variation, relatedness, and genetic distance and (2) assessing paternity and maternity. These data suggest that there are higher levels of variation of DNA sequence in birds than had previously been thought to exist.   相似文献

Belonging to sect.Scandentes, it has passed asD. tiliifolia Lam., but is not closely related to that or to any other Meso-american species. The protein hPot1 shares homology with telomere-binding proteins in lower eukaryotes and associates with single-stranded telomeric DNA in vitro as well as colocalizing with telomere-binding proteins in vivo. We now show that hPot1 is coimmunoprecipitated with telomeric DNA and that stable expression of this protein in telomerase-positive cells results in telomere elongation, supporting the idea that hPot1 is a bona fide mammalian telomere-binding protein. We previously found that mutations in the N-terminal DAT domain of the hTERT catalytic subunit of telomerase rendered the enzyme catalytically active but unable to elongate telomeres in vivo. This phenotype could be partially rescued by fusion with the double-stranded telomeric protein hTRF2. Given that hPot1 binds to single-stranded DNA in vitro (at the same site that hTERT binds to in vivo), we addressed whether fusion of hPot1 can rescue the DAT mutations more efficiently than that of hTRF2. We now report that a DAT mutant of hTERT is indeed efficiently rescued upon fusion to hPot1. However, this rescue depended on the ability of hPot1 to localize to telomeres rather than binding to DNA per se. These data support a model whereby the DAT domain of hTERT is implicated in telomere-telomerase associations.  相似文献

Activation of phospholipase C-dependent inositol polyphosphate signaling pathways generates distinct messengers derived from inositol 1,4,5-trisphosphate that control gene expression and mRNA export. Here we report the regulation of telomere length by production of a diphosphorylinositol tetrakisphosphate, PP-IP4, synthesized by the KCS1 gene product. Loss of PP-IP4 production results in lengthening of telomeres, whereas overproduction leads to their shortening. This effect requires the presence of Tel1, the yeast homologue of ATM, the protein mutated in the human disease ataxia telangiectasia. Our data provide in vivo evidence of a regulatory link between inositol polyphosphate signaling and the checkpoint kinase family and describe a third nuclear process modulated by phospholipase C activation.  相似文献

Synopsis Several North American minnow species are known to engage in the action pattern parallel swimming, an activity that allows males competing for nests or nesting sites an opportunity to assess each other. These agonistic displays have been hypothesized to occur only between males of similar size. We provide evidence to support this hypothesis for two minnow species and a sucker species. Competition is strong both intra- and interspecifically for nest sites and we document the occurrence of interspecific parallel swims between three species.  相似文献

Members of at least two unrelated genera of plants, Dalechampia (Euphorbiaceae) and Clusia (Guttiferae), attract pollinators by secreting resins from floral structures. Bees that pollinate these flowers collect and use the resin in nest construction; these include Euglossa, Eulaema, Eufriesea, Trígona and, Hypanthidium in the neotropics, and Heriades in Africa. Floral resins are slow in hardening as compared with many other plant resins; this facilitates collection and storage by bees, hence probably enhancing the attractiveness of the flowers. Floral resins are probably of great utility and dependability and may be an especially important resource to certain bees, which in turn are major pollinators of many species of tropical plants. Bees appear to forage floral resin using a “strategy” of energy efficiency; large bees collect resin only from copious sources, smaller bees collect resin from sources with small amounts as well as from sources with large amounts of resin. Resin secretion in flowers may have originated as defense against herbivores and secondarily assumed the role of pollinator reward.  相似文献

The pollination ecology of four Dalechampia species was studied in three areas in northern Natal, South Africa. All species were pollinated by resin- and/or pollen-collecting megachilid bees (Hymenoptera: Megachilidae). The most common of these at all study sites was Heriades sp. (Megachilini), which collected both pollen and resin. This bee was the primary pollinator of D. galpinii and D. volubilis, and a secondary pollinator of D. aff. parvifolia and D. capensis. The primary pollinators of these latter two Dalechampia were Pachyanthidium near cucullatum and P. cordatum (Anthididiini), respectively. These are larger bees that collected mainly resin. Other visitors varied among Dalechampia species and among sites. Pairs of Dalechampia species were often found in sympatry (within 30 m of each other) and shared pollinators to varying extents. “Transfer experiments,” in which we placed inflorescences of two Dalechampia species together, reinforced observations of flower discrimination by bees foraging among naturally occurring sympatric pairs. Heriades sp. showed no obvious discrimination between Dalechampia species, but Pachyanthidium spp. “preferred” the Dalechampia species with the greater resin reward. Occasionally, however, Pachyanthidium would visit less rewarding species. Thus, although sympatric African Dalechampia species showed some pollinator partitioning, it was much weaker than found among New World species, and cannot alone explain the general absence of Dalechampia hybrids in northern Natal.  相似文献

Using cloned Epstein-Barr nuclear antigen 1 (EBNA) and oriP elements from the Epstein-Barr virus (EBV) in conjunction with liver-specific growth media, we have constructed an EBNA-producing line of well-differentiated human hepatoma cells (Hep-EBNA-2) and appropriate EBV-oriP vectors. These vectors, pBEDC1 and pBEUG1, were maintained as free extrachromosomal elements only in cells that expressed the trans-acting EBNA protein. They were readily rescued from transfected Hep-EBNA-2 cells upon transformation of recA- Escherichia coli with cellular low-Mr DNA. They are true shuttle vectors in that they can propagate as free closed circular elements in both human Hep-EBNA-2 cells and E. coli. Finally, we have demonstrated the vector capability of our shuttle system by inserting into the SV40 expression cassette of pBEUG1 a large full-length cDNA encoding coagulation factor VIII. Our data clearly show that EBV-oriP episomes are able to stably propagate in an hepatic background and that neither high levels of EBNA protein nor multiple copy episomes significantly interfere with the expression of the set of hepatic functions that have been analyzed. These results are discussed in terms of gene amplification and cloning of genes that program liver differentiation.  相似文献

We examined the mandibles of 377 individuals representing 25 species, 12 genera, 5 tribes, and 2 subfamilies of the Loricariidae, a species‐rich radiation of detritivorous–herbivorous neotropical freshwater fishes distinguished by having a ventral oral disk and jaws specialized for surface attachment and benthic feeding. Loricariid mandibles are transversely oriented and bilaterally independent, each rotating predominantly around its long axis, although rotational axes likely vary with mandibular geometry. On each mandible, we measured three traditional and three novel morphological parameters chosen primarily for their functional relevance. Five parameters were linear distances and three of these were analogous to traditional teleost in‐ and out‐levers for mandibular adduction. The sixth parameter was insertion area of the combined adductor mandibulae muscle (AMarea), which correlated with adductor mandibulae volume across a subset of taxa and is interpreted as being proportional to maximum force deliverable to the mandible. Multivariate analysis revealed distributions of phylogenetically diagnosed taxonomic groupings in mandibular morphospace that are consistent with an evolutionary pattern of basal niche conservatism giving rise to multiple adaptive radiations within nested clades. Correspondence between mandibular geometry and function was explored using a 3D model of spatial relationships among measured parameters, potential forces, and axes of rotation. By combining the model with known loricariid jaw kinematics, we developed explicit hypotheses for how individual parameters might relate to each other during kinesis. We hypothesize that the ratio [AMarea/tooth row length2] predicts interspecific variation in the magnitude of force entering the mandible per unit of substrate contacted during feeding. Other newly proposed metrics are hypothesized to predict variation in aspects of mandibular mechanical advantage that may be specific to Loricariidae and perhaps shared with other herbivorous and detritivorous fishes. 2011. © 2011Wiley Periodicals, Inc.  相似文献
