Detection of a protein encoded by a class II mitochondrial intron of Podospora anserina

Summary In the filamentous fungus Podospora anserina, the amplification as circular DNA molecules of the first intron (intron ) of the CO1 mitochondrial gene, encoding the cytochrome oxidase subunit 1, is known to be strongly associated with aging of strains. In this study we have attempted to detect the protein potentially encoded by the open reading frame (ORF) contained in this intron. This was done by the Western blot technique using specific antisera raised against three polypeptides encoded by three non-overlapping fragments of this ORF adapted to the universal code and overexpressed in Escherichia coli. We examined about thirty independent subclones of Podospora derived from two different geographic races (A, s), using wild-type and mutant strains, young and senescent cultures. A 100 kDa polypeptide, encoded by the class II intron , was detected in five senescent subclones which all showed strong amplification of the intronic sequence (Sen DNA ).     

Effect of mifepristone (RU 486) on concentrations of prostaglandin E-2 binding sites in the rat endometrium

Progesterone implants in ovariectomized rats increased endometrial concentrations of PGE-2 receptors. The increase was completely inhibited by simultaneous daily injection (7.5 mg/kg) of mifepristone (RU 486). A single injection of mifepristone on the morning of Day 1 of pseudopregnancy (day of oestrus) decreased the amount of PGE-2 receptors found in the endometrium on Day 5 by 64%. This inhibitory effect probably resulted from the antiprogesterone activity of this compound since it was not counteracted by simultaneous treatment with dexamethasone, shown to reverse totally the antiglucocorticoid action of mifepristone. The inhibition by mifepristone lasted only for 1 day; endometrial PGE-2 receptor levels on Day 6 of pseudopregnancy returned to the high values present in controls. Under these conditions, administration of the mifepristone did not affect the plasma oestradiol and progesterone concentrations during the 1st week of pseudopregnancy. The administration of mifepristone on Days 2 and 3 of pseudopregnancy kept the endometrial PGE-2 receptor levels low, even by 4 days after the end of treatment. We therefore concluded that, in the rat, progesterone priming leading to uterine receptivity can be delayed, at least by 1 day. In contrast, interruption of the progesterone action for a longer period later during the early pseudopregnant period resulted in an altered subsequent evolution of the endometrium, in terms of acquisition of the PGE-2 binding sites.     

Deposition of type and voucher material from the helminthological collection of Elon E. Byrd

Elon E. Byrd was a prominent helminth taxonomist who published between 1930–1965. After his death in 1974, his collection was put in storage at the University of Georgia School of Veterinary Medicine. A recent examination of his collection yielded a number of taxonomically valuable specimens that should be available to research workers worldwide. This paper enumerates the specimens recovered and their deposition. Digenean families and genera represented are: Brachycoeliidae (Brachycoelium), Dicrocoeliidae (Paradistomum), Lecithodendriidae (Prosthodendrium, Pseudosonsinotrema), Microphallidae (Levinseniella), Ochetosomatidae (Dasymetra, Pneumatophilus, Renifer, Neorenifer), Plagiorchiidae (Leptophyllum, Paurophyllum, Stomatrema, Styphlodora), Proterodiplostomidae (Pseudoneodiplostomum, Pseudocrocodilicola), Spirorchiidae (Spirorchis, Henotosoma, Vasotrema, Unicaecum, Hapalorhynchus), Telorchiidae (Cercorchis); the Nematoda are represented by the Diaphanocephalidae (Kalicephalus). No attempt was made to determine current generic status or synonymies.     

Production of a membrane-bound proteins,the human gamma-glutamyl transferase,by CHO cells cultivated on microcarriers,in aggregates and in suspension

Recombinant Chinese Hamster Ovary (CHO) cells, engineered for the production of human gamma-glutamyl transferase (GGT), have been grown on Cytodex 1 microcarriers, as aggregates, or as single cells in suspension after adaptation. GGT is a membrane bound enzyme which was not secreted during the culture period. The maximal enzyme activity was found to be directly related to the achieved maximal cell density. Culture of CHO on microcarriers yielded the fastest growth, with a specific growth rate of 0.04 h^–1, the highest cell density (near 1.3×10⁶ cells ml^–1), and the highest enzyme activity around 300 mU ml^–1, which corresponded to a specific cellular level of 20 mU 10^–5 cells. GGT could also be produced by growing CHO cells in suspension as single cells or as aggregates. Under these conditions, however, the specific CHO growth rate was significantly slower and the GGT level per cell was divided by a factor 6. Growing CHO cells without microcarriers also resulted in differences in cell metabolism, with a higher conversion yield of glutamine into ammonia, and a higher cell lysis. The catalytic kinetic constants of the enzyme were found identical for the three culture systems.     

Heterogeneity of twoBeauveria bassiana strains revealed by biochemical tests,protein profiles and bio-assays ofLeptinotarsa decemlineata (Col.: Chrysomelidae) andColeomegilla macultata lengi (Col.: Coccinellidae) larvae

Biochemical profiles on API Rapid CH^* strips and protein profiles on polyacrylamide gels in the presence of sodium dodecyl sulfate were used to distinguish two strains of the entomopathogenic fungusBeauveria bassiana (Balsamo) Vuillemin, ARSEF 2991 and ATCC 44860. Next, the toxicity of these two strains was determined at concentrations of 10², 10⁴, 10⁶ and 10⁸ blastospores/ml on larvae of the Colorado potato beetleLeptinotarsa decemlineata Say (Coleoptera: Chrysomelidae) and of its predator, the spotted ladybird beetle,Coleomegilla maculata lengi Timberlake (Coleoptera: Coccinellidae). Both strains were highly toxic toL. decemlineata larvae. However, the two strains exhibited different levels of toxicity forC. maculata larvae: ARSEF 2991 was toxic, whereas ATCC 44860 caused little coccinellid larval mortality.

Résumé Les profils biochimiques sur galeries API Rapid CH^* et les profils protéiques sur gels de polyacrylamide ont été utilisés pour distinguer deux souches du champignon entomopathogèneBeauveria bassiana (Balsamo) Vuillemin. La toxicité de ces deux souches a été déterminée à des concentrations de 10², 10⁴, 10⁶ et 10⁸ blastospores/ml sur des larves du doryphore,Leptinotarsa decemlineata Say (Coleoptera: Chrysomelidae) et de la coccinelle maculéeColeomegilla maculata lengi Timberlake (Coleoptera: Coccinellidae). Les deux souches deB. bassiana se sont avérées actives à l'égard des larves deL. decemlineata. Toutefois la souche ARSEF 2991 s'est avérée pathogène pour les larves deC. maculata, alors que la souche ATCC 44860 a provoqué une faible mortalité des larves.

     

IN VIVO EFFECTS OF CARDIOTROPHIN-1

Cardiotrophin-1 (CT-1) is a recently discovered cytokine that was isolated based on its ability to induce cardiac myocyte hypertrophy in vitro. In this study, the effects of chronic administration of CT-1 to mice (0.5 or 2 μg by intraperitoneal injection, twice a day for 14 days) were determined. A dose-dependent increase in both the heart weight and ventricular weight to body ratios was observed in the treated groups. The body weights of the animals were unaffected. These results indicate that CT-1 can induce cardiac hypertrophy in vivo. CT-1 was not specific for the heart, however. It stimulated the growth of the liver, kidney, and spleen, and caused atrophy of the thymus. CT-1 administration also increased the platelet counts by 70%, with no change in mean platelet volume. Red blood cell counts were increased in the treated animals, and there was a concomitant increase in haemoglobin concentration. Thus, CT-1 has a broad spectrum of biological activities in vivo. This observation is consistent with previous in-vitro findings showing that the mRNA for CT-1 is expressed in several tissues, and that CT-1 can function through binding to the leukaemia inhibitory factor (LIF) receptor and signalling through the gp130 pathway.     

Continuous hybridoma culture in a low-protein serum-free medium supplemented with liposomes

A homemade serum-free medium containing a low protein level under 0.1 g l⁻¹ has been proved to support long-term cultures of VO 208 hybridoma cells successfully up to 50 days. The low protein level was achieved by supplying the lipids through liposomes containing cholesterol, oleic acid, - dipalmitoyl phosphatidylcholine, and bovine serum albumin. The influence of the liposome content in the feeding medium was studied in a continuous culture performed with step variations of the liposomes level, from 7.5 to 30 ml l⁻¹. The cell density decreased at the highest liposomes content while it became higher with 7.5 or 12 ml l⁻¹ of liposomes. For each step variation appeared a transitory activation of the specific rates of nutrient consumption, metabolite production and antibody secretion, as well as a transitory decrease of the specific cell growth rate. The overall structure of the antibodies was not affected during the culture.     

The Presence and Role of Transmembrane Transforming Growth Factor-α in Cultures of Rat Liver Epithelial Cells

We assessed the presence and the role of membrane TGF-α in two rat liver epithelial cell lines, either parental or transfected with c-fos proto-oncogene. c-fos overexpressing cells had more TGF-α-like activity in their membranes. When TGF-α was removed by elastase or neutralized, the growth rates of both cell lines were markedly reduced, but to a higher extent for parental cells. If membrane TGF-α seemed to play a key contribution in normal cell growth, both cell lines were unable to react to the addition of soluble TGF-α, showing that these two forms of growth factors are not equivalent.
