Three possible hypotheses could explain the polarity of the histological features of basal archosauriform and archosauromorph reptiles: either, the fibrolamellar complex is basal; or, the lamellar-zonal complex is basal or finally, the condition varied, and each complex evolved more than once in these early groups. The answer to this question would have broad implications for our understanding of the physiological, ecological, and behavioral features of the first archosaurs. To this end, we sampled the bone histology of various archosauriforms and basal archosaurs from the Triassic and Lower Jurassic: erythrosuchids, proterochampsids, euparkeriids, and basal ornithischian dinosaurs, including forms close to the origin of archosaurs but poorly assessed phylogenetically. The new data suggest that the possibility of reaching and maintaining very high growth rates through ontogeny could have been a basal characteristic of archosauriforms. This was partly retained (at least during early ontogeny) in most lineages of Triassic pseudosuchians, which nevertheless generally relied on lower growth rates to reach large body sizes. This trend to slower growth seems to have been further emphasized among Crocodylomorpha, which may thus have secondarily reverted toward more generalized reptilian growth strategies. Accordingly, their “typical ectothermic reptilian condition” may be a derived condition within archosauriforms, homoplastic to the generalized physiological condition of basal amniotes. On the other hand, ornithosuchians apparently retained and even enhanced the high growth rates of many basal archosauriforms during most of their ontogenetic trajectories. The Triassic may have been a time of “experimentation” in growth strategies for several archosauriform lineages, only one of which (ornithodirans) eventually stayed with the higher investment strategy successfully. 相似文献

The sirtuin SIRT1 is an important regulator of energy metabolism through its impact on glucose and lipid metabolism and therefore
we tested the hypothesis that genetic variation in SIRT1 may have an effect on adiposity in a Belgian case/control association study. This study included 1,068 obese patients (BMI ≥ 30 kg/m2) from the outpatient obesity clinic and 313 lean controls (BMI between 18.5 and 25 kg/m2). Anthropometrics were assessed by classical methods and visceral (VFA), subcutaneous (SFA) and total abdominal (TFA) fat
areas were determined by a CT scan. The extent of linkage disequilibrium in SIRT1 allowed us to reduce the number of SNPs to two, sufficient to cover the entire gene. The two tagSNPs (rs7069102 and rs3818292)
were analyzed by LightSNiP assays in all subjects. Rs3818292 genotypes were similarly distributed in cases and controls, whereas
rs7069102 was different for the additive (P = 0.007) and dominant (P = 0.01) model. The variant C-allele of rs7069102 reduced obesity risk with an OR of 0.74 (P = 0.025; 95% CI 0.57–0.96) under a dominant model. In obese male subjects, this variant allele was associated with increased
waist circumference (P = 0.04), WHR (P = 0.02), TFA (P = 0.03) and VFA (P = 0.005) (dominant model; adjusted for age and BMI). Rs3818292 was related to VFA (P = 0.005; adjusted for age and BMI) in obese males while in obese women, no significant associations were detected. Our data
suggest that genetic variation in SIRT1 increases the risk for obesity, and that SIRT1 genotype correlates with visceral obesity parameters in obese men.

Molecular imaging was used to study the biodistribution, pharmacokinetics, and activity of naked small interfering RNAs (siRNAs). siRNAs with riboses chemically modified in the 2' position were compared with unmodified siRNA. In vitro, replacement of the 2'-hydroxyl (2'OH) group of certain nucleotides in an siRNA sequence by a fluorine atom (2'F) on both antisense (AS) and sense (S) strands [2'F(AS/S)], or by a methoxy group (2'OMe) on the S strand [2'OH(AS)/2'OMe(S)], was compatible with RNA interference. Different siRNAs [2'F(AS/S), 2'OH(AS)/2'OMe(S), and 2'OH(AS/S)] were labeled with fluorine-18 (conjugation with [(18)F]FPyBrA), and comparative dynamic and quantitative imaging was performed with positron emission tomography. After intravenous injections of [(18)F]siRNAs in rodents, total radioactivity was rapidly eliminated by the kidneys and the liver. Tissue distribution of the different siRNAs were similar, and their bioavailability (as judged from blood persistence and stability) increased in the order 2'OH(AS/S) = 2'OH(AS)/2'OMe(S) < 2'F(AS/S). However, in our in vivo model, the 2'F(AS/S) siRNA, despite its higher bioavailability, was not able to induce a higher interference effect with respect to the 2'OH(AS/S) siRNA. Molecular imaging approaches, applied in the present work to both natural and chemically modified siRNAs, can contribute to the development of these macromolecules as therapeutic agents. 相似文献

We performed a terrestrial small mammal species inventory in the Agoua and Wari‐Maro forest reserves (Benin). Four localities were sampled, and in each locality, three habitats were surveyed: dense forest, open forest or woodland savannah and shrub savannah. This is the first comprehensive inventory for small mammals in central Benin. We captured 794 small mammals representing twenty species (six shrew species, fourteen rodent species). Three new species that need to be described were recorded. We observed a mixture of both true forest species and of species adapted to a wider range of habitats ranging from savannah to forest clearings. Species with either Sudanian or Guinea–Congolian affinities were recorded, as well as a new species endemic to Togo and Benin. This rich biodiversity underlines the urgent need for an effective protection of these forests. The Sudanian species Crocidura cf. foxi was more abundant in Wari‐Maro than in Agoua forest, while the Guineo–Congolian species Praomys misonnei and Hylomyscus sp were only captured in Agoua forest. Important functions in myogenesis have been proposed for FGF6, a member of the fibroblast growth factor family accumulating almost exclusively in the myogenic lineage. However, the analyses of Fgf6 (-/-) mutant mice gave contradictory results and the role of FGF6 during myogenesis remained largely unclear. Recent reports support the concept that FGF6 has a dual function in muscle regeneration, stimulating myoblast proliferation/migration and muscle differentiation/hypertrophy in a dose-dependent manner. The alternative use of distinct signaling pathways recruiting either FGFR1 or FGFR4 might explain the dual role of FGF6 in myogenesis. A role for FGF6 in the maintenance of a reserve pool of progenitor cells in the skeletal muscle has been also strongly suggested.
