Amino acid sequence of rabbit ventricular myosin light chain-2: Identity with the slow skeletal muscle isoform

Many studies have established a correlation of differences in the activities of various muscle types with differences in the expression of myosin isoforms. In this paper we report the sequence determination of myosin light chain-2 from rabbit slow skeletal (LC2s) and ventricular (LC2v) nmscles. We sequenced tryptic peptides from LC2v which account for all except a few terminal amino acid residues. The major part (87 residues) of the rabbit LC2s sequence, obtained from tryptic and cyanogen bromide (CNBr) peptides, was found to be identical to rabbit LC2v. Our results provide the first sequence information on LC2s from any species, and lend strong support to the hypothesis that LC2s and LC2v are identical. Comparisons of rabbit LC2v and LC2s with rabbit LC2f (from fast skeletal muscle), and also with chicken LC2f and LC2v, show clearly that LC2s and LC2v from mammalian and avian species are more closely related to each other than they are to LC2f isoforms from the same species.     

Production of pectinases byAspergillus sp using differently pretreated lemon peel as the carbon source

Summary Aspergillus sp strains from decaying lemons were tested for extracellular pectinase production, testing differently pretreated lemon peel as the carbon source instead of pectin. It was found that the production of extracellular polygalacturonase was about the same and that of pectinesterase substantially higher when unwashed fresh lemon peel was used instead of pectin. The culture filtrate obtained showed a clarifying capacity similar to that of a commercial pectinase preparation, but the vitamin C of the juice was less affected by the treatment.     

Increased GH responsiveness to dopamine receptor stimulation in alcohol addicts during the late withdrawal syndrome

In humans the release of growth hormone (GH) elicited by dopamine (DA) and DA agonists may represent a reliable model to assess change in sensitivity of DA receptors. We now report that in chronic alcoholics, 4–7 days after the suspension of alcohol consumption, the increase of GH response to DA infusion was higher than that seen in non alcoholic volunteers. The specificity of this GH response to DA administration was demonstrated by the use of domperidone, a novel peripheral antagonist of DA receptors. These results suggest the development of hyper-responsiveness of DA receptors involved in the control of GH secretion in chronic alcoholics during the later phases of the “withdrawal syndrome”.     

The steroids and fatty acids of the basidiomycete Scleroderma polyrhizum

The fruit bodies of the Basidiomycete Scleroderma polyrhizum have been shown to contain the steroids ergosta-4,6,8(14) 22-tetraen-3-one and 5α,8α-epidoxyergosta-6,22-dien-3β-ol and also palmitic and oleic acids.     

Acid phosphatase from maize scutellum: Negative cooperativity suppression by glucose

Acid phosphatase purified from maize scutellum, upon acylation with succinic anhydride, still shows negative co-operativity for the hydrolysis of glucose-6-phosphate at pH 5.4. This phenomenon is abolished by glucose, for both native and succinylated enzymes, through stimulation of the initial velocities at sub-optimal substrate concentrations. However, negative co-operativity for the enzymatic hydrolysis of p-nitrophenylphosphate at pH 5.4 is suppressed only at high concentrations of glucose. Furthermore, the hydrolysis of p-nitrophenylphosphate is noncompetitively inhibited (low affinity form of the enzyme molecule) by glucose, which suggests the existence of different substrate binding sites.     

Acid hydrolases in the perinuclear secretion granules of the rat preputial gland. A light and electron microscope study

Synopsis Four acid hydrolases in the secretory cells and the sebum of the preputial sebaceous gland of the rat were incestigated cytochemically. A strong -glucuronidase activity was found to occur in the matrix of the perinuclear secretion granules, whereas the granule crystalloids were unreactive. The distribution of acid phosphatase at the light microscope level was similar, though the intensity of the reaction was lower and the number of positive granules smaller. By electron microscopy, the final reaction product of acid phosphatase occurred in patches at the periphery of the granule matrix, as well as in the vesicles adjoining the Golgi stacks, from which the perinuclear granules seemed to arise. In the sebum, the two hydrolases occurred in the background material between the unstained crystalloid masses. There was noN-acetyl--glucosaminidase or aryl sulphatase activity in the gland. The perinuclear granules appear to be secretory lysosomes which, after discharge from the disaggregating cell, release their acid hydrolases into the sebum.     

Pyridoxal phosphate-sensitized photoinactivation of glutamate decarboxylase from Clostridium perfringens

1. l-Glutamate decarboxylase (EC 4.1.1.15) from Clostridium perfringens was inactivated by exposure to visible light at pH6.2. 2. Inactivation does not occur at pH4.6 or in the absence of bound pyridoxal phosphate. 3. On prolonged photo-oxidation six histidine residues per molecule of enzyme were destroyed. 4. The loss of six cysteine residues per molecule occurred both in irradiated samples and in controls oxygenated in the dark. 5. This dark-oxidation of cysteine residues is apparently required before the photo-oxidation process. 6. The absorbance, fluorescence and circular-dichroism properties of the enzyme as well as its elution volume during Sephadex gel-filtration were unaffected by prolonged irradiation. 7. However, an apparently homogeneous product of photo-oxidation could be separated from the control enzyme by ion-exchange chromatography. 8. The K(m) for l-glutamate was unchanged in an irradiated sample retaining 22% of control activity. 9. These data and the catalytic role of imidazole residues at the active sites of amino acid decarboxylases are discussed.     

Die Diffusion des 59Fe-markierten Hämoglobins,ein Artefakt der Glutaraldehyd-Fixierung

Zusammenfassung Ratten wurden 50–80 c ⁵⁹Fe-Citrat in die caudale Vene injiziert. Nach 3–5tägigem Einbau des markierten Eisens in das Hämoglobin wurden Milz-Blöckchen (2 × 2 × 5 mm) 2 Std in GA vorfixiert oder in Hanks-Lösung (= Kontrolle) überführt. Ein Teil der Blöckchen wurde anschließend in OsO₄-Lösung nachfixiert.Die autoradiographischen Ergebnisse zeigen eine Diffusion des Hämoglobins vom unfixierten Zentrum zur Peripherie des Blöckchens.Aktivitätsbestimmungen, die am ganzen Blöckchen, dessen abgetrennten zentralen und peripheren Anteilen, sowie im Überstand vorgenommen wurden, bestätigen diese Diffusion. Während der OsO₄-Nachfixierung erfolgte ein weiterer Verlust des markierten Hämoglobins aus dem zentralen Teil des Blöckchens, nicht aber aus der vorfixierten Peripherie.

The diffusion of ⁵⁹Fe-labelled hemoglobin, an artefact of the fixation with glutaraldehyde

Summary Injections of 50–80 c ⁵⁹Fe-citrate into the caudale vein of rats were performed. After 3–5 days of ⁵⁹Fe incorporation into the hemoglobin the spleen was taken off, cut in small blocks (2 × 2 × 5 mm) and prefixed for 2 hours in GA or transfered into Hankssolution (= control). Later on some blocks of the spleen were postfixed in OsO₄ solution.A diffusion of the hemoglobin from the unfixed center to the peripheral tissue of the spleen-block is demonstrated by autoradiographic results.After measuring the radioactivity of the total spleen-block of the separated central and peripherical parts as well as their supernatants a diffusion was confirmed. A further loss of the labelled hemoglobin has been observed during the OsO₄ postfixation from the central part of the spleen-block, but not from the prefixed periphery.

     

A re-evaluation of the mesangial cells of the renal glomerulus

Summary The topographical localization of the mesangial cells in renal glomeruli of rats, and their relationships with the structures at the hilus of the glomerulus were studied in this investigation. It was observed that the mesangial cells occupy a parietal position in the wall of the glomerular capillaries, and that they are anatomically continuous with the smooth muscle cells of the tunica media of the glomerular arterioles.This study was supported by a United States Public Health Service Grant AM 08628 (Institute of Arthritis and Metabolic Diseases).     

THE ETIOLOGY OF SCHIZOPHRENIA—A Review

The causes and the nature of the psychiatric disorder labeled schizophrenia remain vexingly obscure. Perhaps as an expression of a still extant body-mind controversy, most of the experiments and statements made toward an elucidation of the problem follow one or the other of two opposing postulations: (a) That its origin is genetico-organic; (b) that it is environmental. In a review of the outstanding “facts” for either argument, it is notable that they presuppose not only a difference in theoretical frameworks, but two radically distinct outlooks. This is reflected in therapy, a field in which organicists and environmentalists stand even further apart; the organicist, relying heavily on electroshock and drugs, hopes to counteract a hypothetical body malfunction, and the environmentalist, through psychotherapy, attempts to make it possible for the patient to disentangle his own conflicting feelings and reaction patterns.Between the two an eclectic position seems hardly tenable. For, in spite of voluminous research and speculation, it has not been possible to build a bridge between the two camps and integrate different outlooks which, at times, have brought psychiatry almost to the point of schism.
