SUMMARY. Comparison is made of resistance to anoxia at 0°C of an English and a Swedish winter larval population of two closely related forms of Cloeon (dipterum?). The English population was not very resistant but contained a few slightly resistant larvae; the Swedish population was very resistant. Swedish larvae initiated hibernation more rapidly than English larvae and were also more resistant to starvation. The different properties of the populations are seen as an adaptation to different winter conditions in ponds in England and Sweden. Evidence is presented that low temperatures but not decreasing photoperiod are decisive for the final induction of hibernation in the larvae. In Swedish larvae this induction starts at between 4 and 5°C.  相似文献

Direct evidence of pseudogamy in apomictic Brachiaria brizantha (Poaceae)   总被引:1,自引:0,他引:1  
Brachiaria brizantha is a forage grass that has several apomictic accessions. B. brizantha cv. Marandu is a natural tetraploid aposporous apomict widely cultivated in Brazil. Pseudogamy was detected in this species by observation that seed set is suppressed in plants that have had the stigmas excised from the flowers. The egg cell develops parthenogenetically in the apomictic plants, meaning that fertilisation is necessary for the formation of the endosperm. A thorough knowledge of all the events of seed formation in natural apomictic plants is essential for a complete understanding of this mode of reproduction. In this paper, we show direct evidence of pseudogamy in B. brizantha through the cytological analysis of polar nucleus fertilisation and the determination of triploid level of the endosperm tissue. The development of the male gametophyte gives rise to a reduced tri-celled pollen, the viability of which varies throughout the year, reaching 88% in the peak of the flowering period. Discharge of the male gamete takes place around 10 h after pollination and monospermy is the predominant system observed. Precocious embryony was also observed in these plants; embryos arise from egg cells. Endosperm development followed the free nucleus model and was associated with the presence of an embryo. Cellularisation and reserve uptake occurred 2 days after pollination (DAP) and mature endosperm was observed 8 DAP. The triploid level of the endosperm in the apomictic accession confirmed the 2:1 maternal:paternal ratio of genome contribution in the tissue.  相似文献

The addition of bovine serum albumin (BSA) to 25 ± 5 nm diameter single bilayer phosphatidylcholine (PC) vesicles (SBV) (pH 3.5) gives rise to readily visible transient turbidity. Studies of this system, employing a series of techniques, including time-dependent turbidity changes, membrane filtration, centrifugation, Sepharose chromatography and freeze fracture electron microscopy demonstrated that the process involves aggregation and fusion of the vesicles. At least three distinct time-dependent steps have been characterized: (1) the rapid initial formation (in approx. 5 min) of large aggregates (responsible for the visible turbidity) composed of SBV interconnected by BSA in its F form. The formation of these aggregates may be reversed by raising the pH or adding excess BSA to the system at this stage; (2) spontaneous collapse of these large aggregates, in an irreversible step, to form a heterogeneous population of vesicles; (3) fusion produces as the final product of the process, a relatively homogeneous population of larger (50 ± 10 nm diameter) vesicles. This system serves as a convenient and simple model system for the detailed study of protein-mediated aggregation and fusion of membranes at the molecular level.  相似文献

Survey and laboratory studies suggest that several factors, such as social and academic demands, part-time jobs and irregular school schedules, affect the sleep-wake cycle of college students. In this study, we examined the sleep-wake pattern and the role played by academic schedules and individual characteristics on the sleep-wake cycle and academic performance. The subjects were 36 medical students (male = 21 and female = 15), mean age = 20.7 years, SD = 2.2. All students attended the same school schedule, from Monday to Friday. The volunteers answered a morningness-eveningness questionnaire, the Pittsburgh Sleep Quality Index (PSQI) and kept a sleep-wake diary for two weeks. The relationships between sleep-wake cycle, PSQI, chronotypes and academic performance were analyzed by a multiple regression technique. The results showed that 38.9% of the students had a poor sleep quality according to the PSQI. When the medical students were evening type or moderate evening type the PSQI showed a tendency of poor sleep. The multiple regression analysis showed a correlation between sleep onset, sleep irregularity and sleep length with academic performance. These results suggest that chronotypes influence the quality of the sleep-wake cycle and that irregularity of the sleep-wake cycle, as well as sleep deprivation (average length was 6:52), influence the learning of college students.  相似文献

 Batch experiments were conducted to assess the biotransformation potential of four hydrocarbon monoterpenes (d-limonene, α-pinene, γ-terpinene, and terpinolene) and four alcohols (arbanol, linalool, plinol, and α-terpineol) under aerobic conditions at 23°C. Both forest-soil extract and enriched cultures were used as inocula for the biodegradation experiments conducted first without, then with prior microbial acclimation to the monoterpenes tested. All four hydrocarbons and two alcohols were readily degraded. The increase in biomass and headspace CO2 concentrations paralleled the depletion of monoterpenes, thus confirming that terpene disappearance was the result of biodegradation accompanied by microbial growth and mineralization. Plinol resisted degradation in assays using inocula from diverse sources, while arbanol degraded very slowly. A significant fraction of d-limonene-derived carbon was accounted for as non-extractable, dissolved organic carbon, whereas terpineol exhibited a much higher degree of utilization. The rate and extent of monoterpene biodegradation were not significantly affected by the presence of dissolved natural organic matter. Glucose metabolism by sloth fat cells with and without addition of insulin was investigated. The data were compared to the results obtained with rat fat cells incubated under the same experimental conditions. Sloth fat cells showed a very low glucose oxidation to 14CO2 and incorporation into total lipids. The glucose incorporated into lipids is mainly in the glyceride-glycerol moiety. Addition of insulin did not produce an increase on glucose oxidation and a slight increase in the incorporation into total lipids was observed. Since it has been reported that sloths have a very low rate on thyroxine secretion, the results are discussed in relation to data in the literature on carbohydrate and lipid metabolism in hypothyroid animals.  相似文献

Thylakoids isolated from pine chloroplasts were solubilized by sodium dodecyl sulphate and the polypeptides were separated by polyacrylamide gel electrophoresis. The chlorophyll-protein complexes, P700-CPa1 and LH-CPa/b, had apparent molecular weights of 92,000 and 25,000, respectively. When the chlorophyll of P700-CPa1 was extracted or photobleached, the apoprotein of P700-CPa1 appeared as a pronounced peak in the polypeptide scan profile. The molecular weight of the apoprotein was 70,000. During autumn and winter the complex P700-CPa1 was destroyed. This was primarily caused by bleaching of chlorophyll, as the 70,000 apoprotein increased in the scan profile when the complex P700-CPa1 decreased. The winter destruction of P700-CPa1 was less pronounced in old needles than in young. Freezing of frost-hardened seedlings did not change the polypeptide scan profile, unless the temperature was lowered below the frost-killing point followed by thawing and post-treatment in light or darkness above 0°C. Again the main destruction occurred in the P700-CPa1 complex, but in this case no significant increase of the apoprotein was observed. These alterations in the polypeptide scan profile of frost-killed needles were not caused by the low temperature treatment as such, but they occurred after thawing of the needles.  相似文献

Centrins are calcium-binding proteins associated with microtubules organizing centers. Members of two divergent subfamilies of centrins were found in the aquatic fungus Blastocladiella emersonii, contrasting with the occurrence of only one member known for the better explored terrestrial fungi. BeCen1 shows greatest identity with human centrins HsCen1, HsCen2 and green algae centrin CrCenp, while BeCen3 records largest identity with human centrin HsCen3 and yeast centrin Cdc31p. Following the discovery of this unique feature, BeCen1 and BeCen3 centrins were produced to study whether these proteins had distinct features upon calcium binding. Circular dichroism showed opposite calcium binding effects on the α-helix arrangement of the secondary structure. The spectra indicated a decrease in α-helix signal for holo-BeCen1 contrasting with an increase for holo-BeCen3. In addition, only BeCen1 refolds after being de-natured. The fluorescence emission of the hydrophobic probe ANS increases for both proteins likely due to hydrophobic exposure, however, only BeCen1 presents a clear blue shift when calcium is added. ITC experiments identified four calcium binding sites for both proteins. In contrast to calcium binding to BeCen1, which is mainly endothermic, binding to BeCen3 is mainly exothermic. Light-scattering evidenced the formation of large particles in solution for BeCen1 and BeCen3 at temperatures above 30 °C and 40 °C, respectively. Atomic force microscopy confirmed the presence of supramolecular structures, which differ in the compactness and branching degree. Binding of calcium leads to different structural changes in BeCen1 and BeCen3 and the thermodynamic characteristics of the interaction also differ.  相似文献
