Adsorption of fungal spores on a synthetic polymer - High Density Polyethylene was successfully achieved using different pretreatments. Sonication of beads followed by ferric nitrate treatment or use of 0.1% tributyrin gave adsorption upto 46%. Use of dichloromethane as a solvent for sonication is recommended for its better performance in reuse studies (upto 5 times without much decrease in activity). 100 g of immobilized biocatabyst in a 7 L Fluidized Bed Bioreactor was found to perform better than shaker flask at a much lower power input. 相似文献

Abstract A gene encoding a type I topoisomerase (TOP1) was isolated from Candida albicans , sequenced, and expressed in Saccharomyces cerevisiae . The TOP1 gene was identified from a C. albicans genomic library by hybridization with the product of a polymerase chain reaction with degenerate primer sets encoding regions conserved in other TOP1 genes. A clone containing an open reading frame of 2463 bp and predicted to encode a protein of 778 amino acids with sequence similarity to eukaryotic type I topoisomerases was identified. The C. albicans TOP1 gene restored camptothecin sensitivity and increased the topoisomerase activity in S. cerevisiae , indicating that the DNA fragment encodes a functional C. albicans topoisomerase I. 相似文献

High quality RNA with good yield is a prerequisite for carrying out several molecular biology studies. Recalcitrant tissues such as oilseeds pose several problems while isolating good quality RNA. We have standardized a fast and simple protocol for RNA isolation from the seeds of Jatropha curcas, which gives good quality RNA without compromising for the yield. By including pre wash of seed powder with acetone and removal of polysaccharides through selective precipitation, we have been regularly isolating good quality total RNA in the range of 300–450 μg g?1 depending upon tissue type. The RNA isolated by this procedure is devoid of any contaminating DNA. The RNA preparations have been subjected to cDNA synthesis and PCR, and found suitable for these studies. Diabetes from pancreatic β cell death and insulin resistance is a serious metabolic disease in the world. Although the overproduction of mitochondrial reactive oxygen species (ROS) plays an important role in the pathogenesis of diabetes, its specific molecular mechanism remains unclear. Here, we show that the natural Charisma of Aqua (COA) water plays a role in Streptozotocin (STZ) diabetic stress-induced cell death inhibition. STZ induces mitochondrial ROS by increasing Polo-like kinase 3 (Plk3), a major mitotic regulator, in both Beta TC-6 and Beta TC-tet mouse islet cells and leads to apoptosis. Overexpression of Plk3 regulates an increase in mitochondrial ROS as well as cell death, also these events were inhibited by Plk3 gene knockdown in STZ diabetic stimulated-Beta TC-6 cells. Interestingly, we found that natural COA water blocks mitochondrial ROS generation through the reduction of Plk3 and prevents apoptosis in STZ-treated beta cells. Furthermore, using the 3D organoid (ex vivo) system, we confirmed that the insulin secretion of the supernatant medium under STZ treated pancreatic β-cells is protected by the natural COA water. These findings demonstrate that the natural water COA has a beneficial role in maintaining β cell function through the inhibition of mitochondrial ROS-mediated cell death, and it might be introduced as a potential insulin stabilizer. 相似文献

Bacterial biosensors can measure pollution in terms of their actual toxicity to living organisms. A recombinant bacterial biosensor has been constructed that is known to respond to toxic levels of Zn2+, Cd2+ and Hg2+. The zinc regulatory gene zntR and zntA promoter from znt operon of E. coli have been used to trigger the expression of GFP reporter protein at toxic levels of these ions. The sensor was induced with 3–800?ppm of Zn2+, 0.005–4?ppm of Cd2+ and 0.001–0.12?ppm of Hg2+ ions. Induction studies were also performed in liquid media to quantify GFP fluorescence using fluorimeter. To determine the optimum culture conditions three different incubation periods (16, 20 and 24?h) were followed. Results showed an increased and consistent fluorescence in cells incubated for 16?h. Maximum induction for Zn2+, Cd2+ and Hg2+ was observed at 20, 0.005 and 0.002?ppm, respectively. The pPROBE-zntR-zntA biosensor reported here can be employed as a primary screening technique for aquatic heavy metal pollution. 相似文献
