The carbazole ring is the basic structure present in the fluorescence derivatization reagents 9-chlorocarbonylcarbazole and 9-carbazolylacetic acid. The fluorescence behaviour of these carbazole derivatives was studied in solvents with different polarities (cyclohexane, ethanol, acetonitrile, water) and at different pH values (4.5 and 8.8). The influence of the low polarity environment afforded by 2-hydroxypropyl-beta-cyclodextrin (HPbeta-CD) is also described. The behaviour of the fluorescent reagents is compared to the model molecules carbazole and 9-methylcarbazole. For all derivatives studied, a bathochromic shift in the fluorescence emission maxima was observed when the solvent polarity was increased. A bathochromic shift was observed in dioxane solutions, which can be ascribed to the peculiar behaviour of this solvent. The changes in the fluorescence intensity in the case of 9-carbazolylacetic acid can be related to the ionization of the carboxylic acid group. Inclusion into the cavity of HPbeta-CD allows emission spectra to be obtained close to those obtained in ethanolic solutions with a remarkable enhancement in the fluorescence intensity, depending on the chemical structure of the carbazole derivative included.  相似文献

The study of fluorescence quenching of the fluorophores allows the localization of the alkaloids (harmane and harmine) in the micelles (SDS, CTAB, Brij-35) to be established. In aqueous micellar solutions (SDS and Brij-35) at pH 13.0, emission corresponding to the neutral or zwitterionic forms can be observed. In the presence of CTAB (pH = 13.0) it was possible to observe the emission of anionic form. These species are not present in buffered aqueous solutions at these pH values. Bromide ion was added to the different surfactant solutions and the quenching effect was studied according to the Stern-Volmer equation. In the presence of SDS the quenching effect is considerably reduced compared to the aqueous solutions without surfactants, while for Brij-35 micelles were similar to those observed in homogeneous aqueous solution. For CTAB micelles a notable fluorescence quenching was observed for the different pH values studied. The fluorescence quenching studies show that the neutral species are associated inside the micelles, instead of the ionic species (cationic, zwitterionic or anionic) remaining on the surface of the micelles. The anionic surface of SDS micelles prevents the quenching effect by anionic quenchers for both neutral and charged species.  相似文献

Uncovering biological diversity to more accurately understand diversity patterns, and ultimately the processes driving diversification, is important not only from an evolutionary perspective but also a conservation perspective. This is particularly pertinent in Africa's rivers in which overall diversity, as well as how it arose, is poorly understood in comparison with lacustrine environments. Here we investigate population divergence in the sexually dimorphic suckermouth catfish species Chiloglanis anoterus (Crass, 1960) from the African Highveld, in which we observe striking variability in exaggerated male caudal fins across its range. As this trait is likely to be indirect evidence for sexual selection by female choice, a mechanism that has been shown to increase species diversity in different taxa, we used an integrated approach to test if current diversity in this species is underestimated. Results based on phylogenetic inference, population genetics and geometric morphometrics indicate that the recognized species C. anoterus represents five distinct lineages that may be considered confirmed candidate species. We suggest that diversification in these highland catfish has been facilitated through geographical isolation in upper river catchments, and that sexual selection through female choice has probably driven variation in male caudal fin morphology. In contrast to the relatively large range size of the currently recognized species (C. anoterus), our findings highlight highly restricted ranges of the lineages identified here, indicating that these highland habitats may harbour higher levels of endemic diversity than previously thought.  相似文献

Ant colony optimisation (ACO) is a nature-inspired, population-based metaheuristic that has been used to solve a wide variety of computationally hard problems. In order to take full advantage of the inherently stochastic and distributed nature of the method, we describe a parallelization strategy that leverages these features on heterogeneous and large-scale, massively-parallel hardware systems. Our approach balances workload effectively, by dynamically assigning jobs to heterogeneous resources which then run ACO implementations using different search strategies. Our experimental results confirm that we can obtain significant improvements in terms of both solution quality and energy expenditure, thus opening up new possibilities for the development of metaheuristic-based solutions to “real world” problems on high-performance, energy-efficient contemporary heterogeneous computing platforms.  相似文献

The reaction center-light harvesting complex 1 (RC-LH1) purified from the photosynthetic bacterium Rhodobacter sphaeroides has been studied with respect to the kinetics of charge recombination and to the phospholipid and ubiquinone (UQ) complements tightly associated with it. In the antenna-RC complexes, at 6.5 more than three times smaller than that measured in LH1-deprived RCs. At increasing pH values, for which increases, the deceleration observed in RC-LH1 complexes is reduced, vanishing at pH >11.0. In both systems kinetics are described by a continuous rate distribution, which broadens at pH >9.5, revealing a strong kinetic heterogeneity, more pronounced in the RC-LH1 complex. In the presence of the antenna the Q(A)Q(B)(-) state is stabilized by about 40 meV at 6.511. The phospholipid/RC and UQ/RC ratios have been compared in chromatophore membranes, in RC-LH1 complexes and in the isolated peripheral antenna (LH2). The UQ concentration in the lipid phase of the RC-LH1 complexes is about one order of magnitude larger than the average concentration in chromatophores and in LH2 complexes. Following detergent washing RC-LH1 complexes retain 80-90 phospholipid and 10-15 ubiquinone molecules per monomer. The fractional composition of the lipid domain tightly bound to the RC-LH1 (determined by TLC and (31)P-NMR) differs markedly from that of chromatophores and of the peripheral antenna. The content of cardiolipin, close to 10% weight in chromatophores and LH2 complexes, becomes dominant in the RC-LH1 complexes. We propose that the quinone and cardiolipin confinement observed in core complexes reflects the in vivo heterogeneous distributions of these components. Stabilization of the charge separated state in the RC-LH1 complexes is tentatively ascribed to local electrostatic perturbations due to cardiolipin.  相似文献

This qualitative empirical study explores the articulation of meanings about difference, belonging and positionality that are emerging in English talk across transcultural boundaries in Naples, Southern Italy. It shows that the fraught encounters that take place on street markets, on public transport and at community events lead both to tactics of racialized closure, exclusion and division; as well as to the formation of innovative and ambivalent convivialities. In stressing the importance of talking English in intersubjective interactions it not only tells a story about the particular context and history of race relations in the city – where different sorts of speaking are central to a history of internal subordination and mass emigration – but also offers new ways of thinking about the complex and ambiguous multilingual reality that has resulted from intensified migration across the world.  相似文献
