Since 1987, keratinocytes have been cultured at the Queen Astrid Military Hospital. These keratinocytes have been used routinely as auto and allografts on more than 1,000 patients, primarily to accelerate the healing of burns and chronic wounds. Initially the method of Rheinwald and Green was used to prepare cultured epithelial autografts, starting from skin samples from burn patients and using animal-derived feeder layers and media containing animal-derived products. More recently we systematically optimised our production system to accommodate scientific advances and legal changes. An important step was the removal of the mouse fibroblast feeder layer from the cell culture system. Thereafter we introduced neonatal foreskin keratinocytes (NFK) as source of cultured epithelial allografts, which significantly increased the consistency and the reliability of our cell production. NFK master and working cell banks were established, which were extensively screened and characterised. An ISO 9001 certified Quality Management System (QMS) governs all aspects of testing, validation and traceability. Finally, as far as possible, animal components were systematically removed from the cell culture environment. Today, quality controlled allograft production batches are routine and, due to efficient cryopreservation, stocks are created for off-the-shelf use. These optimisations have significantly increased the performance, usability, quality and safety of our allografts. The genetic control of skeletal muscle differentiation at the onset of myogenesis in the embryo is relatively well understood compared to the formation of muscle during the fetal period giving rise to the bulk of skeletal muscle fibers at birth. The Mlc1f/3f (Myl1) locus encodes two alkali myosin light chains, Mlc1f and Mlc3f, from two promoters that are differentially regulated during development. The Mlc1f promoter is active in embryonic, fetal and adult fast skeletal muscle whereas the Mlc3f promoter is upregulated during fetal development and remains on in adult fast skeletal muscle. Two enhancer elements have been identified at the mammalian Mlc1f/3f locus, a 3′ element active at all developmental stages and an intronic enhancer activated during fetal development. Here, using transgenesis, we demonstrate that these enhancers act combinatorially to confer the spatial, temporal and quantitative expression profile of the endogenous Mlc3f promoter. Using double reporter transgenes we demonstrate that each enhancer can activate both Mlc1f and Mlc3f promoters in vivo, revealing enhancer sharing rather than exclusive enhancer-promoter interactions. Finally, we demonstrate that the fetal activated enhancer contains critical E-box myogenic regulatory factor binding sites and that enhancer activation is impaired in vivo in the absence of myogenin but not in the absence of innervation. Together our observations provide insights into the regulation of fetal myogenesis and the mechanisms by which temporally distinct genetic programs are integrated at a single locus.  相似文献

cis-Diamminedichloroplatinum(II) (CDDP) is an important chemotherapeutic agent used in the treatment of a wide variety of solid tumors. We have recently shown that aquated forms of cisplatin (aqua-Pt) rapidly accumulate in K562 and GLC4 cultured cells, in comparison to CDDP. Thus, when cells are incubated with aquated forms of cisplatin a gradient of concentration is observed after a short time, approximately 40 min, with an intracellular concentration of aqua-Pt of 20–30 times higher than that of extracellular aqua-Pt. The same gradient of concentration is observed when cells are incubated with CDDP but it takes a longer time, i.e., about 24 h. Therefore, the question arises as to the identity of the intracellular sites of accumulation of aqua-Pt. Using several agents to modulate membrane potential, acidic compartment pH and/or ATP level, we obtained evidence that aqua-Pt may accumulate rapidly inside mitochondria as this accumulation is energy- and membrane-potential-dependent. However, aqua-Pt complexes are not characterized by a delocalized charge and a lipophilic character that would permit their movement through the inner membrane. Therefore, it is suggested that intracellular aqua-Pt reacts rapidly with glutathione with the resultant complex being transported inside the mitochondria via one of the known glutathione transporters, i.e., dicarboxylate and/or 2-oxoglutarate transporters present in the inner membrane.  相似文献

The allelopathic potential of two invasive alien Ludwigia [Onagraceae: L. peploides (Kunth) Raven and L. grandiflora (Michaux) Greuter and Burdet], that have developed quasi-monotypic stands in many aquatic ecosystems in France, was investigated. Since allelopathy involves the release of compounds into the environment, the water of monospecific experimental cultures was directly tested against two target species: Lactuca sativa L., the standard cultivar for bioassays, and Nasturtium officinale R. Brown, a resistant and widespread native hydrophyte. The treatment was carried out at the three main phases of development of both Ludwigia in February, May and August. For each experiment, the germination, mortality and culture yield percentages, the seedling growth (radicle and hypocotyl elongation) and the health of 15-day-old-seedlings were measured. The water of each Ludwigia tank induced: (1) a decrease in germination for watercress in August (control: 68.6%, L. peploides: 48.6%, L. grandiflora: 61.1%); (2) an increase in mortality in May only for watercress (control: 3.4%, L. peploides: 13.5%, L. grandiflora: 12%) and in August for both target species (up to 22.3% vs. 3% for lettuce and 27% vs. 12.5% for watercress); (3) a disturbance of seedling elongation for lettuce in all seasons; and (4) a seedling chlorosis of both target species, particularly in May and August. This study showed that L. peploides and L. grandiflora possess an allelopathic activity that influences the water quality throughout the year. Combined with the various competitive attributes, allelopathy may contribute to the great success of these two invasive Ludwigia in Europe. In threatened wetland communities of the Mediterranean area, in particular, allelopathy might have an important impact by diminishing the seedling survival of the most vulnerable species.  相似文献

This work describes the synthesis and characterization of four new ligands derived from 1,3-propanediamine in addition to the preparation and characterization of their respective platinum(II) complexes by reaction with K2PtCl4. These ligands were obtained by the reaction of the corresponding alkyl mesylate with 1,3-propanediamine. We have prepared compounds having different carbon chains lengths in an attempt to correlate this factor, which influences the lipophilicity of the compounds, with cytotoxic activity. Octanol/water partition coefficients, the effect of the four complexes on the growth of two tumoral cell lines, and their cellular uptake were investigated. Increasing lipophilicity enhances the rate of cellular uptake and, consequently, the cytotoxic activity.  相似文献

Endocrine disruption is commonly thought to be restricted to a direct endocrine mode of action i.e. the perturbation of the activation of a given type of hormonal receptor by its natural ligand. Consistent with the WHO definition of an endocrine disrupter, a key issue is the “altered function(s) of the endocrine system”. Such altered functions can result from different chemical interactions, beyond agonistic or antagonistic effect at a given receptor. Based on neuroendocrine disruption by polychlorinated biphenyls and bisphenol A, this paper proposes different mechanistic paradigms that can result in adverse health effects. They are a consequence of altered endocrine function(s) secondary to chemical interaction with different steps in the physiological regulatory processes, thus accounting for a possibly indirect endocrine mode of action.  相似文献

Summary In Podospora anserina, positive and very efficient chiasma interference is observed. However, its modalities are different for the two linkage groups 1 (LG1) and 6 (LG6) studied here.In the right arm of LG1, two zones exist in which always occurs only one crossing-over. They are formed independently each other. Moreover, the genetic map consists of clusters of genes located near the centromere and at the limit between the two interference zones. It is postulated that this structure of the map results from the localization of crossing-over in the middle of each zone. We suppose that the type of chiasma interference, in Podospora, is a typical one as it is in Drosophila. The knowledge of higher plant microtubule-associated proteins (MAPs) remains limited to a few examples that illustrate essentially their binding properties to preformed microtubules as described in carrots. Using taxol-stabilized microtubules a putative MAP-enriched fraction has been isolated in maize cultured cell extracts, one of these polypeptides is immunologically related to neural tau. At present, these proteins are being characterized by co-assembly assays that were not possible before. Similar experiments were done also in a heterologous system using brain tubulin. Three polypeptides out of seven that constituted the MAP fraction were found to co-assemble specifically with tubulin subunits of both origins. Their apparent molecular weights are 67, 83 and 125 kDa. A two-dimensional gel immunoblot of the 83 kDa polypeptide with tau antibodies revealed one major spot. Polypeptides were quantiated by scanning the gels. These results shed light on the present debate on higher plant MAPs and their potential activity in the regulation of microtubule assembly and function in the higher plant cell.  相似文献

The radiation inactivation method has been used to determine the molecular mass of membrane-bound acid β-glucosidase (EC 3.2.1.21) in situ, in normal human spleen and in that of two patients with type I Gaucher disease: the molecular mass in Gaucher spleen is about double (125 000 ± 8900) of that found in the normal spleen (67 000 ± 7700) which is compatible with the existence of subunit coupling in the muted acid β-glucosidase. From the results, we conclude that subunit interaction is altered in mutant acid β-glucosidase and that this may be due to a direct effect of the mutation.  相似文献
