Protective properties of moderate wine consumption against cancers, cardiovascular, metabolic and degenerative diseases have been reported in various clinical studies. Here, we analysed the effect of red wine (RW) and white wine (WW) on myelination using an in vitro embryonic co-culture mouse model. The total amount of myelin was found to be significantly increased after RW and WW treatment, while only RW significantly increased the number of internodes. Both types of wine increased rat Schwann cell- (rSC) expression of the NAD+-dependent deacetylase sirtuin-two-homolog 2 (Sirt2), a protein known to be involved in myelination.Detailed chemical analysis of RW revealed a broad spectrum of anthocyanins, piceids, and phenolics, including resveratrol (RSV). In our assay system RSV in low concentrations induced myelination. Furthermore RSV raised intracellular glutathione concentrations in rSCs and in co-cultures and therefore augmented antioxidant capacity.We conclude that wine promotes myelination in a rodent in vitro model by controlling intracellular metabolism and SC plasticity. During this process, RSV exhibits protective properties; however, the fostering effect on myelinaton during exposure to wine appears to be a complex interaction of various compounds.  相似文献

Populations of the copepod Cyclops vicinus in two Danish lakes differed in their life cycles. In Lake Væng C. vicinus was absent during summer, whereas in Lake Søbygård it continued producing distinct cohorts throughout summer. A comparison between the lakes showed that in the lake where C. vicinus was absent during summer, food limitation generally existed. In contrast, abundance of C. vicinus during summer in Lake Søbygård always coincided with available resources sufficient for the whole life cycle. Thus, summer diapause of C. vicinus is suggested to be a strategy to avoid food limitation. Avoidance of fish predation does not seem substantially to influence life cycles.  相似文献

The MR (Mre11 nuclease and Rad50 ABC ATPase) complex is an evolutionarily conserved sensor for DNA double-strand breaks, highly genotoxic lesions linked to cancer development. MR can recognize and process DNA ends even if they are blocked and misfolded. To reveal its mechanism, we determined the crystal structure of the catalytic head of Thermotoga maritima MR and analyzed ATP-dependent conformational changes. MR adopts an open form with a central Mre11 nuclease dimer and two peripheral Rad50 molecules, a form suited for sensing obstructed breaks. The Mre11 C-terminal helix-loop-helix domain binds Rad50 and attaches flexibly to the nuclease domain, enabling large conformational changes. ATP binding to the two Rad50 subunits induces a rotation of the Mre11 helix-loop-helix and Rad50 coiled-coil domains, creating a clamp conformation with increased DNA-binding activity. The results suggest that MR is an ATP-controlled transient molecular clamp at DNA double-strand breaks.  相似文献

Objectives

to determine the prevalence of asymptomatic ischemic heart disease (IHD) in HIV patients by myocardial perfusion scintigraphy (MPS) and to determine the value of coronary artery calcium score (CACS), carotid intima-media thickness (cIMT) and pericardial fat volume as screening tools for detection of IHD in subjects with HIV.

Background

Patients with HIV seem prone to early development of IHD.

Methods

105 consecutive HIV patients (mean age 47.4 years; mean duration of HIV 12.3 years; mean CD4+ cell count 636×106/L; all receiving antiretroviral therapy) and 105 controls matched for age, gender and smoking status, without history of IHD were recruited. MPS, CACS, cIMT, pericardial fat volume, and cardiovascular risk scores were measured.

Results

HIV patients demonstrated higher prevalence of perfusion defects than controls (18% vs. 0%; p<0.001) despite similar risk scores. Of HIV patients with perfusion defects, 42% had a CACS = 0. CACS and cIMT were similar in HIV patients and controls. HIV patients on average had 35% increased pericardial fat volume and increased concentration of biomarkers of atherosclerosis in the blood. HIV patients with myocardial perfusion defects had increased pericardial fat volume compared with HIV patients without perfusion defects (314±43 vs. 189±12 mL; p<0.001).

Conclusions

HIV patients had an increased prevalence of silent IHD compared to controls as demonstrated by MPS. The finding was strongly associated with pericardial fat volume, whereas cardiovascular risk scores, cIMT and CACS seem less useful as screening tools for detection of myocardial perfusion defects in HIV patients.  相似文献

Small shrubs ofCeanothus thyrsiflorus were grown in 19-1 pots irrigated under natural conditions in a chaparral region of Southern California and then subjected to soil drying. Characteristics of leaf gas exchange, leaf water potential, and concentrations of the stress hormone abscisic acid in the xylem sap, ABAxyl, were determined at various stages of drought. Diurnal changes in conductance were strongly correlated with leaf net photosynthesis rate, which provides an effective, integrative predictor of above-ground climate effects on conductance. In drought conditions, ABAxyl concentration increased. Increases in the concentration range of 50–500 nmol/l appeared to induce stomatal closure, restricting water loss and carbon dioxide uptake. When the momentary water potential is related to ABAxyl, ABA appeared to increase significantly only after a threshold of approximately –1.5 MPa was exceeded. At less negative water potentials, large variation in ABAxyl in the 50–1000 nmol/l range occurred for all water-potential values, because ABAxyl remains relatively constant over diurnal courses as water potentials decrease and then recover. When the water potential became more negative than –1.5 MPa, ABAxyl concentrations occurred between approximately 500 and 10 000 nmol/l and even greater in isolated cases. An approximately linear relationship is recognizable between ABAxyl and momentary water potential in this range because in plants under drought conditions, ABAxyl increases during the course of the day as water potential decreases. Increases in ABAxyl in the high concentration range were associated with relatively minor additional restrictions in gas exchange, but they might contribute to improved water use efficiency and explain diurnal changes in the potential for stomatal opening that have been observed in Mediterranean sclerophyllous species. When we examined long-term seasonal change in the response of irrigated plants, changes in average daily temperature greater than 10°C occurred (also associated with shifts in relative humidity and radiation input), which apparently led to small changes in predawn water potential in the –0.1 to –0.7 MPa range. Increases in ABAxyl occurred that were in turn negatively correlated with daily maximum leaf conductance. Thus, chaparral shrubs under non-drought conditions seem to sense even small changes in environmental conditions, in our opinion most probably due to initial drying of the uppermost soil and synthesis of ABA in the shallow roots. The results support the hypothesis that information of photosynthesis rate and predawn water potential may be used as primary variables to predict canopy conductance of Mediterranean sclerophyll shrub vegetation.  相似文献

Seedlings of Ricinus communis L. were cultivated in quartz sandand supplied with media which contained either different concentrationsof nitrate or ammonium nitrogen and were treated with a lowsalt stress. The concentration of ABA was determined in tissuesand in xylem and phloem saps. Between 41 and 51 day after sowing,abscisic acid (ABA) flows between roots and shoots were modelled.Long-distance transport of ABA was not stimulated under conditionsof nitrate deficiency (0.2 mol m–3). However, when ammoniumwas given as the only N source (1.0 mol m–3), ABA transportin both xylem and phloem was increased significantly. Mild saltstress (40 mol m–3 NaCl) increased ABA transport in nitrate-fedplants, but not in ammonium-fed plants. The leaf conductancewas lowered by salt treatment with both nitrogen sources, butit was always lower in ammonium-fed compared to nitrate-fedplants. A negative correlation of leaf conductance to ABA levelsin leaves or flow in xylem was found only in comparison of ammonium-fedto nitrate-fed plants. Abscisic acid (ABA) was detected in aqueous extracts of a range of different soils, beneath a range of crops, pasture and forest species. Assuming that all the ABA is dissolved in the soil solution concentrations ranged from 0.6–2.8 nM. This is in the range which computer simulations predict is required in soils in order to prevent ABA release from the root hair zones of plant roots. The concentration of ABA in the soil solution was highest in acid soils and in soils with reduced moisture, and was lowest in moist, neutral and moderately alkaline soils. ABA in the soil solution of maize fields increased during the vegetative period. After incubation in soil for 72 h, radioactive ABA was degraded by 30–40%. Tetcyclacis, an inhibitor of the oxidative breakdown of ABA, completely prevented the degradation of ABA in the soil solution. Acid conditions and high salt concentrations significantly retarded ABA breakdown.  相似文献

Plants, in contrast to other eukaryotes, possess not only homologs of subunit A (AtSPO11-1, 2, 3) but also of subunit B (AtTOP6B) of the archaebacterial topoisomerase VI. AtTOP6B and AtSPO11-3 are strongly expressed in somatic tissue of Arabidopsis and are able to interact with each other in vitro. A T-DNA insertion in AtTOP6B results in deficient cell proliferation; plants stop growing at the rosette stage, have small crinkled leaves, and die about 4 weeks after germination. Cultured root cells die after a limited number of cell divisions. The mitotic index of the root meristems is strongly reduced. Flow cytometric analysis demonstrates that endoreplication in mutant plants is stopped at the 8C stage; the last cycle is not completed in most cases. Mutant plants show a significant increase in nuclear DNA strand breaks. A T-DNA insertion mutant of AtSPO11-3 has a phenotype that is almost to that of AtTOP6B and the double mutant. Thus, both genes seem to act in vivo as subunits of a functional entity. A loss of this function most likely results in a defect in DNA replication, leading directly, or via the activation of a DNA damage checkpoint, to an arrest of cell division and endoreduplication. The dependence on an archaebacterial topoisomerase VI homolog distinguishes plants from the other eukaryotic kingdoms.  相似文献
