Using Fura-2AM microfluorimetry, we have shown for the first time that sigma-1 receptor agonist, tricyclic antidepressant amitriptyline, significantly inhibits glutoxim- and molixan-induced Ca2+-responses in rat peritoneal macrophages. The results suggest possible involvement of sigma-1 receptors in the signaling cascade induced by glutoxim or molixan and leading to intracellular Ca2+ concentration increase in macrophages. 相似文献

Apolipoprotein A-I can spontaneously associate with phosphatidylcholine and cholesterol in 2-chloroethanol-water mixture. It was demonstrated, using a spin label technique, that dissolved molecules participate in complex formation. The apolipoprotein A-I/phosphatidylcholine/cholesterol complexes were isolated by gel chromatography. Complexes of three types were prepared and characterized: type A, large heterogeneous aggregates with molecular weight 600 000, sedimentation coefficient 10 S and the following molar composition - protein/phosphatidylcholine/cholesterol, 1:(70-100):(10-12); types B and C, with weight average molecular weights 140 000 and 110 000, average sedimentation coefficients 3.6 S and 1.7 S, respectively. Both types have the same molar composition - protein/phosphatidylcholine/cholesterol, 1:25:8. The dissimilar sedimentation coefficients between complexes B and C may be explained by the difference in the monomer/tetramer ratio (monomer molecular weight 50 000). The spin label sn-1-O-stearoyl-2-O-9'-spiro(4',4'-dimethyloxazolidine-3'-oxyl) heptadecanoylglycero-3-phosphocholine introduced into the complexes A and B showed different thermal properties of these complexes, which may be due to differences in the lipid-protein interactions. 相似文献

Using voltage-clamp technique, the involvement of epoxygenases in immunomodulatory drug glutoxim regulation of Na+ transport in frog skin was investigated. We have shown for the first time that preincubation of the frog skin with epoxygenase inhibitors econazole or proadifen almost completely inhibits the stimulatory effect of glutoxim on Na+ transport. The data suggest the involvement of the enzymes and/or products of epoxygenase oxidation pathway of arachidonic acid metabolism in glutoxim effect on Na+ transport in frog skin epithelium. 相似文献

Using Fura-2AM microfluorimetry, we have shown for the first time that preincubation of macrophages with the calsequestrin inhibitor neuroleptic trifluoperazine leads to a significant inhibition of the store-dependent Ca2+ entry induced by endoplasmic Ca2+-ATPase inhibitors thapsigargin or cyclopiazonic acid in rat peritoneal macrophages. We developed PCR primers against highly conserved regions of the rRNA
operon located within the inverted repeat of the chloroplast genome and
used these to amplify the region spanning from the 3' terminus of the 23S
rRNA gene to the 5' terminus of the 5S rRNA gene. The sequence of this
roughly 500-bp region, which includes the 4.5S rRNA gene and two
chloroplast intergenic transcribed spacer regions (cpITS2 and cpITS3), was
determined from 20 angiosperms, 7 gymnosperms, and 16 ferns (21,700 bp).
Sequences for the large subunit of ribulose bisphosphate
carboxylase/oxygenase (rbcL) from the same or confamilial genera were
analyzed in both separate and combined data sets. Due to the low
substitution rate in the inverted repeat region, noncoding sequences in the
cpITS region are not saturated with substitutions, in contrast to
synonymous sites in rbcL, which are shown to evolve roughly six times
faster than noncoding cpITS sequences. Several length polymorphisms with
very clear phylogenetic distributions were detected in the data set.
Results of phylogenetic analyses provide very strong bootstrap support for
monophyly of both spermatophytes and angiosperms. No support for a sister
group relationship between Gnetales and angiosperms in either cpITS or rbcL
data was found. Rather, weak bootstrap support for monophyly of gymnosperms
studied and for a basal position for the aquatic angiosperm Nymphaea among
angiosperms studied was observed. Noncoding sequences from the inverted
repeat region of chloroplast DNA appear suitable for study of land plant
evolution.

The influence of GABA receptor agonists on the terminal differentiation in vitro of dopaminergic (DA) neurons derived from IPS cells was investigated. GABA-A agonist muscimol induced transient elevation of intracellular Ca2+ level ([Ca2+]i) in the investigated cells at days 5 to 21 of differentiation. Differentiation of cells in the presence of muscimol reduced tyrosine hydroxylase expression. Thus, the presence of active GABA-A receptors, associated with phenotype determination via Ca2+-signalling was demonstrated in differentiating human DA neurons. 相似文献

The synaptic responses induced in motoneurones by the stimulations of the dorsal root (DR), single afferent fibres and reticular formation (RF) were intracellularly recorded in the isolated frog spinal cord. It was shown that argiopine (the selective blocker of glutamate receptors of non-NMDA type) in concentrations ranging from 3.10(-7) to 1.10(-5) M effectively suppressed the di- and polysynaptic, but not the monosynaptic components of EPSP's induced by DR stimulation. The initial reaction to argiopine consisted of the increase of this monosynaptic component of EPSP. In the same concentrations range, argiopine reduced both mono- and polysynaptic EPSP, evoked by RF stimulation. 2-amino-phosphonovaleric acid (1.10(-4) M) did not affect, whereas the kinurenate (1--2.10(-3) M) completely blocked the amplitude of all kinds of synaptic responses. The various effects of argiopine on the responses induced by microstimulation of presynaptic nerve terminals were observed. The data obtained speak in favour of heterogeneity of monosynaptic excitatory inputs in the motoneurones of frog spinal cord. Being the glutamatergic by nature, the inputs differ in the properties of postsynaptic receptors. All of these receptors concerning to non NMDA-type can be divided to argiopine-sensitive and argiopine-resistant. The effects of noradrenaline and dopamine on heterosynaptic facilitation (HSF) were investigated during experiments on giant identified neurons from the cerebral ganglion of the freshwater molluskPlanorbis corneus. It was found that catecholamines, while inducing an increase in the amplitude of response obtained by iontophoretic application of acetylcholine, also sharply reduce the amplitude of EPSP occurring as a result of stimulating the cerebral nerve, although acetylcholine also acts as transmitter in this instance. Catecholamines were also found to exert a blocking action on synaptic transmission during the period of HSF. Noradrenaline washout immediately after blockade of HSF was shown to reinstate and promote continuing independent facilitation of synaptic transmission. Duration of this facilitatory after-effect frequently exceeded that of initial HSF. This would imply that if serotonin promotes development of HSF, then the secretion of noradrenaline and dopamine brought about by certain effects act as the mechanism controlling duration and intensity of HSF under naturally-occurring conditions of the mollusk and perhaps even human nervous system activity.A.
