The interaction between calcium-saturated chicken calmodulin and a peptide corresponding to the calmodulin-binding domain of the chicken smooth muscle myosin light chain kinase has been studied by multinuclear and multidimensional nuclear magnetic resonance methods. Extensive 1H and 15N resonance assignments of calmodulin in the complex have been obtained from the analysis of two- and three-dimensional nuclear magnetic resonance spectra. The assignment of calmodulin in the complex was facilitated by the use of selective labeling of the protein with alpha-15N-labeled valine, alanine, lysine, leucine, and glycine. These provided reference points during the main-chain-directed analysis of three-dimensional spectra of complexes prepared with uniformly 15N-labeled calmodulin. The pattern of nuclear Overhauser effects (NOE) seen among main-chain amide NH, C alpha H, and C beta H hydrogens indicates that the secondary structure of the globular domains of calmodulin in the complex closely corresponds to that observed in the calcium-saturated state of the protein in the absence of bound peptide. However, the backbone conformation of residues 76-84 adopts an extended chain conformation upon binding of the peptide in contrast to its helical conformation in the absence of peptide. A sufficient number of NOEs between the globular domains of calmodulin and the bound peptide have been found to indicate that the N- and C-terminal regions of the peptide interact with the C- and N-terminal domains of calmodulin, respectively. The significance of these results are discussed in terms of recently proposed models for the structure of calmodulin-peptide complexes.  相似文献

Given the influence of photoperiod on reproductive development and whole-plant senescence in monocarpic plants, one would suspect that leaf senescence in these plants might be under photoperiodic control. In Arabidopsis thaliana , which is monocarpic and also a nonobligate long-day (LD) plant, LDs (16 h, 300 μmol m−2 s−1) caused leaves to die earlier than did short days (SDs, 10 h). Since leaf longevity was not paralleled by the reproductive development in the present study, the reproductive structures did not seem to be the primary controls of leaf senescence. The LD effect appeared to depend on the amount of light rather than on day length, for leaves given LDs at reduced light intensity (180 μmol m−2 s−1) lived longer than those in LDs with full light. In addition, the higher light intensity promoted chlorophyll loss and anthocyanin accumulation in LDs. Thus, senescence of these leaves seems to be governed by light dosage rather than photoperiod. Light may play a natural role in promoting the senescence of A. thaliana leaves.  相似文献

We have isolated and characterized an aspartate transaminase (glutamate:oxalacetate transaminase, EC 2.6.1.1) from the thermophilic microorganism Bacillus stearothermophilus. The purified enzyme has a molecular mass of 40.5 kDa by sodium dodecyl sulfate gel analysis, a temperature optimum of 95 degrees C, and a pH optimum of 8.0. The corresponding gene, aspC, was cloned and overexpressed in Escherichia coli. The recombinant glutamate:oxalacetate transaminase protein was used in immobilized form together with 4-aminobutyrate:2-ketoglutarate transaminase (EC 2.6.1.19) from E. coli for the production of L-phosphinothricin [L-homoalanin-4-yl-(methyl)phosphinic acid], the active ingredient of the herbicide Basta (AgrEvo GmbH), from its nonchiral 2-keto acid precursor 2-oxo-4-[(hydroxy)(methyl)phosphinoyl]butyric acid (PPO). In this new coupled process conversion rates of ca. 85% were obtained with substrate solutions containing 10% PPO by using only slight excesses of the amino donors glutamate and aspartate. The contamination of the reaction broth with amino acid by-products was < 3%.  相似文献

Viable counts of aerobic and anaerobic chemotrophic sulphur-oxidizers as well as phototrophic sulphur bacteria were determined in sediment samples taken from two different areas along the Baltic Sea shore which were known to regularly develop sulphidic conditions. Depth profiles of bacterial cell counts were correlated with concentration profiles of chloride, sulphate, sulphide, nitrate and phosphate in the pore water of these sediments and with potential activities of nitrate reduction, thiosulphate transformation and sulphate formation. The data revealed a complex multilayered structure within the sediments. Sulphide was released into the water from sediments of both sampling areas, but it was found that light and the availability of oxygen significantly reduced this amount. In the highly reduced sediment at Hiddensee, the highest numbers of phototrophic and chemotrophic sulphur-oxidizers were found near the sediment surface. Therefore, it was concluded that the combined action of both groups of bacteria most efficiently oxidizes reduced sulphur compounds in the top layers of the sediments. Nitrate may replace oxygen as final electron acceptor and will support oxidation of sulphide, in particular when oxygen and light are limiting.  相似文献

Two synthetic fragments, corresponding to the 4–9 and 4–14 sequences of a tetradecapeptide used as a model to test the RXVRG-endoprotease activity from Xenopus laevis skin, have been studied by two-dimensional nmr spectroscopies, correlated spectroscopy, and nuclear Overhauser effect (NOE) spectroscopy. Both peptides wore the 5–9 consensus sequence found in several hormonal precursors. The nmr data for the 4–9 hexapeptide did not indicate any particular organization, either in water or in dimethylsulfoxide (DMSO), whereas, the 4–14 undecapeptide, a substrate for the RXVRG endoprotease, showed, in DMSO solution, significant trends of structural organization involving the amino acids pertaining to the consensus domain. From variations of integrated NOE peaks with temperature, the appearent interproton correlation times τc were estimated and the maxima observed with Va17, the central residue in the consensus sequence. A defined tertiary structure in that domain was also supported by medium-and long-range NOEs between As6 and Arg8, Glu4 and Gly9, and by the likely involvement of Arg8 and Gly9 NHs in intramolecular hydrogen bonds. Most of these observations could be rationalized by an equilibrium between a 5–3 β-turn and a 9 → 4 H-bonded loop. The predominance of one rotamer for the Cα-Cβ bond was established in four residues. Finally, the average ? and ψ angles were derived from two models taking, or not, into account variations in the correlation times along the sequence. This allowed us to discuss the artifacts generated by using an average correlation time through the whole molecule. © 1994 John Wiley & Sons, Inc.  相似文献

Summary The apical cell ofSphacelaria, a tip-growing filamentous brown alga, and its protoplast constitute a model for the investigation of the consequences of cell wall removal on microtubular cytoskeletal organization and cell polarity. In the apical cell, the microtubular cytoskeleton is strongly polarized and, in most cases, extends from two centrosomes to the cortex where it constitutes a fine meshwork. Observations of microtubule dynamics throughout the cell cycle emphasize the coincidence between orientation of the mitotic axis and cell polarity. Just after protoplast isolation, dramatic alterations of initial polarity are observed, whatever the mitotic stage. In particular, the coincidence between cytoplasmic polarity and polarity of the system nucleus-centrosomes is lost in most cases. 12–24 h after protoplast isolation, the cell shows a more symmetrical organization while a dense cortical microtubular network spreads out concomitantly with wall reformation. Our discussion emphasizes the possible relationship between cell polarity and cell totipotency, and the relevance of such a model for higher plant studies.  相似文献

The ipiB and ipiO genes of the potato late blight fungus Phytophthora infestans (Mont.) de Bary were isolated from a genomic library in a screen for genes induced in planta. Expression of these genes was studied during pathogenesis on various host tissues and different host plants, some of which show specific resistance against P. infestans infection. During pathogenesis on leaves and tubers of the fully susceptible potato cultivar (cv.) Ajax and on leaves of the fully susceptible tomato cv. Moneymaker, the P. infestans ipiB and ipiO genes show a transient expression pattern with highest mRNA levels in the early stages of infection. During the interaction with leaves of the partially resistant potato cv. Pimpernel, the expression is also transient but accumulation and disappearance of the mRNAs is delayed. Also in P. infestans inoculated onto a race-specific resistant potato cultivar and onto the nonhost Solanum nigrum, ipiB and ipiO mRNA is detectable during the initial stages of infection. Apparently, the expression of the ipiB and the ipiO genes is activated in compatible, incompatible and nonhost interactions. In encysted zoospores, ipiB and ipiO mRNA accumulation was not detectable, but during cyst germination and appressorium formation on an artificial surface the genes are highly expressed. Expression studies in mycelium grown in vitro revealed that during nutrient starvation the expression of the ipiB and ipiO genes is induced. For ipiO gene expression, carbon deprivation appeared to be sufficient. The ipiO gene promoters contain a sequence motif that functions as a glucose repression element in yeast and this motif might be involved in the regulation of ipiO gene expression.  相似文献

This study assessed the potential of two types of primate exhibit both for enhancing zoo visitors' interest, knowledge, and enjoyment and for promoting conservation education. Visitors' reactions to a free-ranging group of cotton-top tamarins were compared with their responses to caged tamarins in three ways: 1) timing how long visitors spent at each exhibit, 2) recording the comments made by visitors about the exhibits, and 3) asking visitors to respond to questionnaires about the two exhibits. The results showed that the free-ranging group of tamarins provoked wider comment from members of the public than caged groups and that visitors were willing to spend much more time looking for and watching monkeys in trees than monkeys in cages. Visitors felt that improvements in the animals' welfare were obtained from allowing them to live free in the trees, thought that they could learn more from such groups than from caged animals, and enjoyed seeing the free-ranging tamarins more than the monkeys in cages. These results suggest that developing more naturalistic zoo exhibits can have considerable benefits not only for the animals involved, but also for public education in conservation issues. © 1994 Wiley-Liss, Inc.  相似文献

The fungus Mortierella alpina CBS 343.66 was grown in a fermentor at different pH, temperatures and supplies of carbon source (glucose) in order to optimize the culture conditions for rapid biomass and lipid production with a high proportion of polyunsaturated fatty acids, especially arachidonic acid (AA). Good growth and lipid production with 31% AA was obtained at pH 6.5 and 25°C. A temperature decrease to 18°C gave a significantly higher degree of polyunsaturated fatty acids. Eicosapentaenoic acid was not detected at 25°C, but was formed at 18°C at about 10%. The AA concentration but was similar for 18°C about 10%. The AA concentration was similar for 18°C and 25°C (around 30%): 18°C allowed relatively good growth and had a beneficial effect on the fungus morphology, i.e. pellets were formed. Best lipid production and a AA content of up to 33% was achieved at an excess of glucose (carbon source) and a deficit of ammonium chloride (nitrogen source). The percentage of AA of the total fatty acid composition was constant as ong as glucose was present. At glucose exhaustion, the proportion of AA increased to 57%. The increase in AA corresponded to a decrease in palmitic acid, stearic acid and oleic acid. Correspondence to: G. Molin  相似文献
