Three new Lycopodium alkaloids, lycoparins A-C (1-3), have been isolated from the club moss Lycopodium casuarinoides. Structures and stereochemistry of 1-3 were elucidated on the basis of 2D NMR correlations. In many animals, cytochalasins have generally been used as cytoskeletal inhibitors for the diploid complement retention of somatic cell nuclear transfer (SCNT) embryos. However, limited information is available on the effects of cytochalasins on the in vitro development of SCNT embryos. Hence, we compared the effects of cytochalasin B (CB) and cytochalasin D (CD) on pseudo-polar body (pPB) extrusion, cortical actin filament (F-actin) distribution in porcine parthenogenetic oocytes and in vitro development of SCNT embryos that were reconstructed using foetal fibroblasts in the G0/G1 phase derived from miniature pigs. CB (7.5 microg/ml) and CD (2.5 microg/ml) treatments effectively inhibited pPB extrusion in SCNT embryos. CB (2.5 microg/ml) treatment could not inhibit pPB extrusion and insufficiently destabilized F-actin immediately following artificial activation. In parthenogenetic oocytes treated with 2.5 microg/ml CD, normal reorganization and uniform distribution of cortical F-actin at the cytoplasmic membrane were observed at 8 h after artificial activation; this finding was similar to that of control oocytes. In contrast, parthenogenetic oocytes treated with 7.5 microg/ml CB showed non-uniform distribution of F-actin at 8 h after artificial activation. On day 5 after in vitro cultivation, the blastocyst formation rate of SCNT embryos treated with 2.5 microg/ml CD was significantly higher than that of SCNT embryos treated with 2.5 and 7.5 microg/ml CB (p < 0.05). Hence, the present findings suggest that CD is more effective than CB as the cytoskeletal inhibitor for the production of SCNT embryos in miniature pigs. 相似文献

Nitric oxide (NO) and reactive oxygen species (ROS) are known to play various functional and pathophysiological roles as an intracellular messenger in the heart. In this study, we investigated whether the increased production of NO and/or ROS was involved in the cholinergic regulation of rhythmic contraction in spontaneously beating cultured cardiac myocytes from neonatal rats. Exposure of cultures to carbachol, an agonist of muscarinic acetylcholine receptors (mAchR), produced a dose-dependent decrease in the beat rate of cultured cardiac myocytes, and such a effect was significantly attenuated by pre-treatment with an NOS inhibitor, as well as an NO scavenger. In addition, exposure to an NO donor (SNAP) also decreased the beat rate dose-dependently. Carbachol- or SNAP-induced suppression of the contraction rhythm was significantly attenuated by co-treatment with 5-hydroxydecanoate (5-HD). In contrast, treatment with diazoxide decreased the beat rate dose-dependently. Carbachol treatment increased the intensity of 2',7'-dichlorodihydrofluorescein fluorescence, suggesting that the production of ROS was enhanced by the treatment. In addition, the carbachol- or diazoxide-induced suppression of contraction rhythm was attenuated by co-treatment with 2-mercaptopropionyl glycine, a scavenger of ROS. The present study has suggested that the mAchR-NO-mitoK ATP -ROS pathway is a factor responsible for carbachol-induced suppression of contraction rhythm in cultured cardiac myocytes. 相似文献

The genealogical and geographic structure of variation in spikelet morphology was analyzed for central Eurasian wild wheat Aegilops tauschii Coss. using a diverse array of 203 sample accessions that represented the entire species range. In this sample set, two subspecies were identified on the basis of sensu-stricto criteria: only the accessions having markedly moniliform spikes were assigned to Ae. tauschii Coss. subspecies strangulata (Eig) Tzvel., whereas those having mildly moniliform and cylindrical spikes to Ae. tauschii Coss. subspecies tauschii. In a graph of the first two axes from a principal component analysis based on nine spikelet traits, the plots of the two subspecies formed separate clusters, indicating that subspecies strangulata sens. str. is a practically usable taxon. Chloroplast-DNA-based genealogical analyses suggested that subspecies strangulata diverged from an ancestor that carried a specific chloroplast DNA type, whereas, after divergence, this subspecies became polyphyletic, likely through hybridization. Geographically, significant longitudinal and latitudinal clines were detected for spikelet size, with spikelets tending to be small in the eastern and southern regions. These results shed some light on the patterns of subspecies divergence and spikelet-shape diversification in the course of Ae. tauschii’s long-distance dispersal from the Transcaucasus to China. 相似文献

The aim of this study is to develop a prototype system for noncontact, noninvasive and unconstrained vital sign monitoring using microwave radar and to use the system to measure the respiratory rate of a Japanese black bear (Ursus thibetanus japonicus) during hibernation for ensuring the bear's safety. Ueno Zoological Gardens in Tokyo planned to help the Japanese black bear (female, approximately 2 years of age) going into hibernation. The prototype system has a microwave Doppler radar antenna (10-GHz frequency, approximately 7 mW output power) for measuring motion of the body surface caused by respiratory activity without making contact with the body. Monitoring using this system was conducted from December 2006 to April 2007. As a result, from December 18, 2006, to March 17, 2007, similar behaviors reported by earlier studies were observed, such as sleeping with curled up posture and not eating, urinating or defecating. During this hibernation period and also around the time of hibernation, the prototype system continuously measured cyclic oscillations. The presence of cyclic vibrations at 8-sec intervals (about 7 bpm) was confirmed by the system before she entered hibernation on December 3, 2006. The respiratory rate gradually decreased, and during the hibernation period the respiratory rate was extremely low at approximately 2 bpm with almost no change. The results show that motion on the body surface caused by respiratory activity can be measured without touching the animal's body. Dwarf bamboo species are monocarpic. They flower simultaneously and die after several decades. The type of flowering in the
genus Sasa ranges from sporadic to gregarious. In order to determine whether or not the sporadic flowering of dwarf bamboo is fixed
genetically, we investigated the distribution of clones using eight microsatellite (SSR) loci in a sporadic flowering patch
of Sasa cernua Makino, a major dwarf bamboo species found in central Hokkaido. In May 2006, flowering occurred on 60.5% of living culms
in a 1600 m2 patch. We established a 50 × 10 m study plot in this patch and noted 1267 clumps consisting of 2529 living culms. We investigated
all 1267 clumps and identified six multilocus genotypes as clones using five variable SSR loci. All flowering clumps belonged
to the same clone. On the other hand, non-flowering vegetative clumps were also discovered to be of the same clone. These
data suggest that all flowering culms originated from a single clone of a sporadic flowering patch of S. cernua. Clonal analysis for investigation of sporadic flowering of S. cernua revealed that only a portion of a clone flowers and dies instead of the whole clone. 相似文献

The objective of this study is to determine the effects of substrate moisture and oxygen availability on growth traits of
Salix gracilistyla Miquel, which colonizes gravel bars along rivers, the shoot growth schedule, biomass production, and resource allocation
were examined under greenhouse conditions. We used four treatments representing a range of substrate moisture and oxygen availability:
drought (D), flooding with standing water (FS), flooding with running water (FR), and control without drought or flooding
(C). Cuttings in D stopped flushing and had low biomass production, reduced total leaf mass, and small leaves. Under anaerobic
conditions, cuttings in FS stopped flushing and had low biomass production, small root biomass, low biomass allocation to
roots, shallow roots, high biomass allocation to hypertrophied lenticels, and a few small, thick leaves. Under aerobic conditions,
cuttings in FR showed continuous branch elongation and flushing, large biomass production, and large leaf biomass, similar
to cuttings in C, in addition to low allocation to hypertrophied lenticels and many large leaves. The growth of cuttings was
not inhibited by flooding of the roots throughout the experiment unless the conditions were anaerobic. Thus, cuttings respond
to water stress under low moisture conditions by reducing the transpiration area and respond to flooding under low oxygen
conditions by high allocation to hypertrophied lenticels and reduced transpiration area. Plasticity in the shoot growth schedule,
biomass production, and resource allocation according to moisture conditions and the ability to develop hypertrophied lenticels
upon flooding allow S. gracilistyla to colonize sites in which both desiccation and flooding occur. 相似文献
