Our aim was to evaluate the association between the expression and the polymorphism of TLR4/NF-κB pathways and colon cancer. TLR4 (rs4986790, rs10759932, rs10759931 and rs2770150) were genotyped in blood samples from Colorectal patients and healthy controls. TLR4 and cytokines inflammatory expression were evaluated by real time PCR on 40 matching normal and colon tissues and the protein level by Immunohistochemistry. The high level of TLR4 expression in colon cancer tissues is mainly due to infections by bacteria in the human colon and leads to induction of an acute secretion of inflammatory cytokines mediated by NF-κB. Also, we report here a clear evidence for an association between TLR4 rs10759931 polymorphism (OR = 0.086, CI: 0.04–0.18, P = <0.00001). This polymorphism affects the entire population without being specific to either gender or to any age group. In contrast, the rs2770150 is associated with colon cancer in women aged over 50 years and is closely linked with the decreased levels of female sex hormones during the post-menopausal period (OR = 0.188, CI: 0.074–0.48, P = <0.00084). rs10759932 and rs4986790 appear to have any association with colon cancer. Our data suggest that TLR4 SNPs could possibly serve as biomarkers for decision making in colon cancer treatment.  相似文献

Seedling recruitment is a bottleneck for population dynamics and range shift. The vital rates linked to recruitment by seed are impacted by amplified drought induced by climate change. In the Mediterranean region, autumn and winter seedling emergence and mortality may have strong impact on the overall seedling recruitment. However, studies focusing on the temporal dynamic of recruitment during these seasons are rare. This study was performed in a deciduous Mediterranean oak forest located in southern France and quantifies the impact of amplified drought conditions on autumn and winter seedling emergence and seedling mortality rates of two herbaceous plant species with meso‐Mediterranean and supra‐Mediterranean distribution (respectively, Silene italica and Silene nutans). Seedlings were followed from October 2019 to May 2020 in both undisturbed and disturbed plots where the litter and the aboveground biomass have been removed to create open microsites. Amplified drought conditions reduced seedling emergence and increased seedling mortality for both Silene species but these negative effects were dependent on soil disturbance conditions. Emergence of S. italica decreased only in undisturbed plots (−7%) whereas emergence of Snutans decreased only in disturbed plots (−10%) under amplified drought conditions. The seedling mortality rate of Sitalica was 51% higher under amplified drought conditions in undisturbed plots while that of Snutans was 38% higher in disturbed plots. Aridification due to lower precipitation in the Mediterranean region will negatively impact the seedling recruitment of these two Silene species. Climate change effects on early vital rates may likely have major negative impacts on the overall population dynamic.  相似文献

Arginine vasopressin (AVP), oxytocin (OT) and neurophysins (Np) have been found in the pineal gland and the retina of the rat. Because the retina, pineal gland and Harderian gland (HG) serve analogous functions, we undertook a study to determine the presence of these peptides in these three organs of rats. They were detected by two specific methods: HPLC and specific radioimmunoassays. For Np, total neurophysins (NpT) were measured. To determine a 24 hr rhythm, the animals were maintained under a light/dark cycle of 12 hr/12 hr for 3 weeks. The pineal glands, retinae and HG were collected. Day/night rhythms of AVP, OT and NpT were demonstrated in the retina and HG; but the pineal gland had only AVP rhythm. A significant decrease in the rhythms at 4 a.m. was demonstrated in the retina and HG. The 24 hr variation of AVP in the retina seemed parallel to that of the HG.  相似文献

Endothelin-1(ET) is a recently discovered endothelium-derived vasoconstrictor peptide. Plasma concentrations of immunoreactive (IR-) endothelin were investigated in rats by radioimmunoassay after extraction of endothelin on Sep-Pak Cartridges. In control rats, a mean concentration of 1.03 +/- 0.2 pg/ml was found. After exercise performed until semi-exhaustion or exhaustion, the mean plasma concentration of IR-endothelin decreased. An increase of Atrial Natriuretic Peptide plasma was observed immediately after exercise. There was a negative correlation between Endothelin and Atrial Natriuretic Peptide (r = -0.73, p less than 0.001). This is the first time that this increase following exercise is shown in normotensive rat. The mechanisms of the decrease of Endothelin are unclear. One hypothesis could be that the decrease in Endothelin is related to its rapid removal by the different tissues, specially the lungs.  相似文献

Node cultures of Eucalyptus camaldulensis Dehn inPetri dishes in vitro under darkness in the presence of anauxin developed meristematic agglomerates (4 to 6 diameter),i.e. dense shoot clusters in which outgrowth of numerous successive buds islimited. Similar cultures under a 16 photoperiod produced smallgreen plantlets with reduced leaves often presenting white hypertrophiedlenticels and very short roots crowning the stem bases. The use of half-litreglass vials under light allowed direct development of well-developed rootedplantlets, either in the presence of the same auxin or in the presence of acytokinin. Light favoured an increase in phenolic compounds and a reversevariation of peroxidase activity during the culture cycles. These aspects arediscussed in terms of a possible regulation of the endogenous auxin levelthrough a light control of peroxidase activity and the level of phenoliccompounds.  相似文献

Cytochrome c(550) (cyt c(550)) is a component of photosystem II (PSII) from cyanobacteria, red algae, and some other eukaryotic algae. Its physiological role remains unclear. In the present work, measurements of the midpoint redox potential (E(m)) were performed using intact PSII core complexes preparations from a histidine-tagged PSII mutant strain of the thermophilic cyanobacterium Thermosynechococcus (T.) elongatus. When redox titrations were done in the absence of redox mediators, an E(m) value of +200 mV was obtained for cyt c(550). This value is ~300 mV more positive than that previously measured in the presence of mediators (E(m) = -80 mV). The shift from the high potential form (E(m) = +200 mV) to the low potential form (E(m) = -80 mV) of cyt c(550) is attributed to conformational changes, triggered by the reduction of a component of PSII that is sequestered and out of equilibrium with the medium, most likely the Mn(4)Ca cluster. This reduction can occur when reduced low potential redox mediators are present or under highly reducing conditions even in the absence of mediators. Based on these observations, it is suggested that the E(m) of +200 mV obtained without mediators could be the physiological redox potential of the cyt c(550) in PSII. This value opens the possibility of a redox function for cyt c(550) in PSII.  相似文献
