Pup production and mortality of beach breeding Grey seals was studied at two contrasting types of rookery, the cliff-bound island of Ramsey, Dyfed, and the low, grassy island of Auskerry, Orkney. Thirty-five per cent of pups died on Ramsey compared with 14 % on Auskerry. Almost half of the Ramsey mortality was accounted for by animals being lost from the beaches so that they were not available for analysis. The main proximate causes of death revealed by post-mortem examination were starvation and infections. The other conditions which accounted for a small proportion of deaths were drowning, trauma, non-viability, stillbirths, atelectasis, dystokia and heart abnormality. Certain pathogens associated with disease were common to both sites, while others were specific to Ramsey or Auskerry.
The most important ultimate cause of pup deaths appeared to be failure of the mother/pup bond. The differences in levels of mortality between the sites is thought to be due to differences in the topography of the beaches. Pup survival on the narrow cliff-bound beaches, or in caves, on Ramsey was reduced either directly through pups being washed off, or indirectly by overcrowding of animals at high tide.  相似文献

Phosphoethanolaminetransferase of high specific activity was solubilized from rat liver microsomes with the non-ionic detergent octyl glucoside. The solubilization method is fast and simple, allowing for processing of large amounts of material. The solubilized enzyme is stable. It contains virtually no phosphocholinetransferase activity. A preliminary characterization of the enzyme, with both diacyl- and alkylacyl-glycerol as substrate, is given. For the reaction, the lipid substrates were incorporated into artificial phospholipid bilayers (liposomes).  相似文献

Summary The effects of chronic administration of ACTH (up to 36 consecutive days) on the mitochondria of the zona reticularis of the rat adrenal cortex were investigated by stereologic techniques. It was found that ACTH induces two phases of hypertrophy of mitochondria alternating with two proliferative stages, which are associated with a significant decrease in the average volume of the organelles. It is suggested that, as in the zona fasciculata, ACTH controls the processes of growth and division of mitochondria in the zona reticularis. The mechanism underlying this action of ACTH as well as the differences between the responses to ACTH of the mitochondrial population of the two adrenal zones are discussed in the light of evidence indicating that mitochondria contain a complete genetic apparatus largely independent of nuclear control.The authors wish to thank Miss A. Background: Ehlers-Danlos syndrome (EDS) is a common non-inflammatory, congenital connective tissue disorder. Classical type (cEDS) EDS is one of the more common forms, typically caused by mutations in the COL5A1 and COL5A2 genes, though causative mutations in the COL1A1 gene have also been described. Material and methods: The study group included 59 patients of Polish origin, diagnosed with cEDS. The analysis was performed on genomic DNA (gDNA) with NGS technology, using an Illumina sequencer. Thirty-five genes related to connective tissue were investigated. The pathogenicity of the detected variants was assessed by VarSome. Results: The NGS of 35 genes revealed variants within the COL5A1, COL5A2, COL1A1, and COL1A2 genes for 30 of the 59 patients investigated. Our panel detected no sequence variations for the remaining 29 patients. Discussion: Next-generation sequencing, with an appropriate multigene panel, showed great potential to assist in the diagnosis of EDS and other connective tissue disorders. Our data also show that not all causative genes giving rise to cEDS have been elucidated yet.  相似文献

Feeding ecology is an important factor for the survival of a species and knowledge of its parameters is a prerequisite for successful conservation work. In this study we describe the feeding ecology of the endemic Seychelles Black Parrot Coracopsis barklyi on Praslin, Seychelles, the only island on which this parrot is resident. We compared two methods to evaluate feeding choices: incidental observations and feeding walks on 25 transects in all habitat types. Black parrots fed on 46 different species, bringing the total number of known food plants to 53 species. They predominantly consumed endemic and native species (58% of observed feeding bouts), mainly their fruit pulp (in 68% of feeding bouts), followed by buds (15%) and seeds (37%) with occasional observations of leaves, bark and scale insects. The incidental method rendered many more observed bouts than the transect approach and the ratios of consumed species differed between methods but the transect results are regarded as more representative. The incidental method is not suitable for quantitative conclusions but complements the transect method, providing information about rarely occurring feeding events.  相似文献

The present study focuses on temporal and microscale spatial variation of the community structure and richness of subaerial microalgae growing on the bark of European beech (Fagus sylvatica) trees in temperate deciduous forests. Subaerial phototrophic biofilms present common and conspicuous microalgal communities growing on a variety of natural and man‐made substrata. However, in comparison with other major microalgal communities such as phytoplankton and microphytobenthos, basic patterns of their spatio‐temporal variation remain largely unknown. The bark samples were collected six times each spring and autumn in a period of 3 years (2010–2013) and were cultured on agar plates, and then individual clonal strains were identified by light microscopy. A total of 55 morphotypes (considered as operational taxonomic units for subsequent analyses) were recognized, which mainly belong to the classes Trebouxiophyceae and Chlorophyceae. Interestingly, temporal variation explained the largest proportion of variation in the community structure. This variation was primarily related to seasonal fluctuations, and although the communities recorded in spring and autumn showed many overlapping taxa, a clear distinction in species composition and abundance was observed. However, the microhabitat characteristics such as bark roughness also significantly structured the microalgal community. Conversely, spatial factors such as the height of the samples above ground or distance of the samples on a trunk seemed to be of lesser importance on this scale. Thus, we concluded that the previously unrecognized seasonal changes, resulting from variation in temperature, humidity, and irradiance, as well as the non‐seasonal temporal changes, possibly resulting from local colonization or extinction of individual taxa, should be considered as one of the important factors in structuring aerial microalgal communities.  相似文献

Gene knockout in murine embryonic stem cells (ESCs) has been an invaluable tool to study gene function in vitro or to generate animal models with altered phenotypes. Gene targeting using standard techniques, however, is rather inefficient and typically does not exceed frequencies of 10(-6). In consequence, the usage of complex positive/negative selection strategies to isolate targeted clones has been necessary. Here, we present a rapid single-step approach to generate a gene knockout in mouse ESCs using engineered zinc-finger nucleases (ZFNs). Upon transient expression of ZFNs, the target gene is cleaved by the designer nucleases and then repaired by non-homologous end-joining, an error-prone DNA repair process that introduces insertions/deletions at the break site and therefore leads to functional null mutations. To explore and quantify the potential of ZFNs to generate a gene knockout in pluripotent stem cells, we generated a mouse ESC line containing an X-chromosomally integrated EGFP marker gene. Applying optimized conditions, the EGFP locus was disrupted in up to 8% of ESCs after transfection of the ZFN expression vectors, thus obviating the need of selection markers to identify targeted cells, which may impede or complicate downstream applications. Both activity and ZFN-associated cytotoxicity was dependent on vector dose and the architecture of the nuclease domain. Importantly, teratoma formation assays of selected ESC clones confirmed that ZFN-treated ESCs maintained pluripotency. In conclusion, the described ZFN-based approach represents a fast strategy for generating gene knockouts in ESCs in a selection-independent fashion that should be easily transferrable to other pluripotent stem cells.  相似文献
