为了明确具有极强抗虫特性的‘草原4号紫花苜蓿’(Medicago sativa L.‘Caoyuan No.4’) 营养器官的解剖特征,该研究选择具有抗蓟马特性较强的‘草原2号杂花苜蓿’(Medicago varia Martin.‘Caoyuan No.2’)为对照,采用显微镜观察比较两品种的根、茎、叶解剖结构特征,为揭示‘草原4号紫花苜蓿’ 抗蓟马特性提供理论依据。结果显示:(1)‘草原4号紫花苜蓿’根部解剖结构的皮层薄壁细胞厚度、内皮层厚度、形成层厚度、木质部厚度和木射线宽度等5个指标均极显著高于(P<0.01)‘草原2号杂花苜蓿’,其中木射线宽度(159.37 μm)是‘草原2号杂花苜蓿’的1.82倍。(2)‘草原4号紫花苜蓿’的茎部厚角组织厚度(21.4 μm)极显著高于‘草原2号杂花苜蓿’(P<0.01),而韧皮部宽度、髓直径却均极显著低于‘草原2号杂花苜蓿’(P<0.01)。(3)‘草原4号紫花苜蓿’叶片解剖构造的7个指标均极显著高于‘草原2号杂花苜蓿’(P<0.01),其中栅栏组织层数(2~3层)极明显地高于‘草原2号杂花苜蓿’(1~2层)。研究表明,‘草原4号紫花苜蓿’的组织结构特征具有明显的抗虫特征,且其组织的抗虫特征比‘草原2号杂花苜蓿’更为突出。  相似文献

Bird species diversity of the altitudinal belts of the Eastern Himalayas was analyzed in the early spring of 2005 and 2014. Species richness is revealed to be decreasing from the belts of subtropical mixed and coniferous forests to the alpine belt. Specific species that are not beyond the limits of a corresponding belt are immanent to three of four investigated altitudinal belts. The avifaunas of two adjacent belts also have comparatively many common species. One hundred and thirty-three bird species met in both years belong to six faunal complexes, among which most species are Himalayan endemics and subendemics, as well as Palearctic species. The abundance of background species has been determined for each altitudinal belt.  相似文献

Species composition and quantitative distribution of ichthyoplankton in areas of the Rockall-Hutton seamounts are considered in a comparative aspect, and the spawning biomass of the blue whiting Micromesistius poutassou, the main commercial item of this region, is calculated. Maximal catches of eggs and larvae were recorded in spring 2006 and minimal catches were recorded in spring 2003. The highest number of species was recorded in March–April 2006 and the lowest number was recorded in March–April 2003. In the seasons under consideration, M. poutassou had the most mass of the commercial fish in the plankton; haddock Melanogrammus aeglefinus and gurnard Eutrigla gurnardus were less numerous. Of the noncommercial species, the larvae of myctophids (fam. Myctophidae) and of other mesopelagic fish occurred more frequently in the ichthyoplankton. The species and quantitative composition of ichthyoplankton was considerably more numerous at the Rockall Seamount than at the Hutton Seamount, which is explained by specific oceanographic features of the region and the more southern location of this seamount. According to the data of the ichthyoplanktonic method, the spawning biomass of M. poutassou exhibited a tendency for an increase in the period of 2002 to 2006.  相似文献

Immunofluorescent staining techniques were used to study the distribution of the Ca(2) + Mg(2+)-dependent ATPase and calsequestrin in primary cultures of differentiating rat skeletal muscle cells, grown for different periods of time under various culture conditions. In mononucleated myoblasts calsequestrin was detected after 45 h in culture whereas the ATPase was not detected until 60 h. After cell fusion began, both proteins could be identified in all multinucleated cells. Myoblasts grown for longer than 60 h in low Ca(2+) medium contained calsequestrin and the ATPase, even though they were unable to fuse. These studies at the cellular level confirm biochemical findings on the biosynthesis of calsequestrin and the ATPase. Immunofluorescent staining of myoblasts showed that calsequestrin first appears in a well-defined region of the cell near one end of the nucleus. At later times, the staining occupied progressively larger regions adjacent to the nucleus and took on a fibrous appearance. This suggests that calsequestrin first accumulates in the Golgi region and then gradually spreads throughout the cell. In contrast, the ATPase appeared to be concentrated in many small patches or foci throughout the cytoplasm and was never confined to one particular region, although some parts of the cell often stained more intensely than others. In multinucleated cells, alternating dark and fluorescent strands parallel to the longitudinal axis of the cells were evident.  相似文献

 The mesophyll structure of current and second year needles of mature Scots pines (Pinus sylvestris L.), growing on a survey line from the Karelian Isthmus (N.W. Leningrad region, Russia) to S.E. Finland, was studied at the light microscopic level in 1992 and 1994. The area is affected by sulphur dioxide and nitrogen oxide emissions from Russia (200  000–285 000 ton/year SO2 and about 100  000 ton/year NOX), Estonia (140  000–320  000 ton/year SO2 and 30  000–50  000 ton/year NOX) and Finland (6000–9000 ton/year SO2 and 23 000–25 000 ton/year NOX) and by unknown amounts of neutralizing compounds from Estonia. Mean concentrations of SO2 were between 1.0 and 1.5 μg m–3 and those of ozone between 52 and 61 μg m–3 during growing seasons of 1991–1994. The concentrations tended to be lower in 1993/94 than in 1991/92. The results showed cellular changes related to exposure to sulphur dioxide, ozone and acid wet deposition. The percentage of cells with injuries related to acid deposition was higher in 1994 than in 1992, possibly due to higher reduction of neutralizing than of acidifying compounds in the main emission areas, while the percentage of cells with injuries related to sulphur dioxide correlated positively with needle sulphur concentrations and showed a declining trend between Karelian Isthmus and S.E. Finland in the needles sampled in 1992. These changes and those related to ozone were considerably less numerous in 1994 than in 1992, possibly on account of the lower concentrations of SO2 and ozone in 1993/94 than in 1991/92, especially in early summer, the time of the most sensitive phase of needle growth. In addition, periods of lower wet deposition in summer 1992, and considerably lower temperatures in autumn/winter 1992 than in 1994 may have enhanced the injuries in the needles collected in October/December 1992, relative to those collected in 1994. The results of the microscopic work agreed well with the simultaneous chemical measurements, pointing to the usefulness of cellular analysis for diagnostic work in the field. New experimental and clinical data on the function of the ligamentum capitis femoris (LCF) and its participation in maintaining an erect posture were obtained. It was established that this anatomical element is involved in constraining the hip joint adduction and may fix the joint in the frontal plane, turning it into an analogue of a second-class lever. In both unstrained one-suport and symmetrical two-support orthostatic postures, when the LCF is stretched and the abductor muscle group is exerted, a load equal to the body weight is evenly distributed between the upper and lower hemispheres of the caput femoris. In addition, the LCF function increases the steadiness of the erect posture and unloads the muscle apparatus.  相似文献

Adaptation is a complex process by which populations of organisms respond to long-term environmental stresses by permanent genetic change. Here we present data from the natural "open-field" radiation adaptation experiment after the Chernobyl accident and provide the first evidence of the involvement of epigenetic changes in adaptation of a eukaryote-Scots pine (Pinus silvestris), to chronic radiation exposure. We have evaluated global genome methylation of control and radiation-exposed pine trees using a method based on cleavage by a methylation-sensitive HpaII restriction endonuclease that leaves a 5' guanine overhang and subsequent single nucleotide extension with labeled [3H] dCTP. We have found that genomic DNA of exposed pine trees was considerably hypermethylated. Moreover, hypermethylation appeared to be dependent upon the radiation dose absorbed by the trees. Such hypermethylation may be viewed as a defense strategy of plants that prevents genome instability and reshuffling of the hereditary material, allowing survival in an extreme environment. Further studies are clearly needed to analyze in detail the involvement of DNA methylation and other epigenetic mechanisms in the complex process of radiation stress and adaptive response.  相似文献

Atomic force microscopy has recently provided highly precise measurements of mechanical properties of various viruses. However, molecular details underlying viral mechanics remain unresolved. Here we report atomic force microscopy nanoindentation experiments on T=4 hepatitis B virus (HBV) capsids combined with coarse-grained molecular dynamics simulations, which permit interpretation of experimental results at the molecular level. The force response of the indented capsid recorded in simulations agrees with experimental observations. In both experiment and simulation, irreversible capsid deformation is observed for deep indentations. Simulations show the irreversibility to be due to local bending and shifting of capsid proteins, rather than their global rearrangement. These results emphasize the viability of large capsid deformations without significant changes of the mutual positions of HBV capsid proteins, in contrast to the stiffer capsids of other viruses, which exhibit more extensive contacts between their capsid proteins than seen in the case of HBV.  相似文献
