Aim The analysis of the phylogeographical structures of many European species reveals the importance of Mediterranean glacial refugia for many thermophilic species, but also underlines the relevance of extra-Mediterranean glacial differentiation centres for a number of temperate species. In this context, phylogeographical analyses of species from south-eastern Europe are highly important for a comprehensive understanding of Europe as a whole.
Location Romania and Bulgaria.
Methods We analysed 19 allozyme loci for 615 individuals of the temperate butterfly species Erebia medusa from 28 populations.
Results These populations had an intermediate genetic diversity, but the Bulgarian populations were significantly more diverse than the ones north of the Danube in Romania. The differentiation among populations was strong, and 52.1% of the genetic variance among populations was distributed between these two countries. The genetic differentiation was considerably stronger in Romania than in Bulgaria, but several sublineages were distinguished within each of these countries.
Main conclusions The observed genetic structure is so strong that it is most probably the result of glacial differentiation processes in south-eastern Europe and not a post-glacial structure. The strong differentiation into the two groups north and south of the Danube suggests a separating effect by this river valley. The strong differentiation accompanied with genetic impoverishment in Romania suggests the existence of several differentiation centres: at least two small ones on the southern slopes of the southern Carpathians and one in the eastern Carpathian Basin. The considerably weaker differentiation among the Bulgarian samples and their significantly higher genetic diversity imply that gene flow occurred among different regions of Bulgaria during the last ice age. 相似文献

Six different pyruvate decarboxylase mutants of Saccharomyces cerevisiae were isolated. They belong to two unlinked complementation groups. Evidence is presented that one group is affected in a structural gene. The fact that five of the six mutants had residual pyruvate decarboxylase activity provided the opportunity for an intensive physiological characterization. It was shown that the loss of enzyme activity in vitro is reflected in a lower fermentation rate, an increased pyruvate secretion, and slower growth on a 2% glucose medium. The different effects of antimycin A on leaky mutants grown on ethanol versus the same mutants grown on glucose support the view that glucose induces some of the glycolytic enzymes, especially pyruvate decarboxylase. 相似文献

The bacterial paromomycin resistance gene, aphH, as a dominant selectable marker in Volvox carteri 总被引:1,自引:0,他引:1
The aminoglycoside antibiotic paromomycin that is highly toxic to the green alga Volvox carteri is efficiently inactivated by aminoglycoside 3′-phosphotransferase from Streptomyces rimosus. Therefore, we made constructs in which the bacterial aphH gene encoding this enzyme was combined with Volvox cis-regulatory elements in an attempt to develop a new dominant selectable marker – paromomycin resistance (PmR) – for use in Volvox nuclear transformation. The construct that provided the most efficient transformation was one in which aphH was placed between a chimeric promoter that was generated by fusing the Volvox hsp70 and rbcS3 promoters and the 3′ UTR of the Volvox rbcS3 gene. When this plasmid was used in combination with a high-impact biolistic device, the frequency of stable PmR transformants ranged about 15 per 106 target cells. Due to rapid and sharp selection, PmR transformants were readily isolated after six days, which is half the time required for previously used markers. Co-transformation of an unselected marker ranged about 30%. The chimeric aphH gene was stably integrated into the Volvox genome, frequently as tandem multiple copies, and was expressed at a level that made selection of PmR transformants simple and unambiguous. This makes the engineered bacterial aphH gene an efficient dominant selection marker for the transformation and co-transformation of a broad range of V. carteri strains without the recurring need for using auxotrophic recipient strains. 相似文献

Arabidopsis thaliana has seven genes for functionally active sucrose transporters. Together with sucrose transporters from other dicot and monocot plants, these proteins form four separate phylogenetic groups. Group-IV includes the Arabidopsis protein SUC4 (synonym SUT4) and related proteins from monocots and dicots. These Group-IV sucrose transporters were reported to be either tonoplast- or plasma membrane-localised, and in heterologous expression systems were shown to act as sucrose/H(+) symporters. Here, we present comparative analyses of the subcellular localisation of the Arabidopsis SUC4 protein and of several other Group-IV sucrose transporters, studies on tissue specificity of the Arabidopsis SUC4 promoter, phenotypic characterisations of Atsuc4.1 mutants and AtSUC4 overexpressing (AtSUC4-OX) plants, and functional comparisons of Atsuc4.1 and AtSUC4-OX vacuoles. Our data show that SUC4-type sucrose transporters from different plant families (Brassicaceae, Cucurbitaceae and Solanaceae) localise exclusively to the tonoplast, demonstrating that vacuolar sucrose transport is a common theme of all SUC4-type proteins. AtSUC4 expression is confined to the stele of Arabidopsis roots, developing anthers and meristematic tissues in all aerial parts. Analyses of the carbohydrate content of WT and mutant seedlings revealed reduced sucrose content in AtSUC4-OX seedlings. This is in line with patch-clamp analyses of AtSUC4-OX vacuoles that characterise AtSUC4 as a sucrose/H(+) symporter directly in the tonoplast membrane. 相似文献

Understanding the ecological process of population differentiation and identifying the molecular changes that contribute to adaptation is a central aspect of evolutionary biology. In this study we analyzed the geographic variation in allozyme allele frequencies (based on 15 enzyme systems representing 18 loci) across 18 populations of the butterfly Lycaena tityrus from different altitudes. Population genetic analyses showed that within population genetic diversity (e.g. mean number of alleles per loci: 1.8; expected heterozygosity: 12%) was within the typical value range for the Lepidoptera. The populations of L. tityrus investigated showed a remarkable genetic differentiation (FST : 0.065), being clearly divided into an alpine (high-altitude) and a non-alpine (low-altitude) cluster. This differentiation was almost entirely caused by variation at a single enzyme locus, PGI. Although the involvement of historical events cannot be ruled out, several lines of evidence suggest that the specific pattern of allozyme (and in this case particularly PGI) variation found is caused by thermal selection. For example, genetic variation was highly locus-specific rather than relatively uniform, as should be expected for effects of natural selection. Further, the PGI 2–2 genotype dominating in alpine (in contrast to low-altitude) populations is known to exhibit increased cold stress resistance and relatively long development times typical of alpine populations. Thus, PGI (or possibly a closely linked gene) is an obvious target for thermal selection in L. tityrus . The aim of this study was to test the method Kunos and his colleagues (1999) developed on the first rib to assess adult age-at-death. The method was applied on a sample of known age and sex, selected from a Thai collection. The procedure being subjective, we chose to estimate an age category rather than a precise age.
The results show that only 55% of the individuals are correctly classified and that the number of subjects over 60 years old is under-estimated. Three explanations are given: the modifications of the first rib related with ageing are more variable than previously stated, the technique is too subjective and difficult to apply and the morphological changes of the first rib are different between North-American and Thai people. 相似文献
