To investigate the signal transduction pathway of caspase-2, cell permeable Tat-reverse-caspase-2 was constructed, characterized and utilized for biochemical and cellular studies. It could induce the cell death as early as 2 h, and caspase-2-specific VDVADase activity but not other caspase activities including DEVDase and IETDase. Interestingly, nuclear DNA fragmentation occurred and consistently DNA fragmentation factor (DFF45)/Inhibitor of caspase-activated DNase (ICAD) was cleaved inside the cell as well as in vitro, suggesting a role of caspase-2 in nuclear DNA fragmentation. 相似文献

Conformational dynamics play a crucial role in biological function. Dynein light chain protein (DLC8) acts as a cargo adaptor, and exists as a dimer under physiological conditions and dissociates into monomer below pH 4. In the present NMR study, we identified some dynamic residues in the dimer using chemical shift perturbation approach by applying small pH change. As evidenced by gel filtration and CD studies, this small pH change does not alter the globular structural features of the protein. In fact, these changes result in small local stability perturbations as monitored using temperature dependence of amide proton chemical shifts, and influence the dynamics of the dimer substantially. Further, interaction studies of the protein with a peptide containing the recognition motif of cargo indicated that the efficacy of peptide binding decreases when the pH is reduced from 7 to 6. These observations taken together support the conception that dynamics can regulate cargo binding/trafficking by the DLC8 dimer. 相似文献

Reactions of HgX2 (X = Cl, Br, I) with methylene blue (MB) chloride dihydrate and the reaction of HgCl2 with MB nitrate dihydrate have been undertaken in an attempt to prepare metal derivatives of MB. The products were characterized by elemental analysis, UV-Vis, IR, 1H NMR spectroscopy and X-ray crystallography. The reaction of HgCl2 with MB chloride dihydrate and subsequent crystallization in a DMF/H2O mixture yielded the products (1 and 2) with different crystal morphologies. The structure of complex 1 represents the first structural report of a complex of MB with any metal ion. The efficacy of MB to act as a ligand in spite of its cationic nature is thus demonstrated. The structure of 1 comprises a distorted tetrahedral geometry around Hg(II); the coordination valencies being provided by three chloride ions and a MB cation. The structure of 2 is a salt consisting of an anion and two MB cations. The reaction of HgI2 and HgBr2 with MB chloride dihydrate yielded salts 3 and 4 with (X = Br or I) anions and MB cations. A mixed salt 5, whose anions comprise mercury(II), chloride and nitrate species resulted from the reaction between HgCl2 with MB nitrate dihydrate. The reaction of Hg2F2 with MB chloride dihydrate and the crystallization of the resulting product 6 in aqueous DMF yielded crystals of (MB)2HgCl4 · H2O. 相似文献

The Escherichia coli NapA (periplasmic nitrate reductase) contains a [4Fe-4S] cluster and a Mo-bis-molybdopterin guanine dinucleotide cofactor. The NapA holoenzyme associates with a di-heme c-type cytochrome redox partner (NapB). These proteins have been purified and studied by spectropotentiometry, and the structure of NapA has been determined. In contrast to the well characterized heterodimeric NapAB systems ofalpha-proteobacteria, such as Rhodobacter sphaeroides and Paracoccus pantotrophus, the gamma-proteobacterial E. coli NapA and NapB proteins purify independently and not as a tight heterodimeric complex. This relatively weak interaction is reflected in dissociation constants of 15 and 32 mum determined for oxidized and reduced NapAB complexes, respectively. The surface electrostatic potential of E. coli NapA in the apparent NapB binding region is markedly less polar and anionic than that of the alpha-proteobacterial NapA, which may underlie the weaker binding of NapB. The molybdenum ion coordination sphere of E. coli NapA includes two molybdopterin guanine dinucleotide dithiolenes, a protein-derived cysteinyl ligand and an oxygen atom. The Mo-O bond length is 2.6 A, which is indicative of a water ligand. The potential range over which the Mo(6+) state is reduced to the Mo(5+) state in either NapA (between +100 and -100 mV) or the NapAB complex (-150 to -350 mV) is much lower than that reported for R. sphaeroides NapA (midpoint potential Mo(6+/5+) > +350 mV), and the form of the Mo(5+) EPR signal is quite distinct. In E. coli NapA or NapAB, the Mo(5+) state could not be further reduced to Mo(4+). We then propose a catalytic cycle for E. coli NapA in which nitrate binds to the Mo(5+) ion and where a stable des-oxo Mo(6+) species may participate. 相似文献

The Kulsi River, a major tributary of Brahmaputra River, N. India is reported to have resident population of Ganges river dolphin, Platanista gangetica (Roxburgh), which feeds on fishes and prawns. While surveying for the dolphins of the river, a number of fishes and prawns were collected. On identification, one of the prawns was found to be undescribed, and hence is described herein. The ecology of the river consisted of: temperature fluctuating widely from 15 to 28 °C, depth from 0.8 to 10 m, turbidity of 11-19 cm, sand mining @ 12,500 MT annually, and fish catch of 300-800 kg (from 1.5 km area). All these factors pose a great threat to the fish and prawn wealth of the river.Macrobrachium kulsiense sp. nov. is a very small sized prawn (maximum size - 34.5 mm in total length), exhibiting species-specific characters, such as highly elevated and moderately long rostrum with 9-12 dorsal teeth, a single ventral tooth, and percentage ratios of ischium, merus, carpus, palm, dactylus of first and second chelipeds (19.05:28.57:33.33:09.52:09.52 and 21.43:25.00:21.43:14.28:17.86, respectively). The species shows close similarity with Macrobrachium mirabile (Kemp). The females are larger than males, eggs are large in size (1.2×0.9 mm) and fecundity is low (15-20). 相似文献

Unlike a purely reactive system where the motor output is exclusively controlled by the actual sensory input, a cognitive system must be capable of running mental processes which virtually simulate action sequences aimed at achieving a goal. The mental process either attempts to find a feasible course of action compatible with a number of constraints (Internal, Environmental, Task Specific etc) or selects it from a repertoire of previously learned actions, according to the parameters of the task. If neither reasoning process succeeds, a typical backup strategy is to look for a tool that might allow the operator to match all the task constraints. This further necessitates having the capability to alter ones own goal structures to generate sub-goals which must be successfully accomplished in order to achieve the primary goal. In this paper, we introduce a forward/inverse motor control architecture (FMC/IMC) that relaxes an internal model of the overall kinematic chain to a virtual force field applied to the end effector, in the intended direction of movement. This is analogous to the mechanism of coordinating the motion of a wooden marionette by means of attached strings. The relaxation of the FMC/IMC pair provides a general solution for mentally simulating an action of reaching a target position taking into consideration a range of geometric constraints (range of motion in the joint space, internal and external constraints in the workspace) as well as effort-related constraints (range of torque of the actuators, etc.). In case, the forward simulation is successful, the movement is executed; otherwise the residual "error" or measure of inconsistency is taken as a starting point for breaking the action plan into a sequence of sub actions. This process is achieved using a recurrent neural network (RNN) which coordinates the overall reasoning process of framing and issuing goals to the forward inverse models, searching for alternatives tools in solution space and formation of sub-goals based on past context knowledge and present inputs. The RNN + FMC/IMC system is able to successfully reason and coordinate a diverse range of reaching and grasping sequences with/without tools. Using a simple robotic platform (5 DOF Scorbot arm + Stereo vision) we present results of reasoning and coordination of arm/tool movements (real and mental simulation) specifically directed towards solving the classical 2-stick paradigm from animal reasoning at a non linguistic level. 相似文献

Tuberculosis is a global problem especially in children, more so because of the difficulty in the diagnosis of the disease.
There are about 0.5 million deaths every year by this disease, worldwide. There are some reports that, during the disease,
there are changes in serum concentrations of zinc and copper, which reach towards normal levels during treatment. Such data
for children are scarce, especially in reference to India. Our study indicates that there is an insignificant to significant
rise in serum zinc levels in various age groups of children after antitubercular therapy (mean 61.89 ± 3.21 to 65.24 ± 3.60 μg/dl)
and significant fall in serum copper levels in different age groups of children (mean 129.96 ± 3.18 to 124.91 ± 3.48 μg/dl).
The Cu/Zn ratio also changed significantly from 2.11 ± 0.12 to 1.92 ± 0.12. Because evaluation of available diagnostic criteria
for primary complex in children has been found to have high sensitivity, and although this criterion is presently not recommended
for diagnosis of primary complex of children, further research can prove its utility in diagnosis and treatment of this disease. 相似文献

Exclusion of UV (280–380 nm) radiation from the solar spectrum can be an important tool to assess the impact of ambient UV
radiation on plant growth and performance of crop plants. The effect of exclusion of UV-B and UV-A from solar radiation on
the growth and photosynthetic components in soybean (Glycine max) leaves were investigated. Exclusion of solar UV-B and UV-B/A radiation, enhanced the fresh weight, dry weight, leaf area
as well as induced a dramatic increase in plant height, which reflected a net increase in biomass. Dry weight increase per
unit leaf area was quite significant upon both UV-B and UV-B/A exclusion from the solar spectrum. However, no changes in chlorophyll
a and b contents were observed by exclusion of solar UV radiation but the content of carotenoids was significantly (34–46%) lowered.
Analysis of chlorophyll (Chl) fluorescence transient parameters of leaf segments suggested no change in the F
v/F
m value due to UV-B or UV-B/A exclusion. Only a small reduction in photo-oxidized signal I (P700+)/unit Chl was noted. Interestingly the total soluble protein content per unit leaf area increased by 18% in UV-B/A and 40%
in UV-B excluded samples, suggesting a unique upregulation of biosynthesis and accumulation of biomass. Solar UV radiation
thus seems to primarily affect the photomorphogenic regulatory system that leads to an enhanced growth of leaves and an enhanced
rate of net photosynthesis in soybean, a crop plant of economic importance. The presence of ultra-violet components in sunlight
seems to arrest carbon sequestration in plants.

Our previous studies demonstrated that peripheral overexpression of angiotensin II (ANG II) type 2 receptors (AT(2)R) prevents hypertension-induced cardiac hypertrophy and remodeling without altering high blood pressure. This, coupled with the observations that AT(2)R play a role in the antihypertensive actions of ANG II type 1 receptor (AT(1)R) blockers (ARBs), led us to propose that peripheral overexpression of AT(2)R would improve the antihypertensive action of losartan (Los) in Sprague-Dawley (SD) rats made hypertensive via chronic infusion of ANG II. Here we utilized adenoviral vector-mediated AT(2)R gene transfer to test this hypothesis. A single intracardiac injection of adenoviral vector containing genomic AT(2)R (G-AT(2)R) DNA and enhanced green fluorescent protein (EGFP) gene controlled by cytomegalovirus (CMV) promoters (Ad-G-AT(2)R-EGFP; 5 x 10(9) infectious units) into adult SD rats produced robust AT(2)R overexpression in cardiovascular tissues (kidney, lung, heart, aorta, mesenteric artery, and renal artery) that persisted for 3-5 days postinjection. By 7 days post viral injection, the overexpressed AT(2)R are reduced toward basal values in certain tissues (lung, kidney, and heart) and are undetectable in others (kidney and blood vessels). In two separate protocols, we demonstrated that the hypotensive effect of Los (0.125, 0.5, and 1.0 mg/kg iv) was significantly greater in the AT(2)R-overexpressing animals (-40.7 +/- 4.3, -41.8 +/- 4.8, and -48.1 +/- 2.6 mmHg, respectively) compared with control vector (Ad-CMV-EGFP)-treated rats (-12.4 +/- 2.2, -20.2 +/- 3.4, and -27.3 +/- 3.4 mmHg, respectively). These results provide support for a depressor role of AT(2)R and the proposal that combined AT(2)R agonist and ARB treatment may be an improved therapeutic strategy for controlling hypertension. 相似文献
