The distribution and abundance of planktonic calanoid copepods were studied from samples collected at 13–20 stations during four oceanographic cruises (pre- and post- monsoons, and during northeast (NE) and southwest (SW) monsoons) performed between 1998 and 2000 in the Straits of Malacca. Space and time variations of calanoid copepods were described using univariate (number of species, diversity indices, abundance) as well as multivariate (MDS, ANOSIM, SIMPER) techniques from the Plymouth Routines in Multivariate Ecological Research (PRIMER) package. There were significant differences in abundance between the cruises. k-Dominance curves also revealed significant differences in the relative species abundance distributions among the monsoon periods, and a decrease in diversity from northern to southern parts of the Straits during each cruise. Multi-dimensional scaling revealed four groups of abundances with differences in species composition. Evidence from analysis of similarity (ANOSIM) suggested that the differences in communities among monsoon periods were significant, although spatial differences among samples in geographic locations in the northern, central and southern parts of the Straits were insignificant. These differences resulted from an overall change in the balance of relative abundance of few dominant species, rather from changes of many species. This study was planned to identify the chromosomal location of esterase loci in wheat (Triticum aestivum), in comparison to Aegilops uniaristata, using wheat Ae. uniaristata disomic addition and translocation lines. Challenges associated with the efficient and effective preparation of micro- and nanoscale (micro- and nanogram) clinical specimens for proteomic applications include the unmitigated sample losses that occur during the processing steps. Herein, we describe a simple "single-tube" preparation protocol appropriate for small proteomic samples using the organic cosolvent, trifluoroethanol (TFE) that circumvents the loss of sample by facilitating both protein extraction and protein denaturation without requiring a separate cleanup step. The performance of the TFE-based method was initially evaluated by comparisons to traditional detergent-based methods on relatively large scale sample processing using human breast cancer cells and mouse brain tissue. The results demonstrated that the TFE-based protocol provided comparable results to the traditional detergent-based protocols for larger, conventionally sized proteomic samples (>100 microg protein content), based on both sample recovery and numbers of peptide/protein identifications. The effectiveness of this protocol for micro- and nanoscale sample processing was then evaluated for the extraction of proteins/peptides and shown effective for small mouse brain tissue samples (approximately 30 microg total protein content) and also for samples of approximately 5000 MCF-7 human breast cancer cells (approximately 500 ng total protein content), where the detergent-based methods were ineffective due to losses during cleanup and transfer steps. 相似文献

The insect repellent DEET (0.1% concentration), used as a mosquito oviposition deterrent in the laboratory, influenced the retention and maintenance of mature eggs by caged gravid female Aedes albopictus Skuse. This egg-retention mechanism could benefit survival because the gravid females were ultimately able to lay maintained eggs upon availability of water, but the length of forced egg-retention time reduced the number of eggs laid per female. Gravid females with retained eggs also laid a higher percentage of eggs that failed to tan, and this percentage increased with time duration of egg-retention. Percent egg hatch was not significantly affected by DEET when used as an oviposition deterrent; however, percent hatch was affected by time duration of egg-retention in both treated (exposed to DEET) and untreated (control) gravid females. The rate of egg hatch was considerably reduced after three weeks of retention; this reduction declined to zero for treated and control females at six and four weeks post-treatment, respectively. The fecundity and fertility of gravid female Ae. albopictus were affected by the time duration of forced egg-retention. 相似文献

The reproductive biology of cobia, Rachycentron canadum, from the coastal waters of Dungun, Malaysia was studied from June 2014 to May 2015. From commercial trawls, a total of 201 samples (combined sexes) were collected (fork lengths [FL] 37.5–124.0 cm; body weights 0.5–20.4 g). The overall sex ratio of females to males was 1:0.9, which was not significantly different (χ2 = 2.12, df = 1; p < .05). Estimations of length at 50% maturity (L50) showed that both sexes matured at approximately 75 cm FL; estimated spawning frequency was 6 days. Mean batch fecundity (BF) ranged from 0.55 to 4.32 million eggs. The average number of eggs per gram of ovary was from 2,100 to 5,400 eggs, and relative fecundity 147 eggs/g. There was a weak positive correlation (r2 = .48) between BF and female FL as well as BF with an ovary‐free body weight (r2 = .56), possibly due to females being in a continuous spawning condition and some possibly half‐spent, based on the histological examination of the female gonads. Despite cobia being asynchronous spawners, the gonadosomatic index in both males and females showed peaks in June, November, and particularly March. Based on histological examination, spawning‐capable males were encountered throughout the study period, whereas spawning‐capable females in the late developing subphase were found mostly in March and April. This is the first study on the reproductive aspects of cobia in Malaysian waters. 相似文献

Glasshouse experiments were conducted to evaluate the influence of L-methionine (L-MET) and L-ethionine (L-ETH) added to soil
on the growth of corn (Zea mays L.) and tomato (Lycopersicon esculentum), respectively. The application of L-MET and L-ETH stimulated C2H4 production in soil by 299- and 313-fold, respectively, over an unamended control. An L-MET treatment of 1.85 mg kg−1 soil was the most effective in increasing shoot height, shoot fresh weight, internodal distance, and stem diameter in two
corn cultivars, Kandy Korn and Miracle, while shoot and root dry weights, leaf width, uppermost leaf collar base distance
and resistance to stem breaking were increased in the case of Kandy Korn only. A significant epinastic response was observed
in the second and third leaves of tomato plants when soil was treated with L-ETH. An L-ETH treatment of 0.2 mg kg−1 soil resulted in the maximum fresh fruit yield, while 0.02 and 2.0 mg kg−1 gave the most fruit and greater average weight of fresh fruit, respectively. Concentrations ranging from 0.002 to 2.0 mg
L-ETH kg−1 soil initiated early fruit formation. Early fruit ripening was observed with an application rate of 20 mg L-ETH kg−1 soil. The mechanism of action of these chemicals could either be attributed to i) substrate-dependent C2H4 production in soil by the indigenous microflora, ii) uptake directly by plant roots followed by metabolism within the tissues,
