Background
Small populations are thought to be adaptively handicapped, not only because they suffer more from deleterious mutations but also because they have limited access to new beneficial mutations, particularly those conferring large benefits.Methodology/Principal Findings
Here, we test this widely held conjecture using both simulations and experiments with small and large bacterial populations evolving in either a simple or a complex nutrient environment. Consistent with expectations, we find that small populations are adaptively constrained in the simple environment; however, in the complex environment small populations not only follow more heterogeneous adaptive trajectories, but can also attain higher fitness than the large populations. Large populations are constrained to near deterministic fixation of rare large-benefit mutations. While such determinism speeds adaptation on the smooth adaptive landscape represented by the simple environment, it can limit the ability of large populations from effectively exploring the underlying topography of rugged adaptive landscapes characterized by complex environments.Conclusions
Our results show that adaptive constraints often faced by small populations can be circumvented during evolution on rugged adaptive landscapes. 相似文献73.

In order to preserve their genome integrity, organisms have developed elaborate tactics for genome protection and repair. The Deinococcus radiodurans bacteria famous for their extraordinary tolerance toward high doses of radiations or long period of desiccation, possess some specific genes with unknown function which are related to their survival in such extreme conditions. Among them, ddrA is an orphan gene specific of Deinococcus genomes. DdrA, the product of this gene was suggested to be a component of the DNA end protection system. Here we provide a three-dimensional reconstruction of the Deinococcus deserti DdrA((1-160)) by electron microscopy. Although not functional in vivo, this truncated protein keeps its DNA binding ability at the wild-type level. DdrA((1-160)) has a complex three-dimensional structure based on a heptameric ring that can self-associate to form a larger molecular weight assembly. We suggest that the complex architecture of DdrA plays a role in the substrate specificity and favors an efficient DNA repair. 相似文献

Goal, Scope and Background The EU 5th framework project OMNIITOX will develop models calculating characterisation factors for assessing the potential toxic impacts
of chemicals within the framework of LCA. These models will become accessible through a web-based information system. The
key objective of the OMNIITOX project is to increase the coverage of substances by such models. In order to reach this objective,
simpler models which need less but available data, will have to be developed while maintaining scientific quality. Methods.
Experience within the OMNIITOX project has taught that data availability and quality are crucial issues for calculating characterisation
factors. Data availability determines whether calculating characterisation factors is possible at all, whereas data quality
determines to what extent the resulting characterisation factors are reliable. Today, there is insufficient knowledge and/or
resources to have high data availability as well as high data quality and high model quality at the same time.
Results The OMNIITOX project is developing two inter-related models in order to be able to provide LCA impact assessment characterisation
factors for toxic releases for as broad a range of chemicals as possible: 1) A base model representing a state-of-the-art
multimedia model and 2) a simple model derived from the base model using statistical tools. Discussion. A preliminary decision
tree for using the OMNIITOX information system (IS) is presented. The decision tree aims to illustrate how the OMNIITOX IS
can assist an LCA practitioner in finding or deriving characterisation factors for use in life cycle impact assessment of
toxic releases.
Conclusions and Outlook Data availability and quality are crucial issues when calculating characterisation factors for the toxicity impact categories.
The OMNIITOX project is developing a tiered model approach for this. It is foreseen that a first version of the base model

Field experiments in managed grassland have shown that the response of vegetative growth to elevated CO2 is nitrogen‐dependent in grasses, but independent in N2‐fixing legumes. In the present study, we tested whether this is also true for reproduction. We evaluated reproductive growth, flowering phenology, seed development, reproductive success and seed germination in the grass Lolium perenne L. and the legume Trifolium repens L., growing in monocultures in a free air carbon dioxide enrichment (FACE) system at ambient (35 Pa) and elevated (60 Pa) partial pressure of CO2 and two levels of nitrogen fertilization (14 and 56 g N m?2 a?1). In both species, elevated CO2 had no significant effect on sexual reproduction. In L. perenne, reproduction was mainly nitrogen‐dependent. The weak interactions between CO2 and mineral N supply (13% more flowers and 8% more grains per spike at high N, 7% less flowers and 8% less grains at low N) were not significant. Under elevated CO2, grain maturation was slightly enhanced and grain weight tended to decrease. No influence could be ascertained in the date of anthesis, the temporal pattern of grain growth, the rate of grain abortion and germination. Trifolium repens, grown under CO2 enrichment at both levels of N fertilization, flowered 10 d earlier, tended to form more inflorescences per ground area and more flowers (8–12%) and seeds (>18%) per inflorescence than at ambient CO2. The temporal pattern of seed growth was about the same in all treatments; embryo development, however, was accelerated in fumigated plants. The number of aborted seeds per pod, seed size, thousand‐seed weight and germinability did not show any influence of CO2. Fumigated plants at high N were attacked slightly more frequently by seed‐eating weevils, which lowered the seed output per pod. In summary, the reproductive response of L. perenne and T. repens to CO2 enrichment on the flower and inflorescence level was far weaker than expected from the results on vegetative growth. 相似文献

Technical enzymes are used in many industrial applications. Nowadays technical enzymes are often produced in transgenic host organisms. The use of transgenic plants with respect to high level of expression at low costs as a prerequisite for successful commercial production of technical enzymes is discussed. This review summarises recently published examples for production of technical enzymes in plants. The impact of regional factors (such as speciation or dispersal) on the species richness in local communities (SL) has received increasing attention. A prominent method to infer the impact of regional factors is the comparison of species richness in local assemblages (SL) with the total number of species in the region (SR). Linear relations between SR and SL have been interpreted as an indication of strong regional influence and weak influence of interactions within local communities. We propose that two aspects bias the outcome of such comparisons: (1) the spatial scale of local and regional sampling, and (2) the body size of the organisms. The impact of the local area reflects the scales of ecological interactions, whereas the ratio between local and regional area reflects the inherent moment of autocorrelation. A proposed impact of body size on the relation is based on the high dispersal and high abundance of small organisms. We predict strongest linearity between SR and SL for large organisms, for large local areas (less important ecological interactions) and for sampling designs where the local habitat area covers a high proportion of the regional area (more important autocorrelation). We conducted a meta-analysis on 63 relations obtained from the literature. As predicted, the linearity of the relationship between SL and SR increased with the proportion of local to regional sampling area. In contrast, neither the body size of the organisms nor the local area itself was significantly related to the relation between SL and SR. This indicated that ecological interactions played a minor role in the shape of local to regional richness plots, which instead was mainly influenced by the sampling design. We found that the studies published so far were highly biased towards larger organisms and towards high similarity between the local and regional area. The proposed prevalence of linear relationships may thus be an artefact and plots of SL to SR are not a suitable tool with which to infer the strength of local interactions. 相似文献

A method is designed to change geometrical proportions of muscles in the rat. After a tibial osteotomy, the application of a stainless steel prosthesis resulted in an elongation of about 8% of the tibia. In 23 out of 28 operated rats a correct application of the prosthesis was maintained till the rats were sacrificed. The operation caused hardly any muscle tissue damage. Characterization of Nicotine-Induced Desensitization of Evoked Dopamine Release from Rat Striatal Synaptosomes 总被引:4,自引:1,他引:3
Abstract: Nicotine has been shown to stimulate neurotransmitter release from brain tissue by acting on presynaptic receptors. In this study, the ability of nicotine pretreatment to produce functional desensitization was investigated in rat striatal synaptosomes in which the release of [3 H]dopamine was measured with an in vitro superfusion system. Pretreatment of synaptosomes with low concentrations of l -nicotine resulted in a decrease in the ability of a subsequent nicotine challenge to evoke [3 H]dopamine release. The IC50 for nicotine-induced desensitization was found to be 12 n M with a maximum inhibition of >90% at 300 n M . Nicotine pretreatment did not affect the release evoked by amphetamine, veratridine, or 15 m M K+ . The onset of nicotine-induced desensitization occurred with a t 1/2 of 43 s at 30 n M nicotine. The temperature dependence of onset yielded a Q10 of 1.2.Recovery from desensitization was slower ( t 1/2 = 4.33 min), and both the onset and recovery appeared to follow a single first-order process. Several intermittent schedules of nicotine treatment were found to be effective at inducing and maintaining desensitization. The results of this study show that nonstimulating concentrations of nicotine can produce a complete functional desensitization of subsequent nicotine-induced neurotransmitter release. 相似文献
