In the present study, we evaluated the immunopotentiating efficacy of tuftsin against experimental murine aspergillosis in both normal and immunodebilitant BALB/c mice. The animals were challenged with an isolate of Aspergillus fumigatus (1x10(8) cfu/mouse) that was showing less susceptibility to lower doses of amphotericin B in murine animal model. Co-administration of the immunomodulator tuftsin and liposomised-amphotericin B was found to be highly effective in the treatment of systemic infection of A. fumigatus in both immunocompetent and leukopenic mice. Moreover, pre-treatment of mice with liposomised-tuftsin prior to challenging them with A. fumigatus infection and subsequent treatment with tuftsin-bearing liposomised-amphotericin B was found to be extremely efficient in successful elimination of fungal pathogen. In another set of experiments, tuftsin-mediated antigen-specific memory antibody response was also assessed by immunizing the animals with A. fumigatus cytosolic antigen. The animals that received a booster 150 days after the first immunization with tuftsin-liposomes-antigen showed more resistance to A. fumigatus infection in comparison with the na?ve animals.  相似文献

The ethanolic extract derived from aerial parts of an indigenous medicinal plant Paeonia emodi was screened for enzyme inhibition activities against Urease (jack bean and Bacillus pasteurii) and alpha-Chymotrypsin. The extract was also investigated for its radical scavenging activity using DPPH assay. The crude extract was found to possess significant enzyme inhibition activities against jack bean (74%) and Bacillus pasteurii (80%) urease and a moderate activity (54%) against alpha-Chymotrypsin. The extract also displayed excellent (83%) radical scavenging activity. On the basis of these results, the crude extract was subsequently fractionated into n-hexane, chloroform, ethyl acetate, n-butanol and water fractions and tested independently for the aforesaid activities. Significant inhibitory activity against urease enzyme was observed for the ethyl acetate, n-butanol and water fractions while the n-hexane and chloroform fractions were devoid of any such activity. In the alpha-Chymotrypsin enzyme inhibition studies the activity was concentrated into the ethyl acetate fraction. All the fractions displayed potent radical scavenging activity. The crude extract and fractions thereof were also subjected to total phenolic content determination. A correlation between radical scavenging capacities of extracts and total phenolic content was observed in the majority of cases.  相似文献

A study was conducted to compare the ensiling characteristics, chemical composition, and the ruminal and total tract nutrient degradabilities of leafy (Cargill F227) and brown midrib (Mycogen TMF94) corn silage hybrids. Corn was grown in Saint-Jean-sur-Richelieu, Quebec, Canada, harvested at a target 350 g kg(-1) dry matter (DM) content, and ensiled in mini-silos for 0, 2, 4, 8, 16, and 45 d. Two non-lactating Holstein cows fitted with ruminal and proximal duodenal cannulae were used to determine ruminal and whole tract nutrient degradability. Forage from both hybrids went through a rapid fermentation with a sharp decline in pH during the first 2 d of ensiling, pH in both silage being less than 4.0 after 45 d. Lactic acid concentration was however greater for leafy than brown midrib corn. Chemical analysis of silage after 45 d of ensiling revealed that hybrids differed in their composition. Compared to leafy corn, brown midrib corn had lower neutral detergent fiber (NDF), acid detergent fiber (ADF), acid detergent lignin (ADL), crude protein (CP), and neutral detergent and acid detergent insoluble proteins, but higher starch and net energy of lactation (NEL) values. Results of the in situ incubation experiment indicated that compared to leafy corn brown midrib corn had greater ruminal DM (64 vs. 54%), CP (73 vs. 71%), and NDF (32 vs. 24%) degradabilities. Brown midrib corn silage also had greater DM ruminal (53 vs. 48%) and total tract (67 vs. 61%) digestibilities, as well as greater NDF ruminal (34 vs. 25%), intestinal (10 vs. 8%), and total tract (43 vs. 33%) digestibilities. Type of corn hybrid will thus greatly affect silage chemical composition and nutrient digestibility.  相似文献

The reactions of the half-open trozircene [(η7-C7H7)Zr(η5-2,4-C7H11)] (1) with the two-electron donor ligands tert-butyl isocyanide (CN-tBu), 1,2-bis(dimethylphosphino)ethane (dmpe), trimethylphosphine (PMe3) and 1,3,4,5-tetramethylimidazolin-2-ylidene (IMe, :C[N(Me)C(Me)]2) have led to the 1:1 adducts 3, 4, 5 and 6, respectively. The latter three were structurally characterized by X-ray diffraction analysis. Additionally, the stability of the adducts was probed by DFT calculations employing the B3LYP and M05-2X functionals showing that the strongly σ-basic N-heterocyclic carbene forms a thermodynamically much more stable adduct than the other three.  相似文献

In marine ecosystems, fluctuations in surface-seawater carbon dioxide (CO2), significantly influence the whole metabolism of marine algae, especially during the early stages of macroalgal development. In this study, the response of the green alga Ulva fasciata for elevating ocean acidification was investigated using four levels of pCO2 ~ 280, 550, 750 and 1050 µatm. Maximum growth rate (6.6% day−1), protein (32.43 %DW) and pigment (2.9 mg/g) accumulation were observed at pCO2-550 with an increase of ~2-fold compared to control. On the other hand, lipid and carbohydrate contents recorded their maximum production (4.23 and 46.96 %DW, respectively) at pCO2-750 while control showed 3.70 and 42.37 %DW, respectively. SDS-PAGE showed the presence of unique bands in response to pCO2, especially at 550 µatm. Dominant associated bacteria was shifted from Halomonas hydrothermalis of control to Vibrio toranzoniae at pCO2-1050. These findings suggest that ocean acidification at 550 µatm might impose noticeable effects on growth, protein, pigments, and protein profile of U. fasciata, which could be a good source for fish farming. While, pCO2-750 was recommended for energetic purpose, due to its high lipid and carbohydrate contents.  相似文献

Noncompetitive NMDA receptor antagonist (+)MK-801 is known to induce neurotoxicity and schizophrenia-like symptomatology where atypical neuroleptic clozapine is effective in contrast to typical neuroleptic, haloperidol. Although neuropeptides are implicated in memory and cognition, their roles in schizophrenia are not well understood. In the present study, we therefore examined the possible roles of neuropeptides, cholecystokinin (CCK) and somatostatin (SS) in the posterior cingulate/retrosplenial cortices (PC/RSC), frontal cortex, and hippocampus of a MK-801-induced schizophrenia-like model rat brain. This study further investigated the pretreated effect of atypical versus typical neuroleptics on the peptidergic system. SS mRNA and peptide levels significantly decreased in the PC/RSC and hippocampus but not in the frontal cortex 3 days after 0.5 mg/kg MK-801 treatment whereas CCK mRNA and peptide levels significantly decreased in all of the brain regions examined. Pretreatment with clozapine but not haloperidol completely recovered the changes in both mRNA and peptide levels of SS and CCK in those brain regions. These data suggest that peptidergic system in the brain presumably plays an important role in the control of negative schizophrenia.  相似文献

In the present study, we have demonstrated the suitability of microspheres in removal of plasma bilirubin from systemic circulation of hyperbilirubinemic rats. Poly (lactide co-glycolide) microspheres (PLGA microspheres) have been shown to bind with bilirubin in both a concentration and time dependent manner. The binding affinity of bilirubin to microspheres was enhanced when rat serum albumin (RSA) was loaded into the microspheres. On evaluating the potential of microspheres in elimination of bilirubin from the systemic circulation, RSA bearing microspheres were found to be competent in both removing bilirubin from the systemic circulation and controlling elevated plasma levels of liver function enzymes in temporarily hyperbilirubinemic rats. On the basis of results of the present study, we suggest that microsphere-based delivery system may help in development of safe, effective and alternate strategy for the treatment of hyperbilirubinemic conditions in model animals.  相似文献

Inhibition of Bacillus pasteurii urease enzyme by 3,7,15-tri-O-acetyl-5-O-nicotinoyl-13,14-dihydroxymyrsinol (1), a diterpene ester with a myrsinol-type skeleton, isolated from Euphorbia decipiens Boiss. and Buhse, was un-competitive consistent with the molecular docking results.
