We present an analysis of the electrostatic properties in the catalytic site of papain (EC 3.4.22.2), an archetype enzyme of the C1 cysteine proteinase family, and we investigate their possible role in the formation, stabilization and regulation of the Cys25((-))...His159((+)) catalytic ion pair. The electrostatic properties were computed using a reassociation method based in multicentered multipolar expansions obtained from ab initio quantum calculations of overlapping protein fragments. Solvent effects were introduced by coupling the use of multicentered multipolar expansions to two continuum boundary element methods to solve the Poisson and the linearized Poisson-Boltzmann equations. The electrostatic profile found in the proton transfer region of papain showed that this enzyme has a well-defined electrostatic environment to favor the formation and stabilization of the catalytic ion pair. The papain catalytic site electrostatic profile can be considered as an electrostatic fingerprint of the papain family with the following characteristics: (i) the presence of a net electric field highly aligned in the (Cys25)-SG-->(His159)-ND1 direction; (ii) the electrostatic profile has a saddle-point character; (iii) it is basically a local environmental effect. Furthermore, our analysis describes a possible regulatory mechanism (the E(SG-->ND1) attenuation effect) controlling the ion pair reactivity and permits to infer the Asp57 acidic residue as the most probable candidate to act as the electrostatic modulator.  相似文献

We determined the amounts of [35S]-glycosaminoglycans (GAGs) found on the intracellular, pericellular and extracellular compartments of primary cultures of astrocytes derived from newborn rat cortex and cerebellum in vitro. Our results show that the greatest portion of newly synthesized GAGs were found in different cellular compartments, depending on the source of the astrocytes. In the cells derived from the cerebellum, the proportion of [35S]-GAGs secreted to the culture medium preponderates over the amount found in the two other compartments, whereas cells derived from the cortex accumulated higher proportions of [35S]-GAGs in the intracellular compartment than in the two other compartments. Cortical and cerebellar glial cells synthesised and secreted heparan sulfate (HS) and chondroitin 4-sulfate (C-4S). HS was predominantly accumulated on the pericellular surface, while C-4S was mostly secreted to the culture medium. Beside the difference on the distribution of total [35S]-GAGs among the three cellular compartments, no difference was observed on the relative proportions of HS and C-4S within each compartment. By defining the source of GAGs, the present study may help to complement and extend information on biosynthesis of these compounds by mammalian glial cells.  相似文献

The antimicrobial activity of a methanol extract and isolated constituents of Mitracarpus scaber, a species used in folk medicine by West African native people, was evaluated against Staphylococcus aureus and Candida albicans strains. The mitracarpus methanol extract possesses both antibacterial and antimycotic activities (minimum inhibitory concentration-MIC 31.25 and 62.50 microg ml-, respectively). This extract was subsequently fractioned and monitored by bioassays leading to the isolation of seven compounds screened for antibacterial and antimycotic activities. Among these compounds, gallic acid and 3,4,5-trimethoxybenzoic acid inhibited the growth of Staph. aureus (MIC 3.90 and 0.97 microg ml-). 4-Methoxyacetophenone and 3,4,5-trimethoxyacetophenone effectively inhibited C. albicans (MIC 1.95 microg ml-). The other compounds (kaempferol-3-O-rutinoside, rutin and psoralen) which were also isolated showed low antibacterial and antimycotic activities (125-500 microg ml-).  相似文献

Morphology of the postotic laterosensory canal was surveyed across loricarioid and outgroup catfishes in order to resolve conflicting statements regarding homology and phylogenetic significance of intrinsic character variation. A pterotic branch is widespread among catfishes and has been identified as a synapomorphy for siluriforms, but its presence in loricarioid catfishes has been disputed. In contrast to previous statements that absence of a pterotic branch is synapomorphic for loricarioids, we confirm the presence of a pterotic branch in Nematogenys inermis and other trichomycterids, callichthyids, and loricariids. The pterotic branch is secondarily absent in scoloplacids and astroblepids. We present criteria for establishing homology of the pterotic branch and review character state optimization schemes on the currently accepted phylogeny. The postotic region of loricariids is further specialized in having an expanded swimbladder capsule that incorporates the trunk lateral line canal and has a lateral opening covered by a greatly expanded pterotic complex. The trunk lateral line enters the swimbladder capsule mesial to the pterotic lateral wall and passes anteromedially as a fleshy tube before forming the postotic canal in the pterotic, a morphology reported previously for a single loricariid representative. Variation in the relative extent and topographic position of postotic canal branches and other morphologies is diagnostic of certain loricariid taxa, suggesting a rich character complex of potential utility in phylogeny reconstruction.  相似文献

A novel 3alpha,6beta,7beta-triol tropane alkaloid esterified by two benzoyl residues was isolated from the leaves of the endemic cuban species, Erythroxylon alaternifolium. Another novel 3alpha,6alpha,7beta-triol tropane alkaloid esterified by trimethoxycinnamoyl and trimethoxybenzoyl residues was isolated from the leaves and stem bark of a second endemic cuban species, Erythroxylon rotundifolium. Their structures were elucidated as 3alpha,7beta-dibenzoyloxy-6beta-hydroxy-tropane and 3alpha-(3,4,5-trimethoxycinnamoyloxy)-7beta-(3,4,5- trimethoxybenzoyloxy)-6alpha-hydroxy-tropane by spectroscopic methods including 2D-NMR techniques.  相似文献

We report that the Bjornstad syndrome gene maps to chromosome 2q34-36. The clinical association of sensorineural hearing loss with pili torti (broken, twisted hairs) was described >30 years ago by Bjornstad; subsequently, several small families have been studied. We evaluated a large kindred with Bjornstad syndrome in which eight members inherited pili torti and prelingual sensorineural hearing loss as autosomal recessive traits. A genomewide search using polymorphic loci demonstrated linkage between the disease gene segregating in this kindred and D2S434 (maximum two-point LOD score = 4.98 at theta = 0). Haplotype analysis of recombination events located the disease gene in a 3-cM region between loci D2S1371 and D2S163. We speculate that intermediate filament and intermediate filament-associated proteins are good candidate genes for causing Bjornstad syndrome.  相似文献

Introduction – The use of the average analytical signal for the construction of curves by the least squares method (LSM) over the standard addition method (SAM) is widespread. It would be advantageous, however, to find a way to avoid intermediary averages, which are known to be the cause of significant increases in standard deviations (SD). Objective – To develop a protocol that uses all gathered data to create curves by LSM over SAM. To use Excel® for the estimation of y = mx + b and R2 rather than using LSM equations for the SD of m, x and b. Methodology – The level of lead (II) in the bark (cork) of Quercus suber Linnaeus was determined using differential pulse anodic stripping voltammetry (DPASV). Three current samples were taken for each of the four standard additions. These signals were combined for adjustment by LSM. The results were compared with those obtained after averaging the current for each addition, and the expression of uncertainty in the measurements determined. Results – The new method shows an expanded uncertainty of ± 0.3321 μg/g (nearly 1.42%). The difference between the results obtained by the new and the old method is 0.01 μg/g (23.41 and 23.40 μg/g). The limit of detection changed approximately from 4.8 to 4 μg/g and the relative SD approximately from 9 to 6%. Conclusion – The absence of intermediary averages in curves improved the determination of lead (II) in cork by DPASV. Estimation of SD only with LSM equations produced results that were significantly worse. The changes are large enough to transform an apparently internally non‐validated procedure (repeatability for precision) into an internally validated procedure. Genetic diversity of present American populations results from very complex demographic events involving different types and degrees of admixture. Through the analysis of lineage markers such as mtDNA and Y chromosome it is possible to recover the original Native American haplotypes, which remained identical since the admixture events due to the absence of recombination. However, the decrease in the effective population sizes and the consequent genetic drift effects suffered by these populations during the European colonization resulted in the loss or under-representation of a substantial fraction of the Native American lineages. In this study, we aim to clarify how the diversity and distribution of uniparental lineages vary with the different demographic characteristics (size, degree of isolation) and the different levels of admixture of extant Native groups in Colombia. We present new data resulting from the analyses of mtDNA whole control region, Y chromosome SNP haplogroups and STR haplotypes, and autosomal ancestry informative insertion-deletion polymorphisms in Colombian individuals from different ethnic and linguistic groups. The results demonstrate that populations presenting a high proportion of non-Native American ancestry have preserved nevertheless a substantial diversity of Native American lineages, for both mtDNA and Y chromosome. We suggest that, by maintaining the effective population sizes high, admixture allowed for a decrease in the effects of genetic drift due to Native population size reduction and thus resulting in an effective preservation of the Native American non-recombining lineages.  相似文献
