The Antarctic soil microbial community has a crucial role in the growth and stabilization of higher organisms, such as vascular plants. Analysis of the soil microbiota composition in that extreme environmental condition is crucial to understand the ecological importance and biotechnological potential. We evaluated the efficiency of isolation and abundance of strict anaerobes in the vascular plant Deschampsia antarctica rhizosphere collected in the Antarctic’s Admiralty Bay and associated biodiversity to metabolic perspective and enzymatic activity. Using anaerobic cultivation methods, we identified and isolated a range of microbial taxa whose abundance was associated with Plant Growth-Promoting Bacteria (PGPB) and presences were exclusively endemic to the Antarctic continent. Firmicutes was the most abundant phylum (73 %), with the genus Clostridium found as the most isolated taxa. Here, we describe two soil treatments (oxygen gradient and heat shock) and 27 physicochemical culture conditions were able to increase the diversity of anaerobic bacteria isolates. Heat shock treatment allowed to isolate a high percentage of new species (63.63 %), as well as isolation of species with high enzymatic activity (80.77 %), which would have potential industry application. Our findings contribute to the understanding of the role of anaerobic microbes regarding ecology, evolutionary, and biotechnological features essential to the Antarctic ecosystem.  相似文献

  • Gall inducers use these structures as shelters and sources of nutrition. Consequently, they cause multiple physiological changes in host plants.
  • We studied the impact caused by seed coat galls of a braconid wasp on the performance of fruits, seeds and seedlings of tree Inga laurina. We tested whether these seed galls are ‘nutrient sinks’ with respect to the fruit/seed of host plant, and so constrain the reproductive ability and reduce seedling longevity. We measured the influence of such galls on the secondary compounds, fruit and seed parameters, seed viability and germination and seedling performance.
  • Inga laurina has indehiscent legumes with polyembryonic seeds surrounded by a fleshy sarcotesta rich in sugars. The galls formed inside the seed coat and galled tissues presented higher phenol concentrations, around 7‐fold that of ungalled tissues. Galls caused a significant reduction in parameters such as fruit and seed size, seed weight and the number of embryos. Fluctuating asymmetry (a stress indicator) was 31% higher in leaves of galled seed plants in comparison to ungalled seed plants. However, the negative effects on fruit and seed parameters were not sufficient to reduce seed germination (except the synchronization index) or seedling performance (except leaf area and chlorophyll content).
  • We attributed these results to the ability of I. laurina to tolerate gall attack on seeds without a marked influence on seedling performance. Moreover, because of the intensity of seed galling on host plant, we suggest that polyembryony may play a role in I. laurina reproduction increasing tolerance to seed damage.

The objective of this work was to develop and evaluate agrometeorological models to simulate the production of Guineagrass. For this purpose, we used forage yield from 54 growing periods between December 2004–January 2007 and April 2010–March 2012 in irrigated and non-irrigated pastures in São Carlos, São Paulo state, Brazil (latitude 21°57′42″ S, longitude 47°50′28″ W and altitude 860 m). Initially we performed linear regressions between the agrometeorological variables and the average dry matter accumulation rate for irrigated conditions. Then we determined the effect of soil water availability on the relative forage yield considering irrigated and non-irrigated pastures, by means of segmented linear regression among water balance and relative production variables (dry matter accumulation rates with and without irrigation). The models generated were evaluated with independent data related to 21 growing periods without irrigation in the same location, from eight growing periods in 2000 and 13 growing periods between December 2004–January 2007 and April 2010–March 2012. The results obtained show the satisfactory predictive capacity of the agrometeorological models under irrigated conditions based on univariate regression (mean temperature, minimum temperature and potential evapotranspiration or degreedays) or multivariate regression. The response of irrigation on production was well correlated with the climatological water balance variables (ratio between actual and potential evapotranspiration or between actual and maximum soil water storage). The models that performed best for estimating Guineagrass yield without irrigation were based on minimum temperature corrected by relative soil water storage, determined by the ratio between the actual soil water storage and the soil water holding capacity.irrigation in the same location, in 2000, 2010 and 2011. Whenever food is placed in the mouth, taste receptors are stimulated. Simultaneously, different types of sensory fibre that monitor several food attributes such as texture, temperature and odour are activated. Here, we evaluate taste and oral somatosensory peripheral transduction mechanisms as well as the multi-sensory integrative functions of the central pathways that support the complex sensations that we usually associate with gustation. On the basis of recent experimental data, we argue that these brain circuits make use of distributed ensemble codes that represent the sensory and post-ingestive properties of tastants.  相似文献

Classical models of phytoplankton–zooplankton interaction show that with nutrient enrichment such systems may abruptly shift from limit cycles to stable phytoplankton domination due to zooplankton predation by planktivorous fish. Such models assume that planktivorous fish eat only zooplankton, but there are various species of filter-feeding fish that may also feed on phytoplankton. Here, we extend these classical models to systematically explore the effects of omnivory by planktivorous fish. Our analysis indicates that if fish forage on phytoplankton in addition to zooplankton, the alternative attractors predicted by the classical models disappear for all realistic parameter settings, even if omnivorous fish have a strong preference for zooplankton. Our model also shows that the level of fish biomass above which zooplankton collapse should be higher when fish are omnivorous than when fish are zooplanktivorous. We also used the model to explore the potential effects of the now increasingly common practice of stocking lakes with filter-feeding fish to control cyanobacteria. Because omnivorous filter-feeding fish forage on phytoplankton as well as on the main grazers of phytoplankton, the net effect of such fish on the phytoplankton biomass is not obvious. Our model suggests that there may be a unimodal relationship between the biomass of omnivorous filter-feeding fish and the biomass of phytoplankton. This implies that to manage for reductions in phytoplankton biomass, heavy stocking or strong reduction of such fish is best.  相似文献

Taurine is a conditionally essential amino acid for human that is involved in the control of glucose homeostasis; however, the mechanisms by which the amino acid affects blood glucose levels are unknown. Using an animal model, we have studied these mechanisms. Mice were supplemented with taurine for 30 d. Blood glucose homeostasis was assessed by intraperitoneal glucose tolerance tests (IPGTT). Islet cell function was determined by insulin secretion, cytosolic Ca2+ measurements and glucose metabolism from isolated islets. Islet cell gene expression and translocation was examined via immunohistochemistry and quantitative real-time polymerase chain reaction. Insulin signaling was studied by Western blot. Islets from taurine-supplemented mice had: (i) significantly higher insulin content, (ii) increased insulin secretion at stimulatory glucose concentrations, (iii) significantly displaced the dose-response curve for glucose-induced insulin release to the left, (iv) increased glucose metabolism at 5.6 and 11.1-mmol/L concentrations; (v) slowed cytosolic Ca2+ concentration ([Ca2+]i) oscillations in response to stimulatory glucose concentrations; (vi) increased insulin, sulfonylurea receptor-1, glucokinase, Glut-2, proconvertase and pancreas duodenum homeobox-1 (PDX-1) gene expression and (vii) increased PDX-1 expression in the nucleus. Moreover, taurine supplementation significantly increased both basal and insulin stimulated tyrosine phosphorylation of the insulin receptor in skeletal muscle and liver tissues. Finally, taurine supplemented mice showed an improved IPGTT. These results indicate that taurine controls glucose homeostasis by regulating the expression of genes required for glucose-stimulated insulin secretion. In addition, taurine enhances peripheral insulin sensitivity.  相似文献

Regulation of NFκB activity is central to many processes during development and disease. Activation of NFκB family members depends on degradation of inhibitory IκB proteins. In Drosophila, a nuclear gradient of the NFκB/c-rel protein Dorsal subdivides the embryonic dorsal–ventral axis, defining the extent and location of mesodermal and ectodermal territories. Activation of the Toll pathway directs Dorsal nuclear translocation by inducing proteosomal degradation of the IκB homologue Cactus. Another mechanism that impacts on Dorsal activation involves the Toll-independent pathway, which regulates constitutive Cactus degradation. We have shown that the BMP protein Decapentaplegic (Dpp) inhibits Cactus degradation independent of Toll. Here we report on a novel element of this pathway: the calcium-dependent protease Calpain A. CalpainA knockdowns increase Cactus levels, shifting the Dorsal gradient and dorsal–ventral patterning. As shown for mammalian IκB, this effect requires PEST sequences in the Cactus C-terminus, implying a conserved role for calpains. Alteration of CalpainA or dpp results in similar effects on Dorsal target genes. Epistatic analysis confirms CalpainA activity is regulated by Dpp, indicating that Dpp signals increase Cactus levels through Calpain A inhibition, thereby interfering with Dorsal activation. This mechanism may allow coordination of Toll, BMP and Ca2+ signals, conferring precision to Dorsal-target expression domains.  相似文献

In this study, the cell-associated and extracellular peptidases of Trypanosoma cruzi grown in modified Roitman’s complex (MRC) medium were analyzed by measuring peptidase activity in gelatin-containing zymograms. Our results showed that the cell-associated peptidases as well as peptidases extracellularly released by T. cruzi displayed two distinct proteolytic classes: cysteine and metallopeptidase activities. The major cysteine peptidase, cruzipain, synthesized by T. cruzi cells was detected in cellular parasite content, as a 50 kDa reactive polypeptide, after probing with anti-cruzipain antibody. In addition, metallo-type peptidases belonging to the matrix metallopeptidase-9 (MMP-9) family were revealed, after Western blotting, as a 97 kDa protein band in cellular extract and an 85 kDa polypeptide in both cellular and secreted parasite extracts. The MMP-9-like activity present in cells and spent culture medium was immunoprecipitated by an anti-MMP-9 polyclonal antibody. The surface location of MMP-9-like proteins in T. cruzi was also evidenced by means of flow cytometry analysis. Furthermore, doxycycline that has direct MMP-9 inhibiting properties in vitro, inhibited MMP-9-like activities in gel zymography, immunoprecipitation and flow cytometry analyses. This is the first report of the presence of MMP-9-like molecules in T. cruzi. The presence of a matrix extracellular-degrading enzyme may play a role in the T. cruzi-host cell interaction, making this enzyme a potential target for future drug development against this pathogenic trypanosomatid.  相似文献

Sticholysin I (StnI) is an actinoporin produced by the sea anemone Stichodactyla helianthus that binds biological and model membranes forming oligomeric pores. Both a surface cluster of aromatic rings and the N‐terminal region are involved in pore formation. To characterize the membrane binding by StnI, we have studied by 1H‐NMR the environment of these regions in water and in the presence of membrane‐mimicking micelles. Unlike other peptides from homologous actinoporins, the synthetic peptide corresponding to residues 1–30 tends to form helix in water and is more helical in either trifluoroethanol or dodecylphosphocholine (DPC) micelles. In these environments, it forms a helix‐turn‐helix motif with the last α‐helical segment matching the native helix‐α1 (residues 14–24) present in the complete protein. The first helix (residues 4–9) is less populated and is not present in the water‐soluble protein structure. The characterization of wild‐type StnI structure in micelles shows that the helix‐α1 is maintained in its native structure and that this micellar environment does not provoke its detachment from the protein core. Finally, the study of the aromatic resonances has shown that the motional flexibility of specific rings is perturbed in the presence of micelles. On these bases, the implication of the aromatic rings of Trp‐111, Tyr‐112, Trp‐115, Tyr‐132, Tyr‐136, and Tyr‐137, in the interaction between StnI and the micelle is discussed. Based on all the findings, a revised model for StnI interaction with membranes is proposed, which accounts for differences in its behavior as compared with other highly homologous sticholysins.
