Scavenging and anti-Staphylococcus aureus activities of extracts, fractions and subfractions from the in vitro mycelium of two strains of the edible mushroom Volvariella volvacea were determined. Chloroform subfractions of the ATCC62890 strain showed the highest inhibition percentage of the DPPH radical (over 80%) after 180 min. When chloroform and hexane subfractions of the R83 strain were combined they showed moderate (inhibition zone of 8.99 ± 0.78 mm) to high (inhibition zone of 13.06 ± 0.41 mm) activity against the Gram-positive bacteria Staphylococcus aureus, which are 74.4 and 51.2% of tetracycline (inhibition zone of 17.55 ± 0.11 mm). Partitioning suggested that the substances in the chloroform and hexane fractions of the R83 strain act synergistically to give the antimicrobial activity, while separating the substances of the ATCC62890 subfractions reduced their activity. In general, the R83 strain seems to be a source of antimicrobial substances, while the ATCC62890 strain appears to be an alternative source of antioxidants.  相似文献

For many insect species, pheromones are important communication tools, but chemical analysis and experimental study can be technically challenging because they require the detection and handling of complex chemicals in small quantities. One drawback of traditional mass spectrometry methods such as gas chromatography mass spectrometry is that whole‐body extractions from one to several hundred individuals are required, with the consequence that intra‐ and interindividual differences cannot be detected. Here, we used the recently introduced UV‐LDI MS (ultraviolet laser desorption/ionization mass spectrometry) to profile the ‘osmeterium’ of the sepsid fly Themira superba that is located on the edge of the hind tibia of males. Based on analyses of individual legs, we established that the gland produced a secretion that consisted of oxygenated hydrocarbons and putative isoprenoids. The secretion was first detected 24 h after eclosion, and its transfer to the wings of females during mating was demonstrated using UV‐LDI MS. We then tested whether the secretion had an anti‐aphrodisiac function, but experimental transfer of the secretion to virgin females did not affect mating success or copulation duration. Throughout the study, UV‐LDI MS proved invaluable, because it allowed tracking the natural and experimental transfer of small quantities of pheromones to specific body parts of small flies.  相似文献

The Chapeu-de-couro (Echinodorus macrophyllus (Kunth) Micheli) is a native plant from Brazil, which has been mainly used in medicinal application being a potent antirheumatic and diuretic, in the production of soft drinks, and also in the ornamentation of aquariums. In this paper, the metals and metalloids for the leaves of chapeu-de-couro collected in the Paraguacu River from the city Cachoeira, Bahia State, Brazil, was determined and evaluated using multivariate analysis. The samples were digested using nitric acid and hydrogen peroxide and were analyzed using inductively coupled plasma mass spectrometry. The accuracy of the method was confirmed by analysis of a certified reference material of apple leaves, furnished by National Institute of Standard and Technology. The study involved 15 samples of the Paraguacu River. The results expressed as milligrams of element per kilogram of sample demonstrated that the concentration ranges varied: 1.39–5.27 for chromium, 44.85–165.39 for manganese, 0.55–0.84 for arsenic, 0.01–3.94 for antimony, and 0.18–0.31 for lead. The principal component analysis and hierarchical cluster analysis evidenced that the concentrations of the metals and metalloids varied according with the variations in the water of the Paraguacu.  相似文献

The global decline of freshwater mussels can be partially attributed to their complex life cycle. Their survival from glochidium to adulthood is like a long obstacle race, with juvenile mortality as a key critical point. Mass mortality shortly after entering into a juvenile state has been reported in both wild and captive populations, thus weakening the effective bivalve population. A similar phenomenon occurs during metamorphosis in natural and hatchery populations of juvenile marine bivalves. Based on a morphological analysis using scanning electron microscopy of newly formed juveniles of the freshwater species Margaritifera margaritifera (L.) (Margaritiferidae) and Unio mancus Lamarck (Unionidae), we show that a second metamorphosis, consisting of drastic morphological changes, occurs that leads to suspension feeding in place of deposit feeding by the ciliated foot. We hypothesize that suspension feeding in these two species improves due to a gradual development of several morphological features including the contact between cilia of the inner gill posterior filaments, the inner gill reflection, the appearance of the ctenidial ventral groove and the formation of the pedal palps. Regardless of the presence of available food, a suspension feeding mode replaces deposit feeding, and juveniles unable to successfully transition morphologically or adapt to the feeding changes likely perish.  相似文献

Experiments performed to determine the influence of the C5 component of complement in experimental Toxoplasma infection revealed that mice deficient in C5 had reduced mortality due to acute toxoplasmosis. Similar results were noted when inbred congenic mice of known complement type, as well as random-bred mice selected for complement type, were used. In both, mice with high complement activity were less resistant to Toxoplasma than were mice deficient in C5. However, many factors must interact in susceptibility to infection with T. gondii. Thus, lower resistance to Toxoplasma was noted in C5-deficient DBA/2J mice, whereas a high degree of resistance was noted in DBA/1J mice, which are not related to DBA/2J mice and which possess a normal sequence of complement. This accentuates the importance of using both random-bred and where possible cogenic lines in assessing the importance of individual factors in infectious immunity.  相似文献

We report a simple and fast microwave-assisted method to grow silver nanoparticle films with tunable plasmon resonance band. Microwaving time controls nucleation and growth as well as particle agglomeration, cluster formation, particle morphology, and the plasmonic properties. Films produced with times shorter than 30 s presented a single well-defined plasmon resonance band (~ 400 nm), whereas films produced with times longer than 40 s presented higher wavelength resonances modes (> 500 nm). Plasmon band position and intensity can be easily tuned by controlling microwaving time and power. SEM and AFM images suggested the growth of asymmetrical silver nanoparticles. Simulated extinction spectra considering particles as spheres, hemispheres, and spherical caps were performed. The films were employed to enhance the sensitivity of ionizing radiation detectors assessed by optically stimulated luminescence (OSL) via plasmon-enhanced luminescence. By tuning the plasmon resonance band to overlap with the OSL stimulation (530 nm), luminescence enhancements of greater than 100-fold were obtained, demonstrating the importance of tuning the plasmon resonance band to maximize the OSL intensity and detector sensitivity. This versatile method to produce silver nanoparticle films with tunable plasmonic properties is a promising platform for developing small-sized radiation detectors and advanced sensing technologies.
