The virilis group species Drosophila montana is widely distributed around the northern hemisphere. Here we show that it consists of at least two well differentiated populations (Finnish and North-American populations) that have been diverging for the last 0.55–0.95 My. These populations show significant chromosomal, behavioural and morphological differences, but no apparent postzygotic isolation. Evidence for introgression is found for both Finnish and North-American populations at two out of the three X-linked genes (fused, elav and su(s)) studied here. In the light of these findings, previously reported evidence for selective sweeps in D. montana populations is re-evaluated.  相似文献

Homozygosity was induced in transgenic barley by microspore culture. Spikes of transgenic barley plants carrying microspores in the late uni-nucleate stage were cold pretreated. Teflon rod maceration and a density of 100 000 viable micropores per plate were used. The developed calli were regenerated and plantlets were treated with colchicine. The microspore culture of 16 mother plants (three transgenic lines) resulted in 927 green regenerants. Of these plants, 476 were transferred to soil, 380 were transgenic, 358 reached maturity and 350 were fertile with a normal seed-set carrying a yield of 6.9 kg. A production efficiency of 0.8 fertile transgenic doubled haploid barley plants per spike used for microspore isolation was recorded. The produced transgenic seeds were used in malting experiments.  相似文献

The males of most species of the HawaiianDrosophila planitibia group produce songs when vibrating their wings during courtship. In four of the most recently evolved species,D. differens, D. planitibia, D. heteroneura, andD. silvestris, these songs are simple in structure and possess a higher and more variable carrier frequency than the songs of the more ancestral species of the group. In the present paper, we studied the variation in wing song production and song characters in aD. planitibia population. Some males vibrated their wings at a very low amplitude or slow speed, producing no detectable sound. Other males produced sound bursts varying in carrier frequency and burst length. The carrier frequency of the song depended mainly on the wing posture of the male during wing vibrations and was consistent for individual males. Variation between males of different isofemale progenies was not significant in any measured song trait. Songs of the males recorded in the present study differed, however, from songs of the males of a laboratory strain recorded earlier.  相似文献

Physiol. 82(3): 760-764, 1997.We analyzednitrate, a major stable end product of nitric oxide (NO) metabolism invivo in plasma and urine from groups of healthy subjects with differentworking capacities. Resting plasma nitrate was higher in athleticsubjects than in nonathletic controls [45 ± 2 vs. 34 ± 2 (SE) µM; P < 0.01]. In other subjects, both the resting plasma nitrate level(r = 0.53; P < 0.01) and the urinary excretionof nitrate at rest (r = 0.46; P < 0.01) correlated to thesubjects' peak work rates, as determined by bicycle ergometry. Twohours of physical exercise elevated plasma nitrate by 18 ± 4 (P < 0.01) and 16 ± 6%(P < 0.01), respectively, in athletes and nonathletes, compared with resting nitrate before exercise. We conclude that physical fitness and formation of NO at restare positively linked to each other. Furthermore, a single session ofexercise elicits an acute elevation of NO formation. The observedpositive relation between physical exercise and NO formation may helpto explain the beneficial effects of physical exercise oncardiovascular health.

Humans are protected against infection from most African trypanosomes by lipoprotein complexes present in serum that contain the trypanolytic pore-forming protein, Apolipoprotein L1 (APOL1). The human-infective trypanosomes, Trypanosoma brucei rhodesiense in East Africa and T. b. gambiense in West Africa have separately evolved mechanisms that allow them to resist APOL1-mediated lysis and cause human African trypanosomiasis, or sleeping sickness, in man. Recently, APOL1 variants were identified from a subset of Old World monkeys, that are able to lyse East African T. b. rhodesiense, by virtue of C-terminal polymorphisms in the APOL1 protein that hinder that parasite’s resistance mechanism. Such variants have been proposed as candidates for developing therapeutic alternatives to the unsatisfactory anti-trypanosomal drugs currently in use. Here we demonstrate the in vitro lytic ability of serum and purified recombinant protein of an APOL1 ortholog from the West African Guinea baboon (Papio papio), which is able to lyse examples of all sub-species of T. brucei including T. b. gambiense group 1 parasites, the most common agent of human African trypanosomiasis. The identification of a variant of APOL1 with trypanolytic ability for both human-infective T. brucei sub-species could be a candidate for universal APOL1-based therapeutic strategies, targeted against all pathogenic African trypanosomes.  相似文献

The antibiotic drug fusidic acid (FA) is commonly used in the clinic against gram-positive bacterial infections. FA targets ribosome-bound elongation factor G (EF-G), a translational GTPase that accelerates both messenger RNA (mRNA) translocation and ribosome recycling. How FA inhibits translocation was recently clarified, but FA inhibition of ribosome recycling by EF-G and ribosome recycling factor (RRF) has remained obscure. Here we use fast kinetics techniques to estimate mean times of ribosome splitting and the stoichiometry of GTP hydrolysis by EF-G at varying concentrations of FA, EF-G and RRF. These mean times together with previous data on uninhibited ribosome recycling were used to clarify the mechanism of FA inhibition of ribosome splitting. The biochemical data on FA inhibition of translocation and recycling were used to model the growth inhibitory effect of FA on bacterial populations. We conclude that FA inhibition of translocation provides the dominant cause of bacterial growth reduction, but that FA inhibition of ribosome recycling may contribute significantly to FA-induced expression of short regulatory open reading frames, like those involved in FA resistance.  相似文献

Purification is a bottleneck and a major cost factor in the production of antibodies. We set out to engineer a bifunctional fusion protein from two building blocks, Protein A and a hydrophobin, aiming at low‐cost and scalable antibody capturing in solutions. Immunoglobulin‐binding Protein A is widely used in affinity‐based purification. The hydrophobin fusion tag, on the other hand, has been shown to enable purification by two‐phase separation. Protein A was fused to two different hydrophobin tags, HFBI or II, and expressed transiently in Nicotiana benthamiana. The hydrophobins enhanced accumulation up to 35‐fold, yielding up to 25% of total soluble protein. Both fused and nonfused Protein A accumulated in protein bodies. Hence, the increased yield could not be attributed to HFB‐induced protein body formation. We also demonstrated production of HFBI–Protein A fusion protein in tobacco BY‐2 suspension cells in 30 l scale, with a yield of 35 mg/l. Efficient partitioning to the surfactant phase confirmed that the fusion proteins retained the amphipathic properties of the hydrophobin block. The reversible antibody‐binding capacity of the Protein A block was similar to the nonfused Protein A. The best‐performing fusion protein was tested in capturing antibodies from hybridoma culture supernatant with two‐phase separation. The fusion protein was able to carry target antibodies to the surfactant phase and subsequently release them back to the aqueous phase after a change in pH. This report demonstrates the potential of hydrophobin fusion proteins for novel applications, such as harvesting antibodies in solutions.  相似文献

Species of the D. virilis group are widely used in evolutionary research, but the individuals of different species are difficult to distinguish from each other morphologically. We constructed a fast and easy microsatellite-based identification method for the species of the group occurring sympatrically in northern Europe. The neighbor joining tree based on 14 microsatellite loci also gave a good resolution of the species divergence pattern in the whole group.  相似文献

We aimed to study the association of perceived control over scheduling of shifts with objectively measured working hour characteristics in shift workers. The participants were 5128 hospital employees (91% women, 85% nursing personnel, average age 43 years) in period-based work (114:45h/3 weeks) from the 2015 Finnish Public Sector study. Survey responses to a measure of control over scheduling of shifts were linked to payroll data on working hour characteristics during the 91 days preceding the survey. We used multinomial logistic regression to assess differences in dichotomized proportion of working hour characteristics (being full-time worker, number of work shifts, long work weeks (>40h and >48h/week), long work shifts (>12-h), evening and night shifts, quick returns (<11h shift interval), single days off, weekend work, >4 consecutive work shifts, and variability of shift length with cut points at 10% or 25% between employees with high, intermediate, or low control over scheduling of shifts. Analyses were adjusted for age, sex, education, full-/part-time work (where applicable), duration of shift work experience, perceived work ability, children < 18 years in the household, and overall stressfulness of the life situation. Differences between age groups, men and women, and levels of work ability were examined using interaction terms. In adjusted analyses, the proportion of full-time workers was lower among employees with intermediate control over scheduling of shifts compared to those with high control (OR 0.78, 95% CI 0.61–0.98). High proportion (>25%) of weekend work was lower among employees with low control over scheduling of shifts compared to high control (OR 0.75, 95% CI 0.61–0.93). High proportion (>25%) of having >4 consecutive work shifts was associated with lower control over scheduling of shifts (OR 1.35, 95% CI 1.13–1.62). Variability of shift length was lower among employees with intermediate and low control over scheduling of shifts compared to those with high control (OR 0.78, 95% CI 0.66–0.93; OR 0.62, 95% CI 0.51–0.75, respectively). No association was observed between the level of control over scheduling of shifts and high proportion of long work weeks (>25% of >40h weeks and >10% of >48h weeks), long work shifts (>25%), quick returns (>25%), single days off (>25%), and evening or night shifts (>10%) in the whole sample. In subgroup analyses, women with low control over scheduling shifts had lower odds ratio (OR 0.58, 95% CI 0.37–0.91) and men had higher odds ratio (OR 2.97, 95% CI 1.26–6.98) for large proportion of >12-h shifts. In conclusion, the employees with high control over scheduling of shifts had slightly more often unsocial working hour characteristics than those with intermediate or low control over scheduling of shifts. The findings, however, suggest that good work time control in shift work can be possible without compromising shift ergonomics.  相似文献
