Background
The metabolic pathway of folate is thought to influence DNA stability either by inducing single/double stranded breaks or by producing low levels of S-adenosyl-methionine leading to abnormal gene expression and chromosome segregation. Polymorphisms in the genes encoding enzymes in the folate metabolism pathway show distinct geographic and/or ethnic variations and in some cases have been linked to disease. Notably, the gene Methylenetetrahydrofolate reductase (MTHFR) in which the homozygous (TT) state of the polymorphism c.665C>T (p.A222V) is associated with reduced specific activity and increased thermolability of the enzyme causing mild hyperhomocysteinemia. Recently several studies have suggested that men carrying this polymorphism may be at increased risk to develop infertility.Methodology/Principal Findings
We have tested this hypothesis in a case/control study of ethnic French individuals. We examined the incidence of polymorphisms in the genes MTHFR (R68Q, A222V and E429A), Methionine synthase reductase MTRR; (I22M and S175L) and Cystathionine beta-synthase (CBS; G307S). The case population consisted of DNA samples from men with unexplained azoospermia (n = 70) or oligozoospermia (n = 182) and the control population consisted of normospermic and fertile men (n = 114). We found no evidence of an association between the incidence of any of these variants and reduced sperm counts. In addition haplotype analysis did not reveal differences between the case and control populations.Conclusions/Significance
We could find no evidence for an association between reduced sperm counts and polymorphisms in enzymes involved in folate metabolism in the French population. 相似文献193.

The aim of the present study was to investigate a possible role of the AP-1 signaling cascade in the process of wasting associated with cancer cachexia at the level of skeletal muscle. The injection of virus containing the TAM67 protein (a blocker of the AP-1 protein) to the gastrocnemius muscle of tumour-bearing rats resulted in a significant recovery of the muscle mass (which is dramatically reduced as a result of tumour burden), therefore suggesting that AP-1 is certainly involved in the signaling associated with muscle protein accretion. In conclusion, the gene therapy approach presented here clearly suggests an important role for AP-1 in muscle signaling during catabolic states. 相似文献

ortho-Hydroxybenzaldehyde semicarbazone (salicylaldehyde semicarbazone) undergoes chemo-selective hydrolysis of the iminic carbon nitrogen double bond through its reaction with [RuCl2(dmso)4] in ethanol in the presence of water, yielding free salicylaldehyde and semicarbazide that remains coordinated to the ruthenium ion as a bidentate N,O-donor to afford [RuCl2(dmso)2(semicarbazide)] · 2H2O complex. The ruthenium-semicarbazide complex has been characterized by 1H NMR and FTIR spectroscopies and X-ray diffraction methods. Related semicarbazones, derived from p-hydroxybenzaldehyde and benzaldehyde, were not hydrolyzed under the same conditions, suggesting a significant role of the structural o-hydroxy motive in the reaction. Theoretical studies were performed in order to gain further insight on the mechanism of reaction. Results support the hypothesis that the ortho-hydroxy moiety, in the keto tautomeric form, participates in the chemo-selective hydrolysis promoted by [RuCl2(dmso)4]. 相似文献

Shotgun genome sequencing is rapidly emerging as the method of choice for the identification of microsatellite loci in nonmodel organisms. However, to the best of our knowledge, this approach has not been applied to marine algae so far. Herein, we report the results of using the 454 next‐generation sequencing (NGS) platform to randomly sample 36.0 and 40.9 Mbp (139,786 and 139,795 reads, respectively) of the genome of two red algae from the northwest Iberian Peninsula [Grateloupia lanceola (J. Agardh) J. Agardh and a still undescribed new member of the family Cruoriaceae]. Using data mining tools, we identified 4,766 and 5,174 perfect microsatellite loci in 4,344 and 4,504 sequences/contigs from G. lanceola and the Cruoriaceae, respectively. After conservative removal of potentially problematic loci (redundant sequences, mobile elements), primer design was possible for 1,371 and 1,366 loci, respectively. A survey of the literature indicates that microsatellite density in our Rhodophyta is at the low end of the values reported for other organisms investigated with the same technology (land plants and animals). A limited number of loci were successfully tested for PCR amplification and polymorphism finding that they may be suitable for population genetic studies. This study demonstrates that random genome sequencing is a rapid, effective alternative to develop useful microsatellite loci in previously unstudied red algae. 相似文献

The effects of three heavy metals (Co, Ni and Cd) on the growth kinetics of five bacterial strains with different characteristics
(Pseudomonas sp., Phaeobacter sp. strain 27-4, Listonella anguillarum, Carnobacterium piscicola and Leuconostoc mesenteroides subsp. lysis) were studied in a batch system. A bivariate model, function of time and dose, is proposed to describe simultaneously all
the kinetic profiles obtained by incubating a microorganism at increasing concentrations of individual metals. This model
combines the logistic equation for describing growth, with a modification of the cumulative Weibull’s function for describing
the dose-dependent variations of growth parameters. The comprehensive model thus obtained—which minimizes the effects of the
experimental error—was statistically significant in all the studied cases, and it raises doubts about toxicological evaluations
that are based on a single growth parameter, especially if it is not obtained from a kinetic equation. In lactic acid bacteria
cultures (C. piscicola and L. mesenteroides), Cd induced remarkable differences in yield and time course of characteristic metabolites. A global parameter is defined
(ED50,τ: dose of toxic chemical that reduces the biomass of a culture by 50% compared to that produced by the control at the time

Streptomyces tacrolimicus (ATCC 55098) was reported to produce the immunosuppressant tacrolimus. The wild-type strain sporulates sparsely and produces very low levels of this immunosuppressant. The lack of genetic knowledge of this strain has hampered strain improvement. In this work, we have cloned the gene encoding a γ-butyrolactone receptor protein (Gbr). The gbr gene is linked to two genes encoding two subunits of the dihydroxyacetone kinase, putatively involved in the biosynthesis of the dihydroxyacetone phosphate precursor of γ-butyrolactone but is not flanked by γ-butyrolactone synthetase genes. The Gbr protein was overexpressed in Escherichia coli and purified. Electrophoretic mobility shift assays showed that Gbr binds to a specific autoregulatory element sequence located 338 bp upstream of the gbr gene, indicating that its expression is self-regulated. The deletion mutant Δgbr showed a very early and intense sporulation in two different media. A phenotype similar to that of the wild-type strain was restored by complementation of the Δgbr mutant with a wild-type gbr allele. Duplication of the gbr gene resulted in a slower sporulation. The Δgbr mutant produced much lower amount (32%) of tacrolimus quantified by high performance liquid chromatography. This analysis, using an optimised system, allowed the resolution of tacrolimus from ascomycin and other contaminant metabolites. Our results indicate that the Gbr protein regulates negatively the sporulation and positively the production of tacrolimus. 相似文献

近年的研究已经揭示出表型可塑性可以通过营养世代遗传。从这个意义上讲,环境诱导的基因调控变化(例如DNA甲基化,即表观遗传变异)会导致可逆的塑性反应传递给后代。这种跨世代的可塑性在克隆植物中尤其重要,因为有性繁殖减少会降低通过遗传变异进行适应的可能性。许多最具侵略性的植物入侵者都具有克隆性,而且克隆性被认为是植物入侵性的关键。本研究中,我们的目标是确定在克隆入侵者喜旱莲子草(Alternanthera philoxeroides)中是否发生了跨代效应,以及这种效应在本地和非本地群落之间是否存在差异。在同质园实验中,我们将采自巴西(原产地)和伊比利亚半岛(非原产地)的喜旱莲子草种群分别种植在养分高和低的土壤中,后代植株被移植到高养分条件下作为对照。为了检验DNA甲基化对跨代可塑性的潜在作用,一半的母株用去甲基化剂5-氮杂胞苷处理。在来自原产地和非原产地的种群中均观察到了跨代效应。有趣的是,在原产地种群的生长变量(分株数、茎生物量、根生物量和总生物量)中发现了跨代效应,而在非原产地种群中发现了生物量分配具有跨代效应。在原产地种群中出现的跨代效应可以用“银勺”效应来解释,而非原产地种群的跨代效应可以归因于DNA甲基化引起的表观遗传传递。本研究强调了跨代效应对克隆植物生长的重要性,这有助于理解众多克隆植物能够成功繁殖扩张的机制。 相似文献
