Background  

Pathway-targeted or low-density arrays are used more and more frequently in biomedical research, particularly those arrays that are based on quantitative real-time PCR. Typical QPCR arrays contain 96-1024 primer pairs or probes, and they bring with it the promise of being able to reliably measure differences in target levels without the need to establish absolute standard curves for each and every target. To achieve reliable quantification all primer pairs or array probes must perform with the same efficiency.  相似文献

Scanning electron microscopy observation was carried out on head morphology, number and construction of teeth, nostril position and the presence of neuromasts of seven species of leptocephali from six families of the Anguilliformes. The number and form of the teeth vary within the different families, from 76 teeth inGnathophis sp. (Congridae) to 32 teeth inAnarchias yoshiae (Muraenidae). In most of the genera the number of teeth on upper and lower jaws is almost equal, except inGnathophis sp.,Anguilla anguilla andKaupichthys hyoproroides. The unusual size and form of the leptocephali's teeth led us to speculate on the feeding behaviour of these larvae. Tentatively we suggest that the tooth formation of the leptocephali may act as a filtering system, rather than as a predatory apparatus, as its vicious appearance might suggest. Given the largely passive nature of a filtering system, the larvae would be able to concentrate on their migration. Of all the investigated leptocephali, onlyNemichthys scolopaceus shows one olfactory opening on each side of the head, whilst all the other species show two openings. The data obtained from these investigations suggest that teeth form as well as form and position of the nostrils are characteristics of the genera, and possibly also of the species.  相似文献

zeta-Crystallin is a novel nicotinamide adenine dinucleotide phosphate:quinone reductase, present at enzymatic levels in various tissues of different species, which is highly expressed in the lens of some hystricomorph rodents and camelids. We report here the complementary DNA (cDNA) cloning of zeta-crystallin from liver libraries in guinea pig (Cavia porcellus), where zeta-crystallin is highly expressed in the lens, and in the laboratory mouse (Mus musculus), where expression in the lens occurs only at enzymatic levels. A 5' untranslated sequence different from the one previously reported for the guinea pig lens cDNA was found in these clones. We also report the isolation of genomic clones including the complete guinea pig zeta-crystallin gene and the 5' region of this gene in mouse. These results show the presence of two promoters in the guinea pig zeta-crystallin gene, one responsible for expression at enzymatic levels and the other responsible for the high expression in the lens. The guinea pig lens promoter is not present in the mouse gene. This is the first example in which the recruitment of an enzyme as a lens crystallin can be explained by the acquisition of an alternative lens- specific promoter.   相似文献

Synopsis The otolith length and the total fish length of 9 leptocephali, 29 elvers and 51 sub-adult eels were measured. For the 51 eels a significant correlation between otolith and fish length was found. No similar correlation was found for leptocephali and elvers because of their similar total length. It was found that the growth of the otolith from leptocephali and elvers differs from the growth of herring larvae otoliths.  相似文献

Summary Chromosomal DNA restriction fragments carrying the nitrogen fixation (nif) and his genes of Klebsiella pneumoniae were identified in hybridization experiments using a plasmid derived from pRD1 as a radioactive probe. Restriction mapping of 26 genetically characterized chromosomal nif deletions provided a map showing the physical location of nif genes along the chromosome.  相似文献

Cnidarian nervous systems utilize chemical transmission to transfer signals through synapses and neurons. To date, ample evidence has been accumulated for the participation of neuropeptides, primarily RFamides, in neurotransmission. Yet, it is still not clear if this is the case for the classical fast neurotransmitters such as GABA, Glutamate, Acetylcholine and Monoamines. A large repertoire of cnidarian Fast Neurotransmitter related Genes (FNGs) has been recently identified in the genome of the sea anemone, Nematostella vectensis. In order to test whether FNGs are localized in cnidarian neurons, we characterized the expression patterns of eight Nematostella genes that are closely or distantly related to human central and peripheral nervous systems genes, in adult Nematostella and compared them to the RFamide localization. Our results show common expression patterns for all tested genes, in a single endodermal cell layer. These expressions did not correspond with the RFamide expressing nerve cell network. Following these results we suggest that the tested Nematostella genes may not be directly involved in vertebrate-like fast neurotransmission.  相似文献
