The Spindle Assembly Checkpoint (SAC) is part of a complex feedback system designed to ensure that cells do not proceed through mitosis unless all chromosomal kinetochores have attached to spindle microtubules. The formation of the kinetochore complex and the implementation of the SAC are regulated by multiple kinases and phosphatases. BubR1 is a phosphoprotein that is part of the Cdc20 containing mitotic checkpoint complex that inhibits the APC/C so that Cyclin B1 and Securin are not degraded, thus preventing cells going into anaphase. In this study, we found that PP2A in association with its B56γ regulatory subunit, are needed for the stability of BubR1 during nocodazole induced cell cycle arrest. In primary cells that lack B56γ, BubR1 is prematurely degraded and the cells proceed through mitosis. The reduced SAC efficiency results in cells with abnormal chromosomal segregation, a hallmark of transformed cells. Previous studies on PP2A's role in the SAC and kinetochore formation were done using siRNAs to all 5 of the B56 family members. In our study we show that inactivation of only the PP2A-B56γ subunit can affect the efficiency of the SAC. We also provide data that show the intracellular locations of the B56 subunits varies between family members, which is consistent with the hypothesis that they are not completely functionally redundant. 相似文献

Indirect genetic effects (IGEs) describe the effect of the genes of social partners on the phenotype of a focal individual. Here, we measure indirect genetic effects using the “coefficient of interaction” (Ψ) to test whether Ψ evolved between Drosophila melanogaster and D. simulans. We compare Ψ for locomotion between ethanol and nonethanol environments in both species, but only D. melanogaster utilizes ethanol ecologically. We find that while sexual dimorphism for locomotion has been reversed in D. simulans, there has been no evolution of social effects between these two species. What did evolve was the interaction between genotype‐specific Ψ and the environment, as D. melanogaster varies unpredictably between environments and D. simulans does not. In this system, this suggests evolutionary lability of sexual dimorphism but a conservation of social effects, which brings forth interesting questions about the role of the social environment in sexual selection. 相似文献

Vermicomposting of different forms of water hyacinth by the earthworm Eudrilus eugeniae,Kinberg 总被引:1,自引:0,他引:1
Six-month long trials were conducted on different vermireactors fed with one of the following forms of water hyacinth: (a) fresh whole plants, (b) dried whole plants, (c) chopped pieces of fresh plants, (d) 'spent' weed taken from reactors after extracting volatile fatty acids (VFAs), (e) precomposted fresh weed and (f) precomposted spent weed. The first four forms were studied with and without cowdung. The experiments revealed three clear trends (i) of the various forms of the weed assessed, the precomposted forms were the most favoured as feed by Eudrilus eugeniae, Kinberg, while the fresh whole form was the least favoured, (ii) the different forms of spent weed were favoured over the corresponding forms of fresh weed, and (iii) blending of cowdung (approximately 14% of the feed mass) with different forms of water hyacinth had a significant positive impact on vermicast output, growth in worm zoomass, and production of offspring relative to the corresponding unblended feed. In all reactors, the 'parent' earthworms steadily grew in size over the six-month span, and produced offspring. There was no mortality. The experiments thus confirm that water hyacinth can be sustainably vermicomposted in any of the forms with E. eugeniae. 相似文献

Avian hemoglobins have attracted much attention in view of the unique oxygen transport characteristics. The present study describes the primary structure of minor hemoglobin component HbD from Tufted duck (Aythya fuligula), a migratory bird seen in Pakistan during the winter season. Separation of the polypeptide subunits was achieved by ion exchange chromatography in the presence of 8M urea. Molecular masses of the intact protein as well as peptides obtained from chemical and enzymatic cleavages were determined by electrospray ionization mass spectrometry. The sequence was studied by automatic Edman degradation of the native chains and their tryptic/hydrolytic fragments in a gas-phase sequencer. Comparison of the hemoglobin sequence with the corresponding sequences of Anseriform representatives and other avian species shows residues like alpha(D)23 Asp, alpha(D)120 Asp as being specific to Tufted duck. The three-dimensional structure analyzed with the protein structure modeling package, WHAT IF, using the crystal structure coordinates of chicken hemoglobin (PDB code=1hbr) shows alpha(D)34 Val, alpha(D)38 Gln, and alpha(D)94 Asp as possible mediators offering alternate pathway for oxygen uptake and release thereby leading to distinct hypoxia tolerance in the Tufted ducks. Serine proteinase inhibitors are widely distributed in nature and inhibit the activity of enzymes like trypsin and chymotrypsin. These proteins interfere with the physiological processes such as germination, maturation and form the first line of defense against the attack of seed predator. The most thoroughly examined plant serine proteinase inhibitors are found in the species of the families Leguminosae, Graminae, and Solanaceae. Leucaena leucocephala belongs to the family Leguminosae. It is widely used both as an ornamental tree as well as cattle food. We have constructed a three-dimensional model of a serine proteinase inhibitor from L. leucocephala seeds (LTI) complexed with trypsin. The model was built based on its comparative homology with soybean trypsin inhibitor (STI) using the program, MODELLER6. The quality of the model was assessed stereochemically by PROCHECK. LTI shows structural features characteristic of the Kunitz type trypsin inhibitor and shows 39% residue identity with STI. LTI consists of 172 amino acid residues and is characterized by two disulfide bridges. The protein is a dimer with the two chains being linked by a disulfide bridge. Despite the high similarity in the overall tertiary structure, significant differences exist at the active site between STI and LTI. The present study aims at analyzing these interactions based on the available amino acid sequences and structural data. We have also studied some functional sites such as phosphorylation, myristoylation, which can influence the inhibitory activity or complexation with other molecules. Some of the differences observed at the active site and functional sites can explain the unique features of LTI. 相似文献

Paper waste, spiked with varying proportions of cowdung, was vermicomposted in 'low-rate' and 'high-rate' reactors. The former type of reactors had earthworm populations and feed loading rates similar to ones recommended by previous workers. The 'high-rate' reactors were operated with 12.5 times higher earthworm densities and feed loading rates. All the reactors were studied for six months to assess the vermicast output, survivability, growth and reproduction of the earthworms hence the sustainability of the reactors--for long-term, continuous operation. The studies revealed the viability of the high-rate vermireactor concept. The high-rate reactors consistently produced over 6.5 times more castings per unit digester volume with no adverse effect on the earthworm population, as reflected by (a) absence of mortality, (b) consistent growth in worm zoomass, and (c) normal rate of reproduction. The studies also revealed that an increase in the cowdung fraction in the feed from 14.3% to 20% (4:1 paper:cowdung blends to 6:1 blends) had little positive impact on the vermicast output or earthworm health. This indicated that spiking of paper feed with approximately 14% cowdung, or perhaps an even smaller fraction, might be adequate to support earthworms in the paper-fed vermireactors. 相似文献

Synchrotron x-ray scattering measurements were performed on dilute solutions of the purified hemocyanin subunit (Bsin1) from scorpion (Buthus sindicus) and the N-terminal functional unit (Rta) from a marine snail (Rapana thomasiana). The model-independent approach based on spherical harmonics was applied to calculate the molecular envelopes directly from the scattering profiles. Their molecular shapes in solution could be restored at 2-nm resolution. We show that these units represent stable, globular building blocks of the two hemocyanin families and emphasize their conformational differences on a subunit level. Because no crystallographic or electron microscopy data are available for isolated functional units, this study provides for the first time structural information for isolated, monomeric functional subunits from both hemocyanin families. This has been made possible through the use of low protein concentrations (< or = 1 mg/ml). The observed structural differences may offer advantages in building very different overall molecular architectures of hemocyanin by the two phyla. 相似文献

This work presents a biophysical study on the interactions of a monorhamnolipid (monoRL) produced by Pseudomonas aeruginosa MA01 with model phosphatidylcholine membranes. The molecular characterization of the biological activities, including the modulation of phospholipid membranes structure, of this monoRL biosurfactant is of importance for the validation of this particular Pseudomonas aeruginosa strain as a useful biosurfactant producer. The marked amphiphilic structure of monoRL is expected to result in strong interactions with the phospholipid constituents of membrane bilayers. Incorporation of monoRL into DMPC completely abolished the pretransition, and the main gel to liquid-crystalline phase transition was progressively broadened and shifted to lower temperatures, as observed by differential scanning calorimetry. Partial phase diagrams for DPPC and DSPC indicated near-ideal behavior. However, the DMPC diagram indicated fluid phase immiscibility. X-ray diffraction showed and apparent increase in d-value for DPPC containing monoRL, which might be the result of an effective increase in the bilayer thickness, or in the thickness of the hydration layer between bilayers. FTIR indicated that interaction of monoRL with the phospholipid acyl chains did not result in a large additional disordering of the acyl chain region of the fluid bilayer. Analysis of the CO stretching band of DPPC indicated an important effect of monoRL on the interfacial region of phosphatidylcholine bilayers, which might contribute to explain some of the biological activities of this glycolipid. 相似文献
