Designing Green Supply Chains (GSCs) requires complex decision-support models that can deal with multiple dimensions of sustainability while taking into account specific characteristics of products and their supply chain. Multi-Criteria Decision Making (MCDM) approaches can be used to quantify trade-offs between economic, social, and environmental criteria i.e. to identify green production options. The aim of this paper is to review the use of MCDM approaches for designing efficient and effective GSCs. We develop a conceptual framework to find relevant publications and to categorise papers with respect to decision problems, indicators, and MCDM approaches. The analysis shows that (1) the use of MCDM approaches for designing GSCs is a rather new but emerging research field, (2) most of the publications focus on production and distribution problems, and there are only a few inventory models with environmental considerations, (3) the majority of papers assume all data to be deterministic, (4) little attention has been given to minimisation of waste, (5) numerous indicators are used to account for eco-efficiency, indicating the lack of standards. This study, therefore, identifies the need for more multi-criteria models for real-life GSCs, especially with inclusion of uncertainty in parameters that are associated with GSCs.  相似文献

Autosomal dominant parkinsonism has been attributed to pathogenic amino acid substitutions in leucine-rich repeat kinase 2 (LRRK2). By sequencing multiplex families consistent with a PARK8 assignment, we identified a novel heterozygous LRRK2 mutation. A referral sample of 248 affected probands from families with autosomal dominant parkinsonism was subsequently assessed; 7 (2.8%) were found to carry a heterozygous LRRK2 6055G-->A transition (G2019S). These seven patients originate from the United States, Norway, Ireland, and Poland. In samples of patients with idiopathic Parkinson disease (PD) from the same populations, further screening identified six more patients with LRRK2 G2019S; no mutations were found in matched control individuals. Subsequently, 42 family members of the 13 probands were examined; 22 have an LRRK2 G2019S substitution, 7 with a diagnosis of PD. Of note, all patients share an ancestral haplotype indicative of a common founder, and, within families, LRRK2 G2019S segregates with disease (multipoint LOD score 2.41). Penetrance is age dependent, increasing from 17% at age 50 years to 85% at age 70 years. In summary, our study demonstrates that LRRK2 G2019S accounts for parkinsonism in several families within Europe and North America. Our work highlights the fact that a proportion of clinically typical, late-onset PD cases have a genetic basis.  相似文献

Human proliferating cell nuclear antigen (hPCNA) containing a single amino acid substitution at position 85, that of lysine for glutamate (E85K), was compared to wild-type (wt) hPCNA for its ability to promote DNA synthesis by purified DNA polymerase delta (pol delta) both on unmodified templates and past chemically defined template base lesions (translesion synthesis; TLS). Significant enhancement (up to 4-5-fold or greater) was seen but depended both on the exact PCNA/pol delta ratio tested and on the specific nature of the template (e.g., unmodified versus lesion-containing; chemical nature of the template base lesion). These results suggest that human PCNA, either mutated to contain lysine (K) at position 85 or bearing similar primary mutations, would promote more secondary mutagenesis in cells and/or tissues where PCNA is normally expressed at low levels relative to pol delta. Over an entire lifetime, such secondary mutagenesis could be biomedically significant.  相似文献

Understanding the relationship between ecological constraints and life-history properties constitutes a central problem in evolutionary ecology. Directionality theory, a model of the evolutionary process based on demographic entropy, a measure of the uncertainty in the age of the mother of a randomly chosen newborn, provides an analytical framework for addressing this problem. The theory predicts that in populations that spend the greater part of their evolutionary history in the stationary growth phase (equilibrium species), entropy will increase. Equilibrium species will be characterized by high iteroparity and strong demographic stability. In populations that spend the greater part of their evolutionary history in the exponential growth phase (opportunistic species), entropy will decrease when population size is large, and will undergo random variation when population size is small. Opportunistic species will be characterized by weak iteroparity and weak demographic stability when population size is large, and random variations in these attributes when population size is small. This paper assesses the validity of these predictions by employing a demographic dataset of 66 species of perennial plants. This empirical analysis is consistent with directionality theory and provides support for its significance as an explanatory and predictive model of life-history evolution.  相似文献

An expression for the entropy of a population was derived in Demetrius (1974) by using a variational principle argument. This entropy measure is precisely the information content of the distribution in the ages of reproducing individuals in a stationary population. This paper introduces another expression for the entropy by considering the variation in the ages at which offspring will be produced by newborn individuals.The relation between these two measures of entropy and their biological significance are discussed.  相似文献

DNA polymerase III, a decameric 420-kDa assembly, simultaneously replicates both strands of the chromosome in Escherichia coli. A subassembly of this holoenzyme, the seven-subunit clamp loader complex, is responsible for loading the sliding clamp (β2) onto DNA. Here, we use structural information derived from ion mobility mass spectrometry (IM-MS) to build three-dimensional models of one form of the full clamp loader complex, γ3δδ′ψχ (254 kDa). By probing the interaction between the clamp loader and a single-stranded DNA (ssDNA) binding protein (SSB4) and by identifying two distinct conformational states, with and without ssDNA, we assemble models of ψχ–SSB4 (108 kDa) and the clamp loader–SSB4 (340 kDa) consistent with IM data. A significant increase in measured collision cross-section (~ 10%) of the clamp loader–SSB4 complex upon DNA binding suggests large conformational rearrangements. This DNA bound conformation represents the active state and, along with the presence of ψχ, stabilises the clamp loader–SSB4 complex. Overall, this study of a large heteromeric complex analysed by IM-MS, coupled with integrative modelling, highlights the potential of such an approach to reveal structural features of previously unknown complexes of high biological importance.  相似文献

Heat shock factor Hsf1 regulates the stress‐inducibility of heat shock proteins (Hsps) or molecular chaperones. One of the functions attributed to Hsps is their participation in folding and degradation of proteins. We recently showed that hsf1?/? cells accumulate ubiquitinated proteins. However, a direct role for Hsf1 in stability of specific proteins such as p53 has not been elucidated. We present evidence that cells deficient in hsf1 accumulate wild‐type p53 protein. We further show that hsf1?/? cells express lower levels of αB‐crystallin and cells deficient in αB‐crystallin also accumulate p53 protein. Reports indicate that αB‐crystallin binds to Fbx4 ubiquitin ligase, and they target cyclin D1 for degradation through a pathway involving the SCF (Skp1‐Cul1‐F‐box) complex. Towards determining a mechanism for p53 degradation involving αB‐crystallin and Hsf1, we have found that ectopic expression of Fbx4 in wild‐type mouse embryo fibroblasts (MEFs) expressing mutant p53 (p53R175H) leads to increase in its degradation, while MEFs deficient in hsf1 or αBcry are defective in degradation of this p53 protein. In addition, immunoprecipitated p53R175H from wild‐type MEFs is able to pull‐down both αB‐crystallin and Fbx4. Finally, immunoprecipitated wild‐type p53 from doxorubicin treated U2OS cells can pull‐down endogenous αB‐crystallin and Fbx4. These results indicate that hsf1‐ and αBcry‐deficient cells accumulate p53 due to reduced levels of αB‐crystallin in these cells. Elevated levels of p53 in hsf1‐ and αBcry‐deficient cells lead to their increased sensitivity to DNA damaging agents. Studies suggest that Hsf4 expression correlates with its role in cell growth and differentiation. However, the role of Hsf4 in tumorigenesis in vivo remains unexplored. In this article, we provide evidence that absence of the Hsf4 gene suppresses evolution of spontaneous tumors arising in p53- or Arf-deficient mice. Furthermore, deletion of hsf4 alters the tumor spectrum by significantly inhibiting development of lymphomas that are normally observed in the majority of mice lacking p53 or Arf tumor suppressor genes. Using mouse embryo fibroblasts deficient in the hsf4 gene, we have found that these cells exhibit reduced proliferation that is associated with induction of senescence and senescence-associated β-galactosidase (SA-β-gal). Cellular senescence in hsf4-deficient cells is associated with the increased expression of the cyclin-dependent kinase inhibitors, p21 and p27 proteins. Consistent with the cellular senescence observed in vitro, specific normal tissues of hsf4(-/-) mice and tumors that arose in mice deficient in both hsf4 and p53 genes exhibit increased SA-β-gal activity and elevated levels of p27 compared with wild-type mice. These results suggest that hsf4 deletion-induced senescence is also present in vivo. Our results therefore indicate that Hsf4 is involved in modulation of cellular senescence, which can be exploited during cancer therapy.  相似文献
