Episodic ataxia (EA) is a rare, familial disorder producing attacks of generalized ataxia, with normal or near-normal neurological function between attacks. Families with autosomal dominant EA represent at least two distinct clinical syndromes. One clinical type of EA (MIM 160120) includes individuals who have episodes of ataxia and dysarthria lasting seconds to minutes. In addition, myokymia (rippling of muscles, diagnosable by electromyography) is evident during and between attacks. Since K+ channel genes are candidate genes for EA, we tested markers near known K+ channel genes for linkage. Using a group of Genethon markers from one such region--chromosome 12p--we found evidence of linkage in four EA/myokymia families. A maximum combined lod score of 13.6 was obtained at theta = 0, with the marker D12S99. A human Ca++ channel gene, CACNL1A1, and three human K+ channel genes--KCNA5, KCNA6, and KCNA1--map close to D12S99, but the Ca++ channel gene is unlikely to be the site of the defect, because crossovers have been observed to occur between the disease gene and a CA-repeat marker located close to this gene. Studies of a large EA family with a different clinical phenotype (MIM 108500), which lacks myokymia but is associated with nystagmus, have excluded the gene causing that disease from the chromosome 12p locus. 相似文献

The mammalian nucleus has a distinct substructure that cannot be visualized directly by conventional microscopy. In this study, the organization of the DNA within the nucleus of multiple myeloma (MM) cells, their precursor cells (monoclonal gammopathy of undetermined significance; MGUS) and control lymphocytes of the representative patients is visualized and quantified by superresolution microscopy. Three‐dimensional structured illumination microscopy (3D‐SIM) increases the spatial resolution beyond the limits of conventional widefield fluorescence microscopy. 3D‐SIM reveals new insights into the nuclear architecture of cancer as we show for the first time that it resolves organizational differences in intranuclear DNA organization of myeloma cells in MGUS and in MM patients. In addition, we report a significant increase in nuclear submicron DNA structure and structure of the DNA‐free space in myeloma nuclei compared to normal lymphocyte nuclei. Our study provides previously unknown details of the nanoscopic DNA architecture of interphase nuclei of the normal lymphocytes, MGUS and MM cells. AbstractConcentrations of heavy metals (Cd, Co, Cr, Cu and Fe) in surface sediments and their partitioning behaviour between exchangeable, reducible (Fe-Mn oxide bound) and organic/residual phases of the sediments in a typical backwater system of Kerala, viz. the southern upstream part of Cochin Estuarine System (South India), have been studied. Spatial and temporal variations of trace metals are discussed with special reference to pH, dissolved oxygen, salinity, organic carbon and texture of sediment. Metal concentrations in the tide gated part of the estuary were found to be significantly higher when compared to metal concentrations reported from the unrestricted part of the Cochin estuarine system and also those from many of the unpolluted estuaries worldwide. The higher levels of heavy metals in the study area and their characteristic distribution and partitioning behaviour in the surficial sediments were attributed to the presence of a salinity barrier across the backwater system and also by the massive use of pesticides and chemical fertilizers in the vast area of agricultural land near the backwater system. 相似文献

Toxins convertthe hepatocellular response to tumor necrosis factor- (TNF-)stimulation from proliferation to cell death, suggesting thathepatotoxins somehow sensitize hepatocytes to TNF- toxicity. Becausenuclear factor-B (NF-B) activation confers resistance to TNF-cytotoxicity in nonhepatic cells, the possibility that toxin-inducedsensitization to TNF- killing results from inhibition ofNF-B-dependent gene expression was examined in the RALA rathepatocyte cell line sensitized to TNF- cytotoxicity by actinomycinD (ActD). ActD did not affect TNF--induced hepatocyte NF-Bactivation but decreased NF-B-dependent gene expression. Expressionof an IB superrepressor rendered RALA hepatocytes sensitive toTNF--induced apoptosis in the absence of ActD. Apoptosis was blockedby caspase inhibitors, and TNF- treatment led to activation ofcaspase-2, caspase-3, and caspase-8 only when NF-B activation wasblocked. Although apoptosis was blocked by the NF-B-dependent factornitric oxide (NO), inhibition of endogenous NO production did notsensitize cells to TNF--induced cytotoxicity. Thus NF-Bactivation is the critical intracellular signal that determines whetherTNF- stimulates hepatocyte proliferation or apoptosis. Althoughexogenous NO blocks RALA hepatocyte TNF- cytotoxicity, endogenousproduction of NO is not the mechanism by which NF-B activationinhibits this death pathway. 相似文献

Lymphoid cell subclasses in rejecting renal allograft in the rat 总被引:4,自引:0,他引:4
We have quantitated the frequency of lymphoid cell subsets in rejecting renal allografts and in the spleen of the allograft recipient during drug-unmodified rejection in the rat. The number of inflammatory (white) cells in the graft was approximately similar to the number of white cells responding to the allograft in the recipient spleen. The inflammatory population of the graft consisted of lymphoid cells and mononuclear phagocytes, with increasing numbers of macrophages toward the end of rejection. Analysis of allograft cellular dispersates with monoclonal antibodies directed to the lymphoid cell subsets demonstrated that although the majority of allograft-infiltrating lymphocytes were T cells, a sizable B-cell proliferation and immunoglobulin synthesis was associated with the inflammatory response of rejection. Within the T-cell subset, the T suppressor/killer cells predominated in the graft whereas the predominant lymphoid cell subset responding to the allograft in the recipient spleen was the T helper cell. 相似文献

Fungi are usually thought not to have a boron (B) requirement. It is not known if mycorrhizas take up B from low concentrations
that are common in forest soils, as fungi might also immobilise B. Here, we studied the B concentrations in sporophores of
49 ectomycorrhizal and 10 saprotrophic fungi to assess whether B is translocated in mycelium or not. Additionally, P and metal
concentrations were measured for comparison. Variability both within species and between species was very large, as the lowest
measured B concentration was 0.01 mg kg−1 in Amanita muscaria, and the highest was 280 mg kg−1 in Paxillus involutus. There was no clear difference between saprotrophic and mycorrhizal fungi. The majority of species did not accumulate B at
more than 0.01–3 mg kg−1, but there were some species that consistently had median concentration values higher than 5–6 mg kg−1 and much higher maximum values, particularly Paxillus involutus, Lactarius necator and several Russula species. Most species increased their B concentration in B fertilised plots, but there were exceptions, particularly Rozites caperatus and Lactarius camphoratus. Boron concentrations did not correlate with those of other elements. In conclusion, B is translocated in the mycelia of most
of the studied species. The differences between species may be due to differences in their water use, or carbohydrates used
in translocation. It remains to be studied, if B concentrations in mycorrhizas or mycelia in soil are in the same order of
magnitude as the larger ones found here, and if this has any effects on the host plants. 相似文献

The Medium Resolution Imaging Spectrometer (MERIS/Envisat) was launched in March 2002 for coastal zone monitoring. Preliminary data from MERIS show that its imagery of large lakes is superior to that of other common ocean colour sensors. The main objective of the present study is to evaluate the MERIS data on large European lakes, Vänern and Vättern in Sweden and Peipsi in Estonia/Russia. In these lakes, coloured dissolved organic matter (CDOM) can be a major contributor to the optical properties of the water. Another characteristic of the waters under investigation is the large temporal and spatial variability in the concentrations of chlorophyll (C Chl) and suspended sediments (C TSS). Potentially toxic cyanobacterial blooms occur in Lake Peipsi in late summer. We have compared the MERIS products from the latest reprocessing (finished in March 2006) with available in situ data. There is a reasonably good correlation between the MERIS algal_2 product and the measured C chl over all three lakes, but no correlation was found for other optically active substances. A significant portion of the pixels (up to 90%) are flagged as invalid results after atmospheric correction. 相似文献

Infertility affects around 1 in 10 men and in most cases the cause is unknown. The Y chromosome plays an important role in spermatogenesis and specific deletions of this chromosome, the AZF deletions, are associated with spermatogenic failure. Recently partial AZF deletions have been described but their association with spermatogenic failure is unclear. Here we screened a total of 339 men with idiopathic spermatogenic failure, and 256 normozoospermic ancestry-matched men for chromosome microdeletions including AZFa, AZFb, AZFc, and the AZFc partial deletions (gr/gr, b1/b3 and b2/b3).AZFa and AZFc deletions were identified in men with severe spermatogenic failure at similar frequencies to those reported elsewhere. Gr/gr deletions were identified in case and control populations at 5.83% and 6.25% respectively suggesting that these deletions are not associated with spermatogenic failure. However, b2/b3 deletions were detected only in men with spermatogenic failure and not in the normospermic individuals. Combined with our previous data this shows an association of the b2/b3 deletion (p = 0.0318) with spermatogenic failure in some populations. We recommend screening for this deletion in men with unexplained spermatogenic failure. 相似文献

Food action plans in many global cities articulate interest in multiple objectives including reducing in‐ and trans‐boundary environmental impacts (water, land, greenhouse gas (GHG)). However, there exist few standardized analytical tools to compare food system characteristics and actions across cities and countries to assess trade‐offs between multiple objectives (i.e., health, equity) with environmental outcomes. This paper demonstrates a streamlined model applied for analysis of four cities with varying characteristics across the United States and India, to quantify system‐wide water, energy/GHG, and land impacts associated with multiple food system actions to address health, equity, and environment. Baseline diet analysis finds key differences between countries in terms of meat consumption (Delhi 4; Pondicherry 16; United States 59, kg/capita/year), and environmental impact of processing of the average diet (21%, 19%, <1%, <1% of community‐wide GHG‐emissions for New York, Minneapolis, Delhi, and Pondicherry). Analysis of supply chains finds city average distance (food‐miles) varies (Delhi 420; Pondicherry 200; United States average 1,640 km/t‐food) and the sensitivity of GHG emissions of food demand to spatial variability of energy intensity of irrigation is greater in Indian than US cities. Analysis also finds greater pre‐consumer waste in India versus larger post‐consumer accumulations in the United States. Despite these differences in food system characteristics, food waste management and diet change consistently emerge as key strategies. Among diet scenarios, all vegetarian diets are not found equal in terms of environmental benefit, with the US Government's recommended vegetarian diet resulting in less benefit than other more focused targeted diet changes. 相似文献
