Summary By application of theBodenheimer's method, in taking 10°C for the threshold of the parasit development, the number of days/degrees necessary for a mean evolution
ofProspaltella has been calculated. With this method, the theoretical number of generations is identical with that observed in several orchards
of some countries. This method is suggested to facilitate the use ofProspaltella by fixing the time necessary to show the parasite's efficiency.
Determinations of carbohydrases, proteases, carboxylesterases and phosphatases in the midgut cells and in the luminal spaces outside and inside the peritrophic membrane of Rhynchosciara americana larvae have been carried out. The data show that alpha-amylase, cellulase and proteinases are present in cells, ecto- and endoperitrophic spaces; aminopeptidases and trehalase in cells and ectoperitrophic space; and finally disaccharidases (except trehalase), carboxypeptidases, dipeptidases, carboxylesterases and phosphatases only in cells. The results support the conclusion that digestion takes place in three spatially organized steps. The first one occurs inside the peritrophic membrane and comprises the dispersion and/or decrease in molecular weight of the food molecules. The second is the hydrolysis of the polymeric food molecules in the ectoperitrophic space to dimers and/or small oligomers. Finally, terminal digestion occurs in the midgut caeca and posterior ventriculus cells by enzymes presumed to be plasma membrane bound. The existence of two extracellular sites for digestion in R. americana is considered to be an adaptation to conserve secreted enzymes, since only those penetrating the endoperitrophic space are lost quickly in the faeces. 相似文献
A fructose-rich diet can induce metabolic syndrome, a combination of health disorders that increases the risk of diabetes and cardiovascular diseases. Diet is also known to alter the microbial composition of the gut, although it is not clear whether such alteration contributes to the development of metabolic syndrome. The aim of this work was to assess the possible link between the gut microbiota and the development of diet-induced metabolic syndrome in a rat model of obesity. Rats were fed either a standard or high-fructose diet. Groups of fructose-fed rats were treated with either antibiotics or faecal samples from control rats by oral gavage. Body composition, plasma metabolic parameters and markers of tissue oxidative stress were measured in all groups. A 16S DNA-sequencing approach was used to evaluate the bacterial composition of the gut of animals under different diets. The fructose-rich diet induced markers of metabolic syndrome, inflammation and oxidative stress, that were all significantly reduced when the animals were treated with antibiotic or faecal samples. The number of members of two bacterial genera, Coprococcus and Ruminococcus, was increased by the fructose-rich diet and reduced by both antibiotic and faecal treatments, pointing to a correlation between their abundance and the development of the metabolic syndrome. Our data indicate that in rats fed a fructose-rich diet the development of metabolic syndrome is directly correlated with variations of the gut content of specific bacterial taxa. 相似文献
The DEG/ENaC ion channel family contributes to channels of striking functional diversity. Neuronally expressed family members include the C. elegans degenerins that mediate touch and are thought to be mechanically gated, and the mammalian ASICs, which are gated by protons. ASICs affect a range of sensory functions that includes perception of gentle touch, harsh touch, heat, sour taste, and pain. The AKin10 gene from Arabidopsis thaliana encoding a putative Ser/Thr protein kinase (PK) has been isolated and characterized. The AKin10-encoding gene is located on a genomic 5.4-kb BamHI fragment and contains ten introns, one being located in the 5' untranslated region. The deduced amino acid sequence of AKin10 is 65% identical over the catalytic domain to the yeast PK (SNF1). SNF1 is essential for the derepression of many glucose-repressible genes, including Suc2 which encodes invertase. Southern blot hybridization experiments suggested the presence of one copy of the gene per haploid genome of A. thaliana. Northern hybridization experiments indicated that this gene is expressed in roots, shoots and leaves. AKin10 may play an important role in a signal transduction cascade regulating gene expression and carbohydrate metabolism in higher plants. 相似文献
UVirradiation of dissolved organic carbon (DOC) in the laboratory can producesmall, labile organic compounds utilizable by microbes, but few studies haveattempted to document this process in situ. 13Cnuclear magnetic resonance (NMR) was used to examine the bulk chemicalcomposition of natural and laboratory-irradiated high-molecular-weight DOC(HMW-DOC) from shaded (150 mol m–2s–1 average light in surface water) and open (1500mol m–2 s–1) field sitesoverone and a half years. 13C NMR revealed only small differences incarbon functional groups between laboratory irradiated and non-irradiatedHMW-DOC. However, bacterial protein productivity per cell (BPP) was enhanced innaturally irradiated samples of HMW-DOC in a field mesocosm experiment (p <0.05), suggesting that bacterial growth was enhanced by photochemicalproductionof labile DOC substrates. Absorbance characteristics such as spectral slope,absorbance at 350 nm, and the absorbance ratio 250nm/365 nm revealed that HMW-DOC was photoreactive,yetno differences in these values were found between samples irradiated with andwithout UV-B. In experiments conducted with simulated solar radiation in thelaboratory and with natural light in the field mesocosm experiment, UV-A(320–400 nm) and photosynthetically active radiation (PAR;400–700 nm) were more effective than UV-B (280–320nm) in HMW-DOC photolysis. 相似文献
Increasing evidence suggests that trabecular meshwork (TM) cells participate in the regulation of intraocular pressure by controlling the rate of filtration of the aqueous humor. Ionic conductances that regulate cell volume and shape have been suggested to play an important role in TM cell volume regulation. Here, we compared ionic currents from TM cells derived from a normal subject (CTM) and from an individual affected by glaucoma (GTM). We found that while the ionic current types were similar, the current amplitudes and percentage of cells endowed with specific current at baseline were different in the two cell lines. Thus, we found that the majority of CTM cells were endowed with a swelling-activated Cl? current at baseline, whereas in the majority of GTM cells this current was not active at baseline and became activated only after perfusion with a hypotonic solution. An inward rectifier K+ current was also more prevalent in CTM than in GTM cells. Our work suggests that disregulation of one or more of these ionic currents may be at the basis of TM cell participation in the development of glaucoma. 相似文献
Stroke is a leading cause of death and disability worldwide. Several mechanisms are involved in the pathogenesis of ischemic stroke (IS). The contributory role of the inflammatory and immunity processes was demonstrated both in vitro and in animal models, and was confirmed in humans. IS evokes an immediate inflammatory response that involves complex cellular and molecular mechanisms. All components of the innate and adaptive immunity systems are involved in several steps of the ischemic cascade. In the early phase, inflammatory and immune mechanisms contribute to the brain tissue damage, whereas, in the late phase, they participate to the tissue repair processes. In particular, damage-associated molecular patterns (DAMPs) appear critical for the promotion of altered blood brain barrier permeability, leukocytes infiltration, tissue edema and brain injury. Conversely, the activation of regulatory T lymphocytes (Tregs) plays protective effects. The identification of specific cellular/molecular elements belonging to the inflammatory and immune responses, contributing to the brain ischemic injury and tissue remodeling, offers the advantage to design adequate therapeutic strategies. In this article, we will present an overview of the knowledge on inflammatory and immunity processes in IS, with a particular focus on the role of DAMPs and leukocytes infiltration. We will discuss evidence obtained in preclinical models of IS and in humans. The main molecular mechanisms useful for the development of novel therapeutic approaches will be highlighted. The translation of experimental findings to the human disease is still a difficult step to pursue. Further investigations are required to fill up the existing gaps.
