The bacterial secretion of glutamate was studied through plasma membrane fluidity, measured by anisotropy using the fluorophore TMA-DPH incorporated in the lipid part of the cell membrane. Cells of Brevibacterium sp. ATCC 13869 (wild type) were switched from the biotin-limited, producing state to the biotin-supplemented, non-producing state, and back. The following conclusions could be drawn: 1. It was not possible to detect any change in anisotropy by switching the cells from biotin-limited biotin-supplemented, as well as from biotin-supplemented, to biotin-limited, media. 2. The anisotropy value in the glutamic acid fermentation remains constant during the lag, exponential, growth, production and stationary phases. 3. The treatment of cells with a neutral synthetic polyester of ethylene-and propyleneoxide with soya oil-fatty acids increased the anisotropy values, indicating incorporation of the surfactant. 4. Glutamate secretion is not coupled with membrane fluidity, so a leak providing a general fluidization of the membrane could not be detected. 相似文献

In a study with rats it was determined whether dietary magnesium concentration affects plasma esterase activities. The feeding of a diet with 0.01% (w/w) instead of 0.04% magnesium reduced plasma magnesium concentration by 50%. Plasma total esterase, arylesterase and butyrylcholinesterase activities were significantly decreased in the magnesium-deficient rats. In rats fed a diet containing 0.02% magnesium, plasma magnesium concentration was lowered by 30%, and group mean plasma total esterase activity was decreased, but not the activities of arylesterase and butyrylcholinesterase. 相似文献

Fertilized stands of Pseudotsuga menziesii were found to have glaucous needles. We investigated the morphological and quantitative characteristics of the epicuticular waxes of needles of fertilized and control trees. Glaucousness was caused by ornate tubular epicuticular wax. Dipping needles in chloroform, which dissolves waxes, eliminated the glaucous appearance. Based on cryostage scanning electron microscopic observations, the epicuticular waxes in the nonstomatal region were much more ornate on the needles of the fertilized trees (experimental needles) than in unfertilized trees (control needles). The stomatal region in both experimental and control needles showed similarly ornate waxes. Quantities of waxes were similar in experimental and control needles. The glaucousness was not the result of greater quantities of wax; rather, fertilization altered wax morphology in the nonstomatal regions. 相似文献

Because of the different mechanical constraints on vines and self-supporting plants, vines are thought to differ from trees and shrubs in a variety of their growth characteristics. I tested the hypotheses that vines grow faster than shrubs and that supported shoots have delayed leaf expansion relative to stem elongation, using western poison oak, Toxicodendron diversilobum (T. & G.) Greene, a plant that grows as a vine when externally supported but otherwise as a shrub. In the field, supported shoots (vines) had significantly higher aboveground biomass and relative growth rates than did their paired unsupported shoots (shrubs) growing nearby. This was not due to differences in leaf phenology, but may have resulted from vines growing into more favorable habitats for growth. In contrast, whereas 2-yr-old cloned plants in a common garden differed in their stem and internode lengths, they had the same aboveground dry weight, proportion of dry weight that was leaf, and relative rate of increase in primary stem length whether grown with stakes (vines) or without stakes (shrubs). These results suggest that there is no inherent requirement of vines to grow faster than shrubs. As hypothesized, leaf elongation was more delayed relative to stem elongation in staked than unstaked individuals in 19 paired plants (each pair cloned from a different source plant). Thus, physical cues resulting from the presence of support can alter the plant's spatial and temporal patterns of development, but do not necessarily dictate the quantity of biomass that will be produced. 相似文献

Extended field studies in Peru and extensive investigation of herbarium material revealed the urgent necessity for a revision. As a result, the number of species is reduced to 12, among them two species are newly described. 9 species are members of an other subgenus. Summary Slot- and dot-blotting are commonly used to evaluate levels of messenger ribonucleic acid (mRNA). Quantitation of bacterially-expressed chloramphenicol acetyl transferase (CAT) mRNA by this method is highly dependent on total RNA immobilised onto the solid support as well as mRNA concentration. mRNA quantitation by comparison with a pure standard results in underestimation. We report an improved method for white clover (Trifolium repens) transformation usingAgrobacterium tumefaciens. High efficiencies of transgenic plant production were achieved using cotyledons of imbibed mature seed. Transgenic plants were recovered routinely from over 50% of treated cotyledons. Thebar gene and phosphinothricin selection was shown to be a more effective selection system thannptII (kanamycin selection) oraadA (spectinomycin selection). White clover was transformed with the soybean auxin responsive promoter, GH3, fused to the GUS gene (-glucuronidase) to study the involvement of auxin in root development. Analysis of 12 independent transgenic plants showed that the location and pattern of GUS expression was consistent but the levels of expression varied. The level of GH3:GUS expression in untreated plants was enhanced specifically by auxin-treatment but the pattern of expression was not altered. Expression of the GH3:GUS fusion was not enhanced by other phytohormones. A consistent GUS expression pattern was evident in untreated plants presumably in response to endogenous auxin or to differences in auxin sensitivity in various clover tissues. In untreated plants, the pattern of GH3:GUS expression was consistent with physiological responses which are regarded as being auxin-mediated. For the first time it is shown that localised spots of GH3:GUS activity occurred in root cortical tissue opposite the sites where lateral roots subsequently were initiated. Newly formed lateral roots grew towards and through these islands of GH3:GUS expression, implying the importance of auxin in controlling lateral root development. Similarly, it is demonstrated for the first time that gravistimulated roots developed a rapid (within 1 h) induction of GH3:GUS activity in tissues on the non-elongating side of the responding root and this induction occurred concurrently with root curvature. These transgenic plants could be useful tools in determining the physiological and biochemical changes that occur during auxin-mediated responses. 相似文献

In July 1992, in the Austrian part of Hanság, a seventy day old young bustard was found dead in a grassland. On the left intertarsal joint a walnut sized open pock was located. Other pocks reaching pea-size were found on both legs. The diagnosis pox was established by light- and electron-microscopic examination of the lesions. A further chick of another hen, fledged in the same year, observed from a distance showed abnormal thickening of the intertarsal joint area. The consequences of pox for such a small group of Great Bustards (total for 1988–1993 15–20 birds) should be watched carefully. 相似文献
