Summary The oncogenic plasmids of Agrobacterium, the Ti-plasmids, carry genes that enable their bacterial host to catabolize opines. Opines are unusual amino acid derivatives that are only produced in crown gall tumours incited by oncogenic strains of Agrobacterium. The 2 opines, octopine and nopaline, are degraded by Agrobacterium strains carrying the octopine or the nopoline Ti-plasmid, respectively, to arginine and pyruvic acid, and to arginine and -ketoglutaric acid. In this paper it is shown that the Ti-plasmids carry gene(s) involved in the utilisation of arginine as a carbon source. Strains harbouring wild type octopine or nopaline Ti-plasmids in the chromosomal context of strain C58C1 do not grow on arginine as a carbon source. However, they are able to grow on arginine provided that they are induced, or constitutive for opine catabolism. The features of ornithine utilisation are identical. The gene(s) involved in arginine and ornithine utilization in C58C1 (pTi-oct) or C58C1 (pTi-nop) are under the control of the regulator gene that controls octopine or nopaline catabolism. A tentative pathway of octopine utilization is proposed, in which at least two steps are Ti-plasmid coded, and probably belong to the same operon: 1-scission of octopine into arginine and pyruvic acid 2-transformation of an arginine derivative (GSA?) to glutamic acid.Arginine utilization as a carbon source is therefore a new function of the Ti-plasmid. As this function is not inducible by arginine but by opines, it provides a method for selecting regulatory mutants of opine catabolism in the genetic background of strain C58.  相似文献

Elevated plasma lipoprotein(a) (Lp(a)) is an independent, causal risk factor for atherosclerotic cardiovascular disease and calcific aortic valve stenosis. Lp(a) is formed in or on hepatocytes from successive noncovalent and covalent interactions between apo(a) and apoB, although the subcellular location of these interactions and the nature of the apoB-containing particle involved remain unclear. Sortilin, encoded by the SORT1 gene, modulates apoB secretion and LDL clearance. We used a HepG2 cell model to study the secretion kinetics of apo(a) and apoB. Overexpression of sortilin increased apo(a) secretion, while siRNA-mediated knockdown of sortilin expression correspondingly decreased apo(a) secretion. Sortilin binds LDL but not apo(a) or Lp(a), indicating that its effect on apo(a) secretion is likely indirect. Indeed, the effect was dependent on the ability of apo(a) to interact noncovalently with apoB. Overexpression of sortilin enhanced internalization of Lp(a), but not apo(a), by HepG2 cells, although neither sortilin knockdown in these cells or Sort1 deficiency in mice impacted Lp(a) uptake. We found several missense mutations in SORT1 in patients with extremely high Lp(a) levels; sortilin containing some of these mutations was more effective at promoting apo(a) secretion than WT sortilin, though no differences were found with respect to Lp(a) internalization. Our observations suggest that sortilin could play a role in determining plasma Lp(a) levels and corroborate in vivo human kinetic studies which imply that secretion of apo(a) and apoB are coupled, likely within the hepatocyte.  相似文献

  • 1 The North African–Asian lion subgroup, which is composed of two subspecies, the Barbary lion, Panthera leo leo, and the Asian lion, P. l. persica, was nearly exterminated during the last centuries. The remaining free‐ranging population of Asian lions consists of c. 350 animals in India. The Barbary lion subsists as captive animals in zoos or circuses, all of which originate from the Moroccan Royal Collection. There have been multiple genetic hybridizations with sub‐Saharan lions.
  • 2 Several incomplete distribution maps of the past occupation of lions were proposed in the literature. I examine the distribution of these lions with the help of a larger database, extracted from zoological, archaeological and historical reports. Some records are however not completely reliable. Data were collected over a long period of time (from the Epipaleolithic to the modern times) in Eurasia, North Africa and the Arabian Peninsula.
  • 3 Dispersal took place in latitudes as far south as 15°N (Yemen) to 18°N (Mali; Chad) and as far north as 45–48°N (Bulgaria); and to longitudes as far west as 5.57°W (Morocco) and as far east as 84°E (India). Expansion was probably constrained by natural ecological factors northwards (higher seasonality, harsher winters) and southwards (extension of aridity after 3000 BP). At latitudes between 40 and 43°N, lions seem to have become a permanent part of the fauna during 6–8 millennia.
  • 4 Lions were brought to extinction by humans through hunting, captures for exhibition in zoos and by the associated natural fragmentation of wild populations. The dates at which extinction took place varied greatly according to the geographical sites: 3000 BC for temperate Europe; 1000 BC in the south of Greece; 12–13th century in the Near East, Arabian Peninsula, Trans‐Caucasia or north of Afghanistan; and 19–20th century for North Africa, the Middle East and India.

In a previous study, we demonstrated that transgenic Lotus plants producing opines (which are small amino acid and sugar conjugates) specifically favor growth of opine-degrading rhizobacteria. The opine-induced bias was repeated and demonstrated with another soil type and another plant species (Solanum nigrum). The use of local anesthetics, such as lidocaine, has been proposed in the treatment of distal ulcerative colitis. Trimebutine maleate (TMB) displays a local anesthetic activity higher than that of lidocaine in rabbit corneal reflex. TMB and nor-TMB its main metabolite in human show similar affinity to that of bupivacaine toward sodium channel labeled by [3H]batrachotoxin and block sodium currents in sensory neurons from rat dorsal root ganglia. The aim of this study was to evaluate the effects of TMB and nor-TMB in comparison to lidocaine and bupivacaine in a rat model of acute colonic inflammation induced by trinitrobenzene sulfonic acid (TNBS). A single intracolonic instillation of TNBS (50 mg/kg dissolved in ethanol 30%) led to early plasma extravasation then macroscopic damage (hyperemia and necrosis), increased colonic weight and tissular MPO, a marker of neutrophilic infiltration. Local administration of TMB at dose of 3 to 60 mg/kg, 30 min before, 24 and 48 h after colitis induction, significantly reduced the severity of colitis. Nor-TMB (1, 3, 10, 30 mg/kg) as well as lidocaine (1, 3, 10 mg/kg) dose-dependently reduced colitis while bupivacaine at 10 mg/kg did not affect it significantly. In contrast systemic administration of TMB, nor-TMB and lidocaine at 10 mg/kg had no significant effect. Furthermore, local administration of TMB (30 mg/kg) and lidocaine (10 mg/kg) significantly reduced plasmatic extravasation. In conclusion, intracolonic treatment with TMB and nor-TMB improved acute experimental TNBS-induced colitis in rat and these effects could be explained by their local anesthetic activity.  相似文献

In this study, we investigate the mechanisms driving biodiversity in floodplain forests with a comparison of the composition and dynamics in the warm-temperate floodplain forests of the lower Mississippi Valley and the cool-temperate floodplain forests of the lower Wisconsin and Rhine River Valleys. We employ data from original research, as well as from the literature. We compare species, genus, and family diversity across regions with respect to species richness, numbers of species per family and genus, and a similarity index. We examine these results within a historical context, as well as with respect to river-floodplain dynamics. We also compare productivity data and successional stages for each region. We find a lower species, genus, and family richness in the cool-temperate forests of the Rhine compared to the cool-temperate forests of the Wisconsin, a probable result of the lack of available refugia for Rhine species in times of glacial expansion. We find the highest richness in the lower Mississippi Valley, likely a result of climatic factors and the availability of refugia in this region. In each of the regions, floodplain forests are more diverse than their upland counterparts, demonstrating the role of river-floodplain dynamics in maintaining species diversity. Each region maintains a high and relatively similar level of productivity in the floodplain forests. They also experience similar stages of succession, although succession becomes more complex in the warm-temperate forests of the Lower Mississippi.  相似文献

Traditional measures of biotic indices (, , ) from phytosociological relevés are used to compare diversity at three scale-levels in the upper Rhine valley (Alsace, northeastern region) and the middle Loire (Bourgogne, central region), France. Both study areas included a tributary. The Ill, which is the Rhine tributary in this sector, has no forest communities in common with the main river because of opposite hydrological and edaphic characteristics. This is not the case in the Loire valley, in which the fluvial characteristics are close to those of its tributary, the Allier.Nearly all forest communities (12/14) were shown to be species-rich, due to the role of natural disturbances. Species richness peaks in late-successional forests of the Rhine valley (for example, up to 50 woody species in Querco-Ulmetum). Sixty per cent of the forest-communities are comon to the two floodplains, which explains why 85% of the families, 65% of the genera and 55% of the species are in common. Compared to the Loire valley, the Rhine valleys shows higher biotic diversity. This was related first to its geographical situation, and second to higher landscape heterogeneity. Human disturbance through river management is responsible for changes in species composition as shown by floristic comparison of flooded and unflooded sites. Insularity of natural forests among man-made landscapes is involved in species diversity. In order to preserve the natural diversity of such rich landscapes, floodable areas should be recreated and the attitudes of foresters should evolve accordingly.  相似文献
