Grasserie, a polyorganotrophic disease caused by Bombyx mori nucleopolyhedrovirus (BmNPV), accounts for lethal infection to fifth instar silkworm larvae. It was found that nanoparticle (NP)-induced morphological transformation of BmNPV polyhedra could reduce the infectivity of BmNPV both in cell line and in silkworm larvae. Initially, 11 NPs were screened for evaluation of their nature of interaction with polyhedra surface through scanning electron microscopy. Amongst these NPs, lipophilically coated silica nanoparticle (SNPL), alumina nanoparticles in the hexagonal close-packed α structure and aspartate capped gold nanoparticle transformed polyhedra were tested for their infectivity in B. mori cell line using cytopathic effect and plaque reduction assay. SNPL was evaluated for its bio-efficacy in fifth instar silkworm larvae. The study of polyhedra morphology as a function of NP concentration showed severe ‘roughening’ of the polyhedra with replacement of the regular facets by a large number of irregular ones by SNPL, and this caused transition of highly infectious polyhedra into a nearly spherical, non-infectious structure. A moderate polyhedra roughening was observed for alumina NPs, and no roughening was noticed for gold NPs. The morphological changes could be correlated with reduction of virus-induced cytopathic effect and plaque formation, and increased survival rate of SNPL transformed polyhedra infected silkworm larvae to 70.09?±?6.61 % after 96 h. In this group, 61.04?±?8.03 % larvae formed normal cocoons from which moths eclosed, laid eggs and larvae emerged. This study could lead to open up newer pathways for designing nano pharmaceuticals to combat other viral diseases.  相似文献

Abstract

Various amino acid similarity matrices have been derived using data on physicochemical properties and molecular evolution. Conformational similarity indices, CSXX′, between different residues are computed here using the distribution of the main-chain and side-chain torsion angles and the values have been used to cluster amino acids in proteins. A subset of these parameters, CSAX′ indicates the extent of similarity in the main-chain and side-chain conformations (φ ψ and χ1) of different residues (X) with Ala (A) and is found to have strong correlation with α-helix propensities. However, no subset of CSXX′ provides any linear relationship with β-sheet propensities, suggesting that the conformational feature favouring the location of a residue in an a-helix is different from the one favouring the β-sheet. Conformationally similar residues (close CSAX values) have similar steric framework of the side-chain (linear/branched, aliphatic/aromatic), irrespective of the polarity or hydrophobicity. Cooperative nucleation of helix may be facile for a contiguous stretch of residues with high overall CSAX values.  相似文献

Despite discovery of the pathogen more than 100 years ago, tuberculosis (TB) continues to be a major killer disease worldwide. Currently a third of world population is infected and multiple-drug-resistant (mdr) TB registers maximum mortality by a single pathogen. Nanomedicine provides enormous opportunity for developing novel drugs. We have recently demonstrated surface-modified-lipophilic-nanosilica as drug to combat malaria and 100% lethal virus, BmNPV. Nanosilver possesses inherent antibacterial properties, but toxicity is a major concern. We hypothesized that capping with nature-inspired biomolecules, bovine serum albumin (BSA) and Poly-n-vinyl-pyrrolidone (PVP) used as blood volume extender, might insure biosafety. BSA-nano-Ag was found to be more stable than PVP-nano-Ag at physiological pH. In this first ever study on clinical isolates collected from TB endemic areas, we report, BSA-nano-Ag act as potent anti-TB drug. Further study with (human serum albumin)-nano-Ag and core-shell-nano-Ag could increase the biocompatibility of oral TB drug formulations without compromising on the efficacy of the drug.  相似文献

In this paper we present a new method for evaluating soil and plant tests for phosphate (P). We apply it to field experiments conducted over 2 years in four widely separated tea gardens of the Terai and the Dooars regions at the Himalayan foothills of India. We applied 5 levels of P to each site and measured yield, P concentration in the leaves, and Bray P values through time. Leaf P concentrations were closely described by a 3-dimensional Mitscherlich equation in which one of the independent variables was the amount of P applied and the other was the initial Bray P value. From this, we calculated the relative effectiveness of the applied P and the Bray P. The effectiveness of the applied P relative to that measured by Bray P, ranged from 0.87 down to 0.55. We added values for applied P and Bray P, with appropriate weighting for relative effectiveness, to calculate an effective level of P for each treatment. The relation between yield and effective P was then closely described by a Mitscherlich equation modified to permit a different maximum for each site. These relationships were used to show that P requirement was greatest on the most productive sites and in the middle of the growing season when growth rates were largest. We also tested the relation between leaf P concentration and yield. Yield was linearly related to leaf P over a concentration range of about 3 to about 4 g P kg?1. The regression lines for different yield levels were close to parallel but with different intercepts. We propose that leaf P analysis could be used to monitor P status and that soil tests plus the values for relative effectiveness of applied P be used to estimate fertilizer requirement.  相似文献

Complement sensitization of red blood cells (RBCs) can cause life-threatening hemolytic anemias. We have previously shown that complement receptor 1 (CR1) derivatives specifically the N-terminal region with decay accelerating activity (DAA) for inactivation of a key enzyme in the complement cascade can reduce complement-mediated RBC destruction in vitro and in an in vivo mouse model of hemolytic transfusion reaction. In the present study, we have modeled the N-terminal CR1 molecule based on the X-ray crystal structure of decay accelerating factor and the NMR structure of a homologous CR1 domain. Based on the homology model, we identified a 34-mer peptide encompassing the putative DAA which in vitro reduced hemolysis, C3a release and surface C3 deposition. More importantly, this peptide at 0.6 mM was effective in prolonging survival of transfused incompatible RBCs in vivo. Our results indicate that CR1-based structure-function studies may provide insights for developing structure-derived transfusion therapeutics in the future.  相似文献

The present study has been undertaken to observe the effect of aqueous extract of M. oleifera (MO) leaf (300mg/kg body weight) on mean ulcer index, enterochromaffin (EC) cells and serotonin (5-hydroxytryptamine; 5-HT) content of ulcerated gastric tissue. Ulceration was induced by using aspirin (500 mg/kg, po), cerebellar nodular lesion and applying cold stress. In all cases increased mean ulcer index in gastric tissue along with decreased EC cell count was observed with concomitant decrease of 5-HT content. Pretreatment with MO for 14 days decreased mean ulcer index, increased both EC cell count and 5-HT content in all ulcerated group, but treatment with ondansetron, a 5-HT3 receptor antagonist, along with MO pretreatment increased mean ulcer index, decreased 5-HT content without any alteration in EC cell count. The results suggest that the protective effect of MO on ulceration is mediated by increased EC cell count and 5-HT levels which may act via 5-HT3 receptors on gastric tissue.  相似文献

The effects of four indole-3-butyric acid (IBA) concentrations and two propagation methods were studied in a lowbush blueberry (Vaccinium angustifolium Ait.) clone collected from natural stands in Newfoundland and Labrador, Canada. Lowbush blueberry cultures were established in vitro from nodal explants on a modified cranberry (V. macrocarpon Ait.) tissue culture medium containing zeatin (2 μM). Blueberry plants propagated by in vitro shoot proliferation (TC) and by conventional softwood cuttings (SC) were evaluated for growth and morphology. Significant interactions for morphological characteristics were observed among the treatments. The IBA concentration had an effect on morphology of propagated plants, increasing the concentration of IBA increased stem length and leaves per stem across propagation methods. Stems per plant increased with IBA concentration up to 20 μM in SC plants, but not in TC plants. Plant vigor was affected by neither IBA concentration nor propagation method. The TC plants produced longer and more stems with more leaves per stem than the conventional cuttings. In vitro culture on zeatin-containing nutrient medium apparently induces the juvenile branching characteristics that favored enhanced vegetative growth with more stems and leaf production. It is suggested that IBA may serve as a physiologically active form of auxin in contributing to increased stem and leaf production in lowbush blueberry SC plants but not in TC plants.  相似文献

We present a new medium‐sized basal sauropodomorph, S efapanosaurus zastronensis gen. et sp. nov. , from the Upper Triassic?Lower Jurassic Elliot Formation of South Africa. It is represented by parts of the postcranial skeleton of at least four individuals, including: cervical, dorsal, sacral and caudal vertebrae, most of the forelimb, and part of the hindlimb. Sefapanosaurus bears several autapomorphies of the astragalus, and referred material also shows autapomorphic features. The inclusion of Sefapanosaurus in a phylogenetic analysis places it within the group of sauropodomorphs more closely related to sauropods than to Massospondylus (i.e. Sauropodiformes), increasing the currently known diversity of the so‐called ‘transitional forms’ leading to Sauropoda. Character optimization revealed the presence of several features that are common for taxa placed within the transitional branches basal to Sauropoda. Sefapanosaurus, together with other transitional sauropodomorphs reported during the last decade, highlights the importance of Gondwanan taxa for understanding the palaeobiodiversity, global distribution, and macroevolutionary changes in the group related to the rise of sauropods. © 2015 The Linnean Society of London  相似文献
