Some strains of Staphylococcus aureus grew as compact colonies in Brain Heart Infusion-serum-soft agar but as diffuse colonies in a modified Staphylococcus 110-serum-soft agar. These strains were designated "pseudocompact." Strains showing compact-type colonial morphology in both media were designated "compact," whereas strains showing diffuse-type growth in both media were designated "diffuse." It was observed that the most recently isolated strains of S. aureus were of the pseudocompact type, whereas most stock culture strains tested were of the compact type. Using cultures recently isolated from clinical material, it was shown that pseudocompact strains convert to compact-type growth after prolonged incubation. Interconversion of compact, diffuse, and pseudocompact growth forms could be induced in vitro by appropriate cultural conditions, and conversion of growth type was also observed in vivo. Femoral abscesses produced in mice by four different compact-type strains showed conversion to diffuse or pseudocompact-type growth during the course of the infection.  相似文献

Summary The structure of a 1.5-kb DNA sequence that is necessary and sufficient for the replication of an 8.2-kb cryptic plasmid, pFTB14, isolated from a strain of Bacillus amyloliquefaciens has been characterized. The 1.5-kb DNA sequence contains an open reading frame, rep, stretching for 1017 bp, a promoter region for rep expression, and a possible replication origin for the plasmid upstream of the promoter. The rep product is trans-active and essential for plasmid replication. The predicted rep protein is a basic protein, as are the RepC protein of pT181, RepB of pUB110 and protein A of pC194 (all these found in staphylococci) and the protein of the R6K plasmid of Escherichia coli. The predicted rep protein has highly homologous amino acid sequences with protein A of pC194 and RepC of pUB110 throughout the protein molecule, but not with RepC of pT181, of R6K or protein RepH encoded by and iniating the replication of pC194.  相似文献

The positional distribution of fatty acids in glycerolipidsfrom thalli of Porphyra yezoensis was studied by enzymatic hydrolysis.In monogalactosyl diacylglycerol, icosapentaenoic acid was amajor fatty acid at both the sn-1 and sn-2 positions of theglycerol moiety, whereas palmitic acid was a minor componentat both positions. In digalactosyl diacylglycerol and sulfoquinovosyldiacylglycerol, icosapentaenoic and palmitic acids were almostexclusively distributed at the sn-1 and sn-2 positions, respectively.In phosphatidylglycerol, palmitic and trans--13-hexadecenoicacid were exclusively located at the sn-2 position. In phosphatidylcholine,icosapentaenoic acid occurred in both the sn-l and sn-2 positions,whereas palmitic acid was confined to the sn-1 position. Itis suggested that monogalactosyl diacylglycerol in P. yezoensissynthesized in both the cytoplasmic and chloroplastic pathways,while the diacylglycerol moieties of the other chloroplast lipidsare virtually all derived from the chloroplastic pathway. In order to prove the acute stimulatory effects of estrogen on pituitary gonadotropin release, we have performed the present experiments in 8 women with a hypergonadotropic state due to surgical castration or primary ovarian failure. They received gonadotropin releasing hormone (Gn-RH) for 12-21 h at the constant rate of 20 micrograms/h. In 5 of the women, estradiol-17 beta was concomitantly administered at the rate of 20 micrograms/h from 6 h after the start of Gn-RH infusion. Blood samples were collected frequently throughout the experiments for the analysis of LH, FSH and estradiol. In response to the sole stimulation of Gn-RH, remarkable and prompt rises in LH (313.5%), but to a lesser degree in FSH (194.2%), were observed within the initial 3 h, and their high levels were maintained throughout the experimental period. However, the additional administration of estradiol brought on a further sudden rise in both gonadotropins levels: 178.3% for LH and 163.5% for FSH within 2 h. These high levels were sustained during estradiol infusions. In 2 of them, blood samples were obtained for several hours after cessation of estradiol infusion. The circulating gonadotropin level dropped precipitously close to the baseline level within 3 h after estradiol infusions. Our data indicate that estrogen has an acute and strong augmentative effect on Gn-RH induced gonadotropin release in addition to its conventional negative and positive feedback effects.  相似文献

Structural studies were carried out on two kinds of teichuronic acid-glycopeptide complexes (designated as TU-GP-I and TU-GP-II) isolated from lysozyme digest of N-acetylated cell walls of Bacillus megaterium AHU 1375 by ion-exchange chromatography and gel chromatography. TU-GP-I, accounting for about 25% of the cell walls, contained N-acetylmannosaminuronic acid, N-acetylglucosamine, glucose, galactose, glycerol, and phosphorus in an approximate molar ratio of 1:1:2:1:0.5:0.5, together with small amounts of glycopeptide components. TU-GP-II, accounting for about 9% of the cell walls, contained glucuronic acid, glucose, and fucose in a molar ratio of about 2:1.5:1, together with small amounts of glycopeptide components. The results of analyses involving Smith degradation, chromium oxidation, methylation, acetolysis, and H-NMR measurement led to the conclusion that the polysaccharide chain of TU-GP-I comprised repeating units,----6) Glc(alpha 1----3)-ManNAcUA(beta 1----4)[Gal(alpha 1----3)][Glc(beta 1----6)]GlcNAc(beta 1----. About half of the repeating units were substituted by glycerophosphoryl residues at C-6 of the beta-glucosyl residues linked to the N-acetylglucosamine residues. By means of a similar procedure, the polysaccharide chain of TU-GP-II was shown to comprise repeating units,----4)GlcUA(alpha 1----3)GlcUA(alpha 1----3)Glc(alpha 1----3)Fuc(alpha 1----, of which about half were substituted by alpha-glucosyl residues at C-3 of the 4-substituted glucuronosyl residues.  相似文献

The distributions of carbon, nitrogen and phosphorus in a moss community-soil system developed on a naked region in Rundvågskollane (69050'S, 39009'E), East Antarctica, were investigated in order to analyze the flow of matter in an Antarctic terrestrial ecosystem.
  1. The moss community was formed from many moss blocks of different sizes and was composed ofBryum pseudotriquetrum (Hedw.) Gaertn., Meyer et Scherb.,Ceratodon purpureus (Hedw.) Brid. andGrimmia lawiana J. H. Willis. The surface of the community was covered with cyanobacteria.
  2. It was estimated that nitrogen fixed by cyanobacteria flowed from these organisms to the moss and that little nitrogen was transported within the moss body.
  3. A large amount of phosphorus existed in the soil. The moss community had a high phosphorus content although the amount itself was relatively little due to the small phytomass.
  4. It seems that absorption of phosphorus from soil is difficult for moss, because of the paucity of water necessary for the movement of phosphorus and the suppressed growth of moss due to the arid conditions.

The ultrastructural changes in plastids ofBuxus sempervirens L. leaves were observed during their seasonal yellowing and regreening. The disintegration of chloroplasts into globular type chromoplasts in yellowing leaves and their direct restoration to functional chloroplasts again in regreening leaves were followed. The results presented an example of recent information indicating the essential sense of the reversible reciprocation of plastid transformation.  相似文献

Novel devices for the spectroscopic and chromatographic analysis of the denaturation curves of the protein are described. A multidimensional spectroscopic measuring system makes it possible to carry out simultaneous and continuous acquisition of a set of data of different spectroscopic dimensions at several wavelengths in the course of increasing or decreasing denaturational perturbation. GuHCl-gradient chromatography can provide information about the progressive change of the protein volume in the course of increasing GuHCl concentration. Thus, denaturation curves with a high data-point density can be obtained. The data-storing function by a magnetic disk memory provides enough precision for a rigorous investigation of the correlation among the curves that probe different aspects of denaturation. The GuHCl denaturation of RNase A, cytochrome c, and pepsinogen are studied to demonstrate the high performance of these devices. Three types of transitions are found in these three proteins and the multiphasic nature of the transitions is clearly detected in the last two proteins.  相似文献

Conformational aspects of the complexation of bis(cyclic tetrapeptide), S,S′-bis[cyclo(Gly-l-hemiCys-Sar-l-Pro)] (BCGCSP) with a metal cation were studied. Binding constants of BCGCSP with several cations were determined in aqueous solution, using circular dichroism (c.d.) titration curves. The values were compared with those of two mono-cyclic tetrapeptides, cyclo[Gly-l-Cys[Bzl(OMe)]-Sar-l-Pro] and cyclo(Sar-l-Pro-Sar-l-Pro). When complexing with alkali metal cations, BCGCSP exhibits selective affinity for Rb+ in preference to Li+, Na+, and K+. Complexing with alkaline earth metal cations, the peptide binds Ba2+ selectively. In addition, BCGCSP shows a marked Ba2+/Ca2+ selectively compared with the other three cyclic peptides. In order to explain these characteristics, a pseudo-inclusion complex with a castanet type structure was proposed as a model of the bis(peptide)—cation complex. The c.d. band ascribed to disulphide (SS) bond transition, showed a red shift upon complex formation. From this observation, it is suggested that conformational fitting of bis(peptide) takes place by changing the geometry of the peptide backbone and covalent CSSC bridge upon complexation with a metal cation.  相似文献

Changes in mechanical properties and chemical nature of the cell walls of the different zones along elongating maize ( Zea mays L. cv. LG 11) roots were analyzed and the following results were obtained. (1) The apical region 2 to 5 mm from the tip of 15 mm long roots showed rapid elongation whereas the region 8–10 mm from the tip showed very little growth. (2) The minimum stress-relaxation time (To) and the mean stress-relaxation rate (R) of the cell wall were small whereas the maximum stress-relaxation time (Tm) was large in the region where cell elongation was optimum. The To and R increased and the Tm decreased gradually towards the base of the root. (3) The amounts of non-cellulosic polysaccharides of the cell wall were highest in the region 1.5–2.5 mm from the tip, decreasing until 5 mm from the tip, and then increasing towards the base. However, the proportion of this fraction in the total cell wall polysaccharides was highest in the extreme tip (cap and meristem, 0–1 mm) and decreased towards the base. (4) Major neutral sugars constituting the non-cellulosic polysaccharides of the cell wall were xylose, arabinose, galactose and glucose, with minor amounts of rhamnosc. mannose and fucose. The 1–15 mm region was on the whole rich in glucose and xylose and contained arabinose to a lesser extent. However, the chemical nature in the apical region, (0–2 mm, was rather special, being rich in galactose and fucose. (5) The cell wall of maize roots contained, as a whole, only little pectic substances but was high in hemicellulose 1 (rich in xylose, arabinose and glucose) and hemicellulose 2 (rich in glucose and xylose). (6) It appeared that in the elongating region (apical 2 to 5 mm) the cell elongation rate (CET) showed a rather good correlation with the parameters of mechanical properties (To, Tm and R) and with neutral sugar compositions in the non-cellulosic polysaccharides.  相似文献
