Small differences in syntopic Agalychnis callidryas and A. spurrelli oviposition sites and embryo predation via snakes and invertebrates provide context for a previously unexplainable difference in embryonic behavioral risk responses. Correlations in reproductive and ecological traits suggest directional selection in the syntopic populations of these species. Background

Rabies is a vaccine-preventable viral zoonosis belonging to the group of neglected tropical diseases. Exposure to a rabid animal may result in a fatal acute encephalitis if effective post-exposure prophylaxis is not provided. Rabies occurs worldwide, but its burden is disproportionately high in developing countries, including Nepal. We aimed to summarize current knowledge on the epidemiology, impact and control of rabies in Nepal.

Methods

We performed a systematic review of international and national scientific literature and searched grey literature through the World Health Organization Digital Library and the library of the National Zoonoses and Food Hygiene Research Centre, Nepal, and through searching Google and Google Scholar. Further data on animal and human rabies were obtained from the relevant Nepalese government agencies. Finally, we surveyed the archives of a Nepalese daily to obtain qualitative information on rabies in Nepal.

Findings

So far, only little original research has been conducted on the epidemiology and impact of rabies in Nepal. Per year, rabies is reported to kill about 100 livestock and 10–100 humans, while about 1,000 livestock and 35,000 humans are reported to receive rabies post-exposure prophylaxis. However, these estimates are very likely to be serious underestimations of the true rabies burden. Significant progress has been made in the production of cell culture-based anti-rabies vaccine and rabies immunoglobulin, but availability and supply remain a matter of concern, especially in remote areas. Different state and non-state actors have initiated rabies control activities over the years, but efforts typically remained focalized, of short duration and not harmonized. Communication and coordination between veterinary and human health authorities is limited at present, further complicating rabies control in Nepal. Important research gaps include the reporting biases for both human and animal rabies, the ecology of stray dog populations and the true contribution of the sylvatic cycle.

Interpretation

Better data are needed to unravel the true burden of animal and human rabies. More collaboration, both within the country and within the region, is needed to control rabies. To achieve these goals, high level political commitment is essential. We therefore propose to make rabies the model zoonosis for successful control in Nepal.  相似文献

Oxidative stress from ozone (O(3)) exposure augments airway neutrophil recruitment and chemokine production. We and others have shown that severe and sudden asthma is associated with airway neutrophilia, and that O(3) oxidative stress is likely to augment neutrophilic airway inflammation in severe asthma. However, very little is known about chemokines that orchestrate oxidative stress-induced neutrophilic airway inflammation in vivo. To identify these chemokines, three groups of BALB/c mice were exposed to sham air, 0.2 ppm O(3), or 0.8 ppm O(3) for 6 h. Compared with sham air, 0.8 ppm O(3), but not 0.2 ppm O(3), induced pronounced neutrophilic airway inflammation that peaked at 18 h postexposure. The 0.8 ppm O(3) up-regulated lung mRNA of CXCL1,2,3 (mouse growth-related oncogene-alpha and macrophage-inflammatory protein-2), CXCL10 (IFN-gamma-inducible protein-10), CCL3 (macrophage-inflammatory protein-1alpha), CCL7 (monocyte chemoattractant protein-3), and CCL11 (eotaxin) at 0 h postexposure, and expression of CXCL10, CCL3, and CCL7 mRNA was sustained 18 h postexposure. O(3) increased lung protein levels of CXCL10, CCL7, and CCR3 (CCL7R). The airway epithelium was identified as a source of CCL7. The role of up-regulated chemokines was determined by administering control IgG or IgG Abs against six murine chemokines before O(3) exposure. As expected, anti-mouse growth-related oncogene-alpha inhibited neutrophil recruitment. Surprisingly, Abs to CCL7 and CXCL10 also decreased neutrophil recruitment by 63 and 72%, respectively. These findings indicate that CCL7 and CXCL10, two chemokines not previously reported to orchestrate neutrophilic inflammation, play a critical role in mediating oxidative stress-induced neutrophilic airway inflammation. These observations may have relevance in induction of neutrophilia in severe asthma.  相似文献

In this report, Ag nanoparticles were fabricated using the single-step glancing angle deposition (SS-GLAD) technique upon In2O3/TiO2 thin film. Afterward, a detailed analysis was done for the two samples such as In2O3/TiO2 thin film and In2O3/TiO2 thin film/Ag nanoparticles, to inspect the field emission scanning electron microscopy (FESEM), energy-dispersive X-ray analysis (EDAX), X-ray diffraction (XRD), ultraviolet (UV) spectroscopy, and electrical properties. The reduction in bandgap energy for the samples of In2O3/TiO2 thin film/Ag nanoparticles (~4.16 eV) in comparison with the In2O3/TiO2 thin film (~4.28 eV) was due to trapped e–h recombination at the oxygen vacancies and electron transmission of Ag to the conduction band of the In2O3/TiO2 thin films. Moreover, under irradiation of photons Ag nanoparticles generated inorganic Ag–O compound attributable to the localized surface plasmon resonance (LSPR). Also, a?~90% high transmittance,?~60% and?~25% low reflectance in UV and visible region, fill factor (FF) of 53%, as well as power conversion efficiency (PCE) of 15.12% was observed for In2O3/TiO2 thin film/Ag nanoparticles than the In2O3/TiO2 thin film. Therefore, the use of Ag nanoparticles textured In2O3/TiO2 thin film–based device is a promising approach for the forthcoming photovoltaic applications.

Objective

Transmission patterns of drug-resistant Mycobacterium tuberculosis (MTB) may be influenced by differences in socio-demographics, local tuberculosis (TB) endemicity and efficaciousness of TB control programs. This study aimed to investigate the impact of DOTS on the transmission of drug-resistant TB in eastern rural China.

Methods

We conducted a cross-sectional study of all patients diagnosed with drug-resistant TB over a one-year period in two rural Chinese counties with varying lengths of DOTS implementation. Counties included Deqing, with over 11 years'' DOTS implementation and Guanyun, where DOTS was introduced 1 year prior to start of this study. We combined demographic, clinical and epidemiologic information with IS6110-based restricted fragment length polymorphism (RFLP) and Spoligotyping analysis of MTB isolates. In addition, we conducted DNA sequencing of resistance determining regions to first-line anti-tuberculosis agents.

Results

Of the 223 drug-resistant isolates, 73(32.7%) isolates were identified with clustered IS6110RFLP patterns. The clustering proportion among total drug-resistant TB was higher in Guanyun than Deqing (26/101.vs.47/122; p,0.04), but not significantly different among the 53 multidrug-resistant isolates (10/18.vs.24/35; p,0.35). Patients with cavitary had increased risk of clustering in both counties. In Guanyun, patients with positive smear test or previous treatment history had a higher clustering proportion. Beijing genotype and isolates resistant to isoniazid and/or rifampicin were more likely to be clustered. Of the 73 patients with clustered drug-resistant isolates, 71.2% lived in the same or neighboring villages. Epidemiological link (household and social contact) was confirmed in 12.3% of the clustered isolates.

Conclusion

Transmission of drug-resistant TB in eastern rural China is characterized by small clusters and limited geographic spread. Our observations highlight the need for supplementing DOTS with additional strategies, including active case finding at the village level, effective treatment for patients with cavities and drug susceptibility testing for patients at increased risk for drug-resistance.  相似文献

We report on the characterization of a group of seven novel Brucella strains isolated in 1964 from three native rodent species in North Queensland, Australia, during a survey of wild animals. The strains were initially reported to be Brucella suis biovar 3 on the basis of microbiological test results. Our results indicated that the rodent strains had microbiological traits distinct from those of B. suis biovar 3 and all other Brucella spp. To reinvestigate these rodent strains, we sequenced the 16S rRNA, recA, and rpoB genes and nine housekeeping genes and also performed multiple-locus variable-number tandem-repeat (VNTR) analysis (MLVA). The rodent strains have a unique 16S rRNA gene sequence compared to the sequences of the classical Brucella spp. Sequence analysis of the recA, rpoB, and nine housekeeping genes reveals that the rodent strains are genetically identical to each other at these loci and divergent from any of the currently described Brucella sequence types. However, all seven of the rodent strains do exhibit distinctive allelic MLVA profiles, although none demonstrated an amplicon for VNTR 07, whereas the other Brucella spp. did. Phylogenetic analysis of the MLVA data reveals that the rodent strains form a distinct clade separate from the classical Brucella spp. Furthermore, whole-genome sequence comparison using the maximal unique exact matches index (MUMi) demonstrated a high degree of relatedness of one of the seven rodent Brucella strains (strain NF 2653) to another Australian rodent Brucella strain (strain 83-13). Our findings strongly suggest that this group of Brucella strains isolated from wild Australian rodents defines a new species in the Brucella genus.Brucella species are facultative intracellular Gram-negative members of the Alphaproteobacteria class capable of causing brucellosis in a range of animal hosts, including domesticated livestock, wildlife, marine mammals, and humans (1, 5, 7, 29, 32, 33, 36, 47). Brucellosis is the most prevalent zoonotic disease worldwide, causing spontaneous abortion and fetal death in animals and severe flu-like symptoms, focal complications, and often, chronic disease in humans (7, 11, 22, 27, 40, 41, 49, 50). Brucella species are typically transmitted to humans through consumption of unpasteurized dairy products or exposure to fluids or tissues from infected animals (45, 49). Animals are primary hosts of all Brucella spp., which include Brucella abortus (cattle), B. canis (dogs), B. melitensis (goats, cows, and sheep), B. suis (swine), B. ovis (rams), and B. neotomae (desert rats) (3, 7, 8). Recently, three additional Brucella species have been recognized: B. pinnipedialis (seals), B. ceti (dolphins) (5), and B. microti. B. microti was initially isolated from the common vole in the Czech Republic (33, 35). In the mid-1980s, DNA-DNA hybridization studies demonstrated a very high level of genetic similarity (98.5%) among the Brucella spp., which led to the adoption of a monospecies concept for the Brucella genus, with all the species at that time renamed as biovars of B. melitensis (46). However, 20 years later, the use of a phylogenetic-evolutionary approach to Brucella taxonomy was accepted. By that approach, host preferences, virulence, and pathogenicity were considered important criteria in the delineation of Brucella species, and consequently, the multispecies taxonomy was restored to the Brucella genus (28). With the development of more advanced molecular typing methodologies such as multilocus sequence analysis (MLSA) (48), multiple-locus variable-number tandem-repeat (VNTR) analysis (MLVA) (23), and techniques interrogating single nucleotide polymorphisms (SNPs) (17, 47), Brucella spp. can be quickly genotyped and strains can be readily examined for their phylogenetic and evolutionary relationships (15).Recently, we reported on two unusual human brucellosis cases, one of which led to the identification of a novel species, Brucella inopinata, whose type strain is strain BO1 and which was associated with a breast implant infection in a patient in Oregon (12, 36). The second brucellosis case involved an atypical Brucella strain (strain BO2) isolated from the lung biopsy fluid of a patient with chronic destructive pneumonia in Australia (44). Because both patients denied common risk factors associated with human brucellosis, the primary hosts of these strains remain unknown. However, nucleotide sequence analysis of the outer membrane proteins (omp2a and omp2b) of both strain BO2 and strain BO1T demonstrated close clustering to an atypical B. suis strain (strain 83-210) isolated from a rodent in Australia (30, 44). Further genetic analysis of the 16S rRNA genes from strains BO1T and BO2 and the 16S rRNA gene from atypical Brucella strain 83-13 (available at http://www.broadinstitute.org), which was isolated from a rodent in Australia and briefly described by Corbel and Brinley-Morgan in 1984 (9), showed that strain 83-13 yielded notable genetic similarity to the novel human isolates, which led us to speculate that atypical human Brucella strains BO1T and BO2 may have an animal reservoir in rodents from Australia.Rodent brucellosis is self-limiting and is mostly associated with wild rodents that cohabitate among domestic livestock presumably infected with classical Brucella spp. (27). Over 22 different wild rodent species worldwide have been reported to be susceptible to Brucella infection, as demonstrated by serology and/or culture (41). Earlier field studies in Argentina, Venezuela, and Denmark reported on the prevalence of B. suis in hares, opossums, and rats and B. abortus in ferrets and capybaras (10, 11, 27). However, two rodent-specific Brucella spp. have been identified, including B. neotomae, isolated from the desert wood rat (Neotomae lepida) in Utah (39), and most recently, B. microti, isolated from the common vole (Microtus arvalis) in the Czech Republic (35). In the early 1960s, Cook et al. reported on the isolation and biochemical identification of seven Brucella suis biovar 3 strains from three known species of wild native rats from Australia (6). We reinvestigated the microbiological characteristics of these seven rodent B. suis biovar 3 strains and performed genetic analyses with respect to the microbiological characteristics and genetics of the classical and atypical Brucella species. In this report, we describe this group of Brucella strains isolated from wild rodents in Australia, which confer unique microbiological and molecular characteristics distinct from those of any of the currently described species.  相似文献

The combination of founder events, random drift and new selective forces experienced by introduced species typically lowers genetic variation and induces differentiation from the ancestral population. Here, we investigate microsatellite differentiation between introduced and native populations of the small Indian mongoose (Herpestes auropunctatus). Many expectations based on introduction history, such as loss of alleles and relationships among populations, are confirmed. Nevertheless, when applying population assignment methods to our data, we observe a few specimens that are incorrectly assigned and/or appear to have a mixed ancestry, despite estimates of substantial population differentiation. Thus, we suggest that population assignments of individuals should be viewed as tentative and that there should be agreement among different algorithms before assignments are applied in conservation or management. Further, we find no congruence between previously reported morphological differentiation and the sorting of microsatellite variation. Some introduced populations have retained much genetic variation while others have not, irrespective of morphology. Finally, we find alleles from the sympatric grey mongoose (Herpestes edwardsii) in one small Indian mongoose within the native range, suggesting an alternative explanation for morphological differentiation involving a shift in female preferences in allopatry.  相似文献

A set of heat conduction equations for a two-component medium simulating biological tissues were formulated. Their solutions were obtained, and the spatial distribution of light and temperature over tissue depth at different times after irradiation by a short light pulse was studied. The local absorption of light by blood vessels and the influence of this effect on the optical parameters of the medium, a more intense heating of blood as compared with its surrounding tissue, heat exchange between them, and heat transfer at the interface with different environments were accounted for. The solutions are expressed via characteristic times of the respective thermal processes to enable one to easily and vividly analyze the features of tissue heating and the influence of optical and thermal parameters on the temperature distributions of the components. A survey of foraminiferal microhabitats at several Gulf of Mexico bathyal/abyssal hydrocarbon seeps reveals that many epibenthic species live not on the sediment, but attached to vestimentiferan tubeworms – centimeters to decimeters above the seafloor – thus avoiding the oxygen depletion and H2S toxicity at the sediment–water interface. This observation explains how certain species with a relatively high oxygen requirement (e.g., Cibicides spp.) may become components of foraminiferal death assemblages in seep sediments. Thus, when the sedimentary record of Foraminifera is used to interpret the history of past methane venting, the species from elevated microhabitats cannot be ignored, but they need to be considered separately from the sediment dwellers.  相似文献

MOTIVATION: Application of mass spectrometry in proteomics is a breakthrough in high-throughput analyses. Early applications have focused on protein expression profiles to differentiate among various types of tissue samples (e.g. normal versus tumor). Here our goal is to use mass spectra to differentiate bacterial species using whole-organism samples. The raw spectra are similar to spectra of tissue samples, raising some of the same statistical issues (e.g. non-uniform baselines and higher noise associated with higher baseline), but are substantially noisier. As a result, new preprocessing procedures are required before these spectra can be used for statistical classification. RESULTS: In this study, we introduce novel preprocessing steps that can be used with any mass spectra. These comprise a standardization step and a denoising step. The noise level for each spectrum is determined using only data from that spectrum. Only spectral features that exceed a threshold defined by the noise level are subsequently used for classification. Using this approach, we trained the Random Forest program to classify 240 mass spectra into four bacterial types. The method resulted in zero prediction errors in the training samples and in two test datasets having 240 and 300 spectra, respectively.  相似文献
