Vernal pools are small, seasonal wetlands that are a common landscape feature contributing to biodiversity in northeastern North American forests. Basic information about their biogeochemical functions, such as carbon cycling, is limited. Concentrations of dissolved methane (CH4) and carbon dioxide (CO2) and other water chemistry parameters were monitored weekly at the bottom and surface of four vernal pools in central and eastern Maine, USA, from April to August 2016. The vernal pools were supersaturated with respect to CH4 and CO2 at all sampling dates and locations. Concentrations of dissolved CH4 and CO2 ranged from 0.4 to 210 μmol L?1 and 72–2300 μmol L?1, respectively. Diffusive fluxes of CH4 and CO2 into the atmosphere ranged from 0.2 to 73 mmol m?2 d?1, and 30–590 mmol m?2 d?1, respectively. During the study period, the four vernal pools emitted 0.1–5.8 kg C m?2 and 9.6–120 kg C m?2 as CH4 and CO2, respectively. The production fluxes (production rates normalized to surface area) of CH4 and CO2 ranged from ? 0.02 to 0.66 and 0.40–4.6 g C m?2 d?1, respectively, and increased significantly over the season. Methane concentrations were best predicted by alkalinity, ortho-phosphate and depth, while CO2 concentrations were best predicted with only alkalinity. Alkalinity as a predictor variable highlights the importance of anaerobic respiration in production of both gases. Our study pools had large concentrations and effluxes of CH4 and CO2 compared to permanently inundated wetlands, indicating vernal pools are metabolically active sites and may be important contributors to the global carbon budget.  相似文献

Butterfly communities can be modified by the activities of large mammals. The wild boar (Sus scrofa) is one of the most common mammals in Europe and has notably expanded its distribution range in recent decades. The present work aimed to investigate the possible effect of wild boar rooting activity on butterflies in olive groves inside the “Cilento, Vallo di Diano and Alburni” National Park. Butterflies were surveyed in five Rooted (R) and five Control areas (C). Transects were made from April to September in 2011 and 2012. Forty-three species and 3659 individuals were found, of which 39 species and 2426 individuals were detected in C areas and 31 species and 1233 individuals in R areas. A community-level approach showed a significantly higher abundance, richness and Shannon–Wiener index in C than in R. An ecological-level approach indicated that more specialised butterflies were significantly more affected by wild boar activity. Finally, the response of single species was investigated: two species (Colias croceus and Polyommatus icarus) were significantly more represented in R, whereas six species (Aricia agestis, Hipparchia statilinus, Lasiommata megera, Melanargia arge, Pyronia cecilia and Thymelicus acteon) were significantly more abundant in C. Three species considered at risk (M. arge; Annex II of Directive 92/43/EEC; H. statilinus and T. acteon: European Red List) were less represented in rooted areas. Overall, the intensive wild boar activity negatively correlated with butterflies in the olive groves studied.  相似文献

Cytolethal distending toxins (CDTs) are tripartite protein exotoxins produced by a diverse group of pathogenic Gram-negative bacteria. Based on their ability to induce DNA damage, cell cycle arrest, and apoptosis of cultured cells, CDTs are proposed to enhance virulence by blocking cellular division and/or directly killing epithelial and immune cells. Despite the widespread distribution of CDTs among several important human pathogens, our understanding of how these toxins interact with host cells is limited. Here we demonstrate that CDTs from Haemophilus ducreyi, Aggregatibacter actinomycetemcomitans, Escherichia coli, and Campylobacter jejuni differ in their abilities to intoxicate host cells with defined defects in host factors previously implicated in CDT binding, including glycoproteins, and glycosphingolipids. The absence of cell surface sialic acid sensitized cells to intoxication by three of the four CDTs tested. Surprisingly, fucosylated N-linked glycans and glycolipids, previously implicated in CDT-host interactions, were not required for intoxication by any of the CDTs tested. Finally, altering host-cellular cholesterol, also previously implicated in CDT binding, affected intoxication by only a subset of CDTs tested. The findings presented here provide insight into the molecular and cellular basis of CDT-host interactions.  相似文献

Introduction

Impaired walking performance is a key predictor of morbidity among older adults. A distinctive characteristic of impaired walking performance among older adults is a greater metabolic cost (worse economy) compared to young adults. However, older adults who consistently run have been shown to retain a similar running economy as young runners. Unfortunately, those running studies did not measure the metabolic cost of walking. Thus, it is unclear if running exercise can prevent the deterioration of walking economy.

Purpose

To determine if and how regular walking vs. running exercise affects the economy of locomotion in older adults.

Methods

15 older adults (69±3 years) who walk ≥30 min, 3x/week for exercise, “walkers” and 15 older adults (69±5 years) who run ≥30 min, 3x/week, “runners” walked on a force-instrumented treadmill at three speeds (0.75, 1.25, and 1.75 m/s). We determined walking economy using expired gas analysis and walking mechanics via ground reaction forces during the last 2 minutes of each 5 minute trial. We compared walking economy between the two groups and to non-aerobically trained young and older adults from a prior study.

Results

Older runners had a 7–10% better walking economy than older walkers over the range of speeds tested (p = .016) and had walking economy similar to young sedentary adults over a similar range of speeds (p = .237). We found no substantial biomechanical differences between older walkers and runners. In contrast to older runners, older walkers had similar walking economy as older sedentary adults (p = .461) and ∼26% worse walking economy than young adults (p<.0001).

Conclusion

Running mitigates the age-related deterioration of walking economy whereas walking for exercise appears to have minimal effect on the age-related deterioration in walking economy.  相似文献

p33ING2 belongs to the ING-gene family that is involved in tumor suppression, DNA repair, cell cycle regulation, and cellular senescence. Most functions are dependent on the tumor suppressor p53. p33ING2 was also shown to bind to trimethylated lysine 4 of histone H3. Here, we show that p33ING2 contains a transferable silencing function, which is independent of p53. p33ING2-mediated gene silencing is resistant to the HDAC-inhibitor trichostatin A indicating that p33ING2 uses a non-HDAC class I or II pathway for gene repression in reporter assays. In line with that we show that p33ING2 is associated with histone methyltransferase (HMT) activity in vitro and in vivo, methylating specifically histone H3. Interestingly, the specificity is distinct from the MeCP2-recruited HMT. Mutation or methylation of lysine 9, a mark well known for repression, abrogates histone methylation by MeCP2 but not by the p33ING2 complex. Instead, the ING2-associated HMT shows an increased methylation activity if lysine 9 is methylated. In contrast, mutation or methylation of lysine 4, a methylation preferentially detected at active genes, led to a reduction of the ING2-associated HMT. Notably, also p33ING1 recruits HMT activity suggesting a more general biochemical interaction between members of p33ING family and HMT activity. Deletion analyses revealed that the ING2 C-terminus recruits HMT activity, which correlates with silencing function.  相似文献

We studied five eutrophic (high phosphorus) and six mesotrophic/oligotrophic (low phosphorus) lakes in Maine, USA, all of which are dimictic and develop anoxic hypolimnia during stratification. The lakes were sampled during the stratified period from May to September 1999. Late summer hypolimnetic total phosphorus (P) concentrations in the high-P lakes ranged from 185 to 460ppb; epilimnetic total P increased up to 30ppb from the spring to the fall overturn. During the same period, the low-P lakes had hypolimnetic total P concentrations in the range of 6–19ppb.Individual high-P lakes demonstrated strong temporal correlations between aqueous hypolimnetic dissolved Fe and total P concentrations (R 20.88) with an average molar Fe:P ratio of 11.9±4.2. For the combined data, the high-P lakes exhibited strong correlation between hypolimnetic Fe and P concentrations (R 2=0.82). The low-P lakes, however, did not show a good correlation between the hypolimnetic Fe and P concentrations. Among the low-P lakes two lakes had hypolimnetic Fe fluxes comparable to the Fe fluxes of the high-P lakes. These two lakes had considerably higher hypolimnetic Fe:P ratios than all other lakes studied here. There were no significant differences in surface sediment Fe(III) or P fractions that correlated with the differences in the relationship between aqueous concentrations of Fe and P in these two outlier low-P lakes. A model for the generation of hypolimnetic acid neutralization capacity (ANC) was developed based on microbially-catalyzed reduction of Fe(III) hydroxide, Mn(IV) oxide and sulfate. Reduction of Fe(III) hydroxide was the most important contributor to the increase in the hypolimnetic ANC in all high-P and the two outlier lakes. Assuming that all hypolimnetic P was due to the reduction of Fe(III) hydroxide by bacteria and sulfide, average summer hypolimnetic P flux for each lake was predicted using the sediment reducible Fe(III):P ratio. The observed and predicted average P fluxes in the high-P lakes corresponded reasonably, suggesting that in these lakes internal P release is closely related to the reduction of Fe(III) hydroxide. Other release or sequestration mechanisms may operate for the release and availability of P in the low-P lakes.  相似文献
