Abundance and distribution of plant cell surface proteins of the hydroxyproline-rich glycoprotein (HRGP) class were studied in the pea- Rhizobium symbiosis using immunoblot analysis. The MAC 265-epitope was especially abundant in pea root nodules containing nitrogen-fixing Rhizobium bacteria. A 180-kDa MAC 265-HRGP dominated in pea shoot plasma membranes, while almost no MAC 265-HRGP was detected in root plasma membranes. We show here that a major difference between the plant-derived peribacteroid membrane of the symbiosomes and the root plasma membrane was the presence of a 100-kDa MAC 265-HRGP in the former. Arabinogalactan proteins (AGPs), as recognized by the monoclonal antibodies MAC 207 and JIM 8, were not detected in the peribacteroid membrane, while two isoforms (100 and 220 kDa) were detected in shoot and root plasma membranes. Specific MAC 265-HRGP isoforms were found in the peribacteroid space fraction of the symbiosomes and thus as soluble proteins in the interface between the symbionts. The abundance of the MAC 265-epitope was much reduced in non-nitrogen-fixing nodules when this phenotype resulted from a dicarboxylate transport mutation in Rhizobium . There was no reduction in the abundance of the MAC 265-epitope in non-fixing phenotypes resulting from a mutation in the plant. The results suggest that bacterial signals related to the bacterial ability to fix nitrogen, might be responsible for the regulation of HRGP expression in root nodules.  相似文献

This study investigated the mechanisms for glucose-induced islet blood flow increase in rats. The effects of adenosine, adenosine receptor antagonists, and vagotomy on islet blood flow were evaluated with a microsphere technique. Vagotomy prevented the islet blood flow increase expected 3, 10, and 20 min after injection of glucose, whereas theophylline (a nonspecific adenosine receptor antagonist) prevented the islet blood flow increase from occurring 10 and 20 min after glucose administration. Administration of selective adenosine receptor antagonists suggested that the response to theophylline was mediated by A1 receptors. Exogenous administration of adenosine did not affect islet blood flow, but local accumulation of adenosine, induced by the adenosine uptake inhibitor dipyridamole, caused a doubling of islet blood flow. In conclusion, the increased islet blood flow seen 3 min after induction of hyperglycemia is caused by the vagal nerve, whereas the increase in islet blood perfusion seen at 10 and 20 min after glucose administration is caused by both the vagal nerve and adenosine.  相似文献

Huge breeding aggregations of red-sided garter snakes (Thamnophis sirtalis parietalis) at overwintering dens in Manitoba provide a unique opportunity to identify sources of mortality and to clarify factors that influence a snake's vulnerability to these factors. Comparisons of sexes, body sizes, and body condition of more than 1000 dead snakes versus live animals sampled at the same time reveal significant biases. Three primary sources of mortality were identified. Predation by crows, Corvus brachyrhynchos (590 snakes killed), was focussed mostly on small snakes of both sexes. Crows generally removed the snake's liver and left the carcass, but very small snakes were sometimes brought back to the nest. Suffocation beneath massive piles of other snakes within the den (301 dead animals) involved mostly small males and (to a lesser extent) large females; snakes in poor body condition were particularly vulnerable. Many emaciated snakes (n = 142, mostly females) also died without overt injuries, probably due to depleted energy reserves. These biases in vulnerability are readily interpretable from information on behavioral ecology of the snakes. For example, sex biases in mortality reflect differences in postemergence behavior and locomotor capacity, the greater attractiveness of larger females to males, and the high energy costs of reproduction for females.  相似文献

One of the most definitive examples of a vertebrate extraorganismal structural protein can be found in three-spined sticklebacks (Gasterosteus aculeatus). In the breeding male the kidney hypertrophies and synthesizes an adhesive protein called "spiggin," which is secreted into the urinary bladder from where it is employed as a structural thread for nest building. This paper describes the first molecular characterization of spiggin and demonstrates that this adhesive is a protein complex assembled from a potential of three distinct subunits (alpha, beta, and gamma). These subunits arise by alternative splicing, and 11-ketoandrogens induce their expression in stickleback kidneys. Analysis of the predicted amino acid sequence of each subunit reveals a modular organization whose structural elements display a similarity to the multimerization domains found within von Willebrand Factor-related proteins. These results implicate that spiggin utilizes a conserved multimerization mechanism for the formation of a viscous agglutinate from its constituent subunits in the urinary bladders of male sticklebacks. This novel extraorganismal structural protein is therefore ideally suited to its function as an adhesive thread.  相似文献

Retinoids have been shown to modulate inflammation and the immune response in many cell types including macrophages, endothelial cells, and vascular smooth muscle cells. However, present knowledge of whether inflammatory mediators modulate vitamin A status in these cells is limited. To identify the role of inflammation on retinoid metabolism in vascular smooth muscle cells, the cells were exposed to a combination of proinflammatory cytokines: interleukin-1beta, interferon-gamma, and lipopolysaccharides. Without stimulation with proinflammatory cytokines, vascular smooth muscle cells expressed retinol dehydrogenases-2 and 5 mRNA detected by RT-PCR. Stimulation with the combination of cytokines induced a substantial increase of retinol dehydrogenase-5 mRNA. This was associated with increased production of ligands for retinoic acid receptors, when assayed in a retinoic acid receptor-dependent luciferase reporter system. Our results demonstrate that inflammatory mediators activate the retinoid metabolic pathway in vascular smooth muscle cells, which potentially may modulate the inflammatory response in the vascular wall.  相似文献

Cultures of Pseudomonas fluorescens DF57 were grown on different carbon and nitrogen sources. Glucose, succinate and acetate were used as carbon source and pulsed to an aerobic steady-state cultivation of P. fluorescens DF57 at D = 0.1 h(-1) with citrate as limiting carbon source. Glucose was utilised with the fastest uptake rate (19.4 C mmol l(-1) h(-1)) compared to succinate (8.8 C mmol l(-1) h(-1)) and acetate (4.3 C mmol l(-1) h(-1)). Acetate triggered an inhibition of cellular metabolism, which resulted in 2-h long growth arrest after its addition to the steady-state culture. The influence of the nitrogen source was investigated in an aerobic cultivation on a mixture of ammonium and nitrate as limiting nitrogen sources and citrate as non-limiting carbon source. When ammonia and nitrate were pulsed to the steady-state culture, they were mainly assimilated into biomass with a maximum uptake rate of 111 and 33 mg N l(-1) h(-1), respectively. Nitrate uptake was never complete as the residual concentration in the chemostat cultivation was 30 mg N l(-1) nitrate. A pulse of nitrite in the cultivation broth resulted in an inhibition of the growth but not of the primary metabolism, as nitrite was taken up at 38 mg N l(-1) h(-1), citrate was consumed and cofactors were produced continuously. In all experiments, oxygen was used as electron acceptor.  相似文献

In zebrafish (Danio rerio) pigmentation is initiated during embryogenesis and begins in the retinal epithelium and in the melanophores. The pigment cells develop rapidly, and within hours they constitute a prominent feature of the embryo. In order to improve signal detection by whole mount in situ hybridization, confocal microscopy, or expression of GFP, embryos may be treated with 1-phenyl 2-thiourea (PTU) during embryogenesis. PTU inhibits melanogenesis by blocking all tyrosinase-dependent steps in the melanin pathway but can be toxic at high concentrations. The embryos remain transparent as long as the PTU treatment is continued. However, PTU treatment must be initiated before the initial pigmentation because it does not remove already formed pigment. Here we provide a protocol for generating transparent zebrafish while avoiding the toxic and teratogenic effects of PTU treatment. Received December 4, 2000; accepted April 7, 2001.  相似文献

This study reports effects on soil solution chemistry and plant uptake of 55 elements (Ag, Al, As, B, Ba, Be, Bi, Br, Ca, Cd, Ce, Co, Cr, Cs, Cu, Dy, Er, Eu, Fe, Gd, Ge, Hf, Hg, Ho, K, La, Li, Lu, Mg, Mn, Mo, Na, Nb, Nd, Ni, P, Pb, Pr, Rb, S, Sb, Sc, Se, Si, Sm, Sr, Tb, Th, Tl, U, V, W, Y, Yb, Zn, Zr) by raising the pH using addition of fine-grained precipitated calcium carbonate at 20 rates (yielding a soil solution pH range of 5.2 – 7.8) to A horizon samples of an acid Cambisol, cultivating a common grass (Agrostis capillaris L.) and determining the soil solution, root and shoot concentrations of these elements at the end of the experiment. For many of these elements, there is little or no previous information about concentrations in soil solutions, or in plant biomass, as related to soil pH/acidity or addition of calcium carbonate. Soil solutions were obtained by high speed centrifugation and ultrafiltration (0.2 m) of samples at 60% water-holding capacity. Concentrations of elements were determined by ICP-ES or (in most elements) ICP-MS, using isotopes specified. Soil solution pH, HCO3 and organic C were also determined.Concentrations of elements in the biomass of A. capillaris were usually inversely related to soil solution pH. The most apparent (p<0.001) inverse, though often curvilinear, relationships between pH and concentrations in shoot biomass were measured for Ag, As, B, Ba, Eu, Ge, Li, Mn, Ni, P and Sr. Positive relationships (p<0.05) were only measured in Ca, Hg, Mg, Mo and S. For concentrations in root biomass, relationships were mostly, but not always, of the same sign and of a similar strength. Though soil solution pH and concentrations of elements were usually quite closely correlated, pH and/or HCO 3 concentration more often accounted for a higher share of the variability in biomass concentration of elements than did soil solution concentration of the same element.  相似文献

Hypothesising that systemically and intrathecally produced nitric oxide might play different roles in the EAE pathogenesis, we administered the NOS inhibitor N-nitro-methyl-L-arginine-ester intrathecally or systemically via osmotic minipumps to DA rats with MOG induced EAE. We demonstrate an protective effect of the NOS inhibitor on EAE severity, the extent of CNS inflammation, and demyelination. Intrathecal administration was more effective when compared to systemic administration. The observed effect was accompanied by enhanced anti-MOG IgG1 production. In our model, the therapeutic effect was concluded to be due to direct inhibition of the NO pathway in the CNS.  相似文献

The ventral sense organ, below the mouth, is composed of 30 primary sensory cells situated in a row perpendicular to the long axis of the animal. Each cell carries one long and slender, modified cilium which arises from an apical pocket in the cell. The sensory cells project 15 axons at each side of the pharynx to the brain, which is rostrally paired and terminates in bulb-like swellings. Each of these bulbs contains four cell bodies, which, according to their fine structure, as well as the synaptic connections with receptor and brain fibers, belong to three different types. It is suggested that the sense organ is a chemosensor and that its remarkable similarity to the vertebrate mechanisms for olfaction makes it probable that the appendicularian Oikopleura dioica possesses a ‘protochordate’ counterpart to the craniate olfactory apparatus.  相似文献
