The synthesis and the binding study of new 3-arylesters and 3-heteroarylpyrazolo[5,1-c][1,2,4]benzotriazine 5-oxide 8-substituted are reported. The nature of these substituents (in terms of lipophilic and electronic features) seems to influence the binding affinity. High-affinity ligands were studied in mice in vivo for their pharmacological effects, considering six potential benzodiazepine actions: anxiolytic-like effects, muscle relaxant effects, motor coordination, anticonvulsant action, spontaneous motor activity, and ethanol-potentiating action. Compounds 4d and 6d showed an inverse-agonist profile. These compounds were evaluated also for their binding at benzodiazepine site on GABAA receptor complex (GABAA/BzR complex) subtype to evaluate their subtype selectivity.  相似文献

Plants continuously generate new tissues and organs through the activity of populations of undifferentiated stem cells, called meristems. Here, we discuss the so-called shoot apical meristem (SAM), which generates all the aerial parts of the plant. It has been known for many years that auxin plays a central role in the functioning of this meristem. Auxin is not homogeneously distributed at the SAM and it is thought that this distribution is interpreted in terms of differential gene expression and patterned growth. In this context, auxin transporters of the PIN and AUX families, creating auxin maxima and minima, are crucial regulators. However, auxin transport is not the only factor involved. Auxin biosynthesis genes also show specific, patterned activities, and local auxin synthesis appears to be essential for meristem function as well. In addition, auxin perception and signal transduction defining the competence of cells to react to auxin, add further complexity to the issue. To unravel this intricate signaling network at the SAM, systems biology approaches, involving not only molecular genetics but also live imaging and computational modeling, have become increasingly important.Plants continuously generate new tissues and organs through the activity of populations of undifferentiated stem cells, called meristems. Apical meristems, positioned at the tip of the shoots and roots, initiate aerial and underground organs, respectively. Along the stems and roots, more diffuse secondary meristems are responsible for secondary thickening of these structures.The plant hormone auxin plays an instrumental role in meristem biology and we discuss here its role in a particular meristem, the shoot apical meristem (SAM) that generates all the aerial organs including the floral meristems (Fig. 1A–C). Over the last three decades, climate abnormalities have been reported to be involved in biodiversity decline by affecting population dynamics. A growing number of studies have shown that the North Atlantic Oscillation (NAO) influences the demographic parameters of a wide range of plant and animal taxa in different ways. Life history theory could help to understand these different demographic responses to the NAO. Indeed, theory states that the impact of weather variation on a species’ demographic traits should depend on its position along the fast–slow continuum. In particular, it is expected that NAO would have a higher impact on recruitment than on adult survival in slow species, while the opposite pattern is expected occur in fast species. To test these predictions, we used long‐term capture–recapture datasets (more than 15,000 individuals marked from 1965 to 2015) on different surveyed populations of three amphibian species in Western Europe: Triturus cristatus, Bombina variegata, and Salamandra salamandra. Despite substantial intraspecific variation, our study revealed that these three species differ in their position on a slow–fast gradient of pace of life. Our results also suggest that the differences in life history tactics influence amphibian responses to NAO fluctuations: Adult survival was most affected by the NAO in the species with the fastest pace of life (Tcristatus), whereas recruitment was most impacted in species with a slower pace of life (Bvariegata and Ssalamandra). In the context of climate change, our findings suggest that the capacity of organisms to deal with future changes in NAO values could be closely linked to their position on the fast–slow continuum.  相似文献

Herps, especially amphibians, are particularly susceptible to climate change, as temperature tightly controls many parameters of their biological cycle—above all, their phenology. The timing of herps’ activity or migration period—in particular the dates of their first appearance in spring and first breeding—and the shift to earlier dates in response to warming since the last quarter of the 20th century has often been described up to now as a nearly monotonic trend towards earlier phenological events. In this study, we used citizen science data opportunistically collected on reptiles and amphibians in the northern Mediterranean basin over a period of 32 years to explore temporal variations in herp phenology. For 17 common species, we measured shifts in the date of the species’ first spring appearance—which may be the result of current changes in climate—and regressed the first appearance date against temperatures and precipitations. Our results confirmed the expected overall trend towards earlier first spring appearances from 1983 to 1997, and show that the first appearance date of both reptiles and amphibians fits well with the temperature in late winter. However, the trend towards earlier dates was stopped or even reversed in most species between 1998 and 2013. We interpret this reversal as a response to cooling related to the North Atlantic Oscillation (NAO) in the late winter and early spring. During the positive NAO episodes, for certain species only (mainly amphibians), the effect of a warm weather, which tends to advance the phenology, seems to be counterbalanced by the adverse effects of the relative dryness.  相似文献

  • Setting up effective conservation strategies requires the precise determination of the targeted species’ distribution area and, if possible, its local abundance. However, detection issues make these objectives complex for most vertebrates. The detection probability is usually <1 and is highly dependent on species phenology and other environmental variables. The aim of this study was to define an optimized survey protocol for the Mediterranean amphibian community, that is, to determine the most favorable periods and the most effective sampling techniques for detecting all species present on a site in a minimum number of field sessions and a minimum amount of prospecting effort. We visited 49 ponds located in the Languedoc region of southern France on four occasions between February and June 2011. Amphibians were detected using three methods: nighttime call count, nighttime visual encounter, and daytime netting. The detection nondetection data obtained was then modeled using site‐occupancy models. The detection probability of amphibians sharply differed between species, the survey method used and the date of the survey. These three covariates also interacted. Thus, a minimum of three visits spread over the breeding season, using a combination of all three survey methods, is needed to reach a 95% detection level for all species in the Mediterranean region. Synthesis and applications: detection nondetection surveys combined to site occupancy modeling approach are powerful methods that can be used to estimate the detection probability and to determine the prospecting effort necessary to assert that a species is absent from a site.

Capsule Woodpigeon Columba palumbus nest-tree selection is affected by human disturbance, grazing and tree characteristics.

Aims To determine the characteristics of trees used for nesting by Woodpigeons under two levels of grazing and disturbance in a Holm oak Middle Atlas forest, Morocco.

Methods Woodpigeon nest-tree selection was investigated in highly disturbed and less-disturbed sites over two years (2010 and 2011). The features of nest-tree characteristics were determined by comparing the characteristics of trees used for nesting to those of randomly selected trees.

Results Tree physical characteristics, grazing intensity and level of human disturbance were the main factors determining selection for nest-trees. Woodpigeons chose nesting trees that were greater in height, larger in DBH, lower in base of the live crown, in denser stands and at greater distance from tracks.

Conclusions Forest management for Woodpigeon nest habitat selection should focus on controlling the access of vehicles especially trucks, introducing a rotating system of forest utilization for grazing and maintaining (without extending) the cereal crops. These recommendations may assist land managers to plan for continued persistence of this species in Middle Atlas forests.  相似文献

During the last decade, the metal hyperaccumulating plants have attracted considerable attention because of their potential use in decontamination of heavy metal contaminated soils. However, in most species, little is known regarding the function, the ecological and the evolutionary significances of hyperaccumulation. In our study, we investigated the parameters influencing the Cd concentration in plants as well as the biological implications of Cd hyperaccumulation in nine natural populations of Thlaspi caerulescens. First, we showed that Cd concentration in the plant was positively correlated with plant Zn, Fe, and Cu concentrations. This suggested that the physiological and/or molecular mechanisms for uptake, transport and/or accumulation of these four heavy metals interact with each other. Second, we specified a measure of Cd hyperaccumulation capacity by populations and showed that T. caerulescens plants originating from populations with high Cd hyperaccumulation capacity had better growth, by developing more and bigger leaves, taller stems, and produced more fruits and heavier seeds. These results suggest a tolerance/disposal role of Cd hyperaccumulation in this species.  相似文献

Organismal aging entails a gradual decline of normal physiological functions and a major contributor to this decline is withdrawal of the cell cycle, known as senescence. Senescence can result from telomere diminution leading to a finite number of population doublings, known as replicative senescence (RS), or from oncogene overexpression, as a protective mechanism against cancer. Senescence is associated with large-scale chromatin re-organization and changes in gene expression. Replication stress is a complex phenomenon, defined as the slowing or stalling of replication fork progression and/or DNA synthesis, which has serious implications for genome stability, and consequently in human diseases. Aberrant replication fork structures activate the replication stress response leading to the activation of dormant origins, which is thought to be a safeguard mechanism to complete DNA replication on time. However, the relationship between replicative stress and the changes in the spatiotemporal program of DNA replication in senescence progression remains unclear.

Here, we studied the DNA replication program during senescence progression in proliferative and pre-senescent cells from donors of various ages by single DNA fiber combing of replicated DNA, origin mapping by sequencing short nascent strands and genome-wide profiling of replication timing (TRT).

We demonstrate that, progression into RS leads to reduced replication fork rates and activation of dormant origins, which are the hallmarks of replication stress. However, with the exception of a delay in RT of the CREB5 gene in all pre-senescent cells, RT was globally unaffected by replication stress during entry into either oncogene-induced or RS. Consequently, we conclude that RT alterations associated with physiological and accelerated aging, do not result from senescence progression. Our results clarify the interplay between senescence, aging and replication programs and demonstrate that RT is largely resistant to replication stress.  相似文献

In this study we used a multi-spatial scale approach to investigate habitat suitability, roosting characteristics, and ecological niche in two flying fox species on the Comoros Islands—Pteropus livingstonii and Pteropus seychellensis comorensis. At a broad scale, we assessed the ecological niche and habitat suitability for both species using the Species Distribution Modeling method based on the recent ensembles of small models (ESM) approach. At a fine scale, Ecological Niche Factor Analysis (ENFA) was applied to assess habitat selection by each species. Direct observation was used at each roost to estimate the total number of individuals and to identify the roost characteristics. At both broad and fine scales, the analyses highlighted clear niche partitioning by the two species. We found that P. livingstonii has a very limited distribution, restricted to steep, high-elevation slopes of the islands’ remaining natural forests, and the patterns were the same for roosting, foraging sites and the entire habitat. By contrast, P. s. comorensis has a relatively large geographic range that extends over low-elevation farmlands and villages and it was negatively correlated to natural forest across the entire area and all roosting sites, but its foraging areas were positively correlated to natural forest and high elevation areas. Both species selected large, tall trees for roosting. The total number of individuals in the studied area was estimated to be 1243 P. livingstonii and 11,898 P. s. comorensis. The results of our study demonstrated that these two species use different habitat types and ensure different ecosystem services in pollination and seed dispersion and thus are both critical for maintaining overall ecosystem dynamics. However, the currently high level of hunting pressure and roost disturbance makes them vulnerable to extinction. To ensure the viability of both species, conservation measures need to be taken by the Comoros government.  相似文献
