A 122‐day growth trial was conducted to observe the impact of polyhouse in winter on growth, conversion efficiencies and body composition of fingerling Labeo rohita (7.9 ± 1.1 cm; 4.60 ± 0.18 g). Fish were stocked (1.5 m?2) in polyhouse and outdoor concrete tanks (250 m2) in duplicate and fed a compound diet [35% crude protein (CP)] to apparent satiation twice daily, at 09.00 and 17.00 hours. A natural photoperiod was maintained during the trials. At the end of the trials, polyhouse‐reared fish produced significantly (P < 0.01) higher values for weight increment, specific growth rate (SGR %), protein efficiency rate (PER), protein productive value (PPV) and better feed conversion ratio (FCR). Polyhouse‐reared fish showed higher (P < 0.01) CP and fat, and lower (P < 0.01) moisture and ash contents in the muscle. In the second part of the study, which also lasted 122 days, polyhouse‐reared young L. rohita (21.7 ± 1.3 cm; 127.8 ± 0.69 g) were raised to marketable size in outdoor earthen ponds (500 m2) at a stocking density of 0.37 m?2 to examine the effects of feeding traditional feed mixture and compound diet (30% CP) on growth and body composition of fish. Labeo rohita fed the compound diet showed higher (P < 0.01) values for weight increment, SGR (%), PER and PPV, and better (P < 0.01) FCR than those fed traditional feed. Proximate composition of fish muscle showed higher (P < 0.05) CP and fat, and lower (P < 0.05) moisture and ash contents in fish fed the compound diet. The results suggest that polyhouse may be used successfully to culture L. rohita during winter, and that thereafter the fish may be reared in earthen ponds using the compound diet to obtain better production. 相似文献

N-formyl-methionyl-leucyl-phenylalanine (fMLP) a potent chemotactic peptide stimulates immune responses by activating macrophages and other cells of the immune system. The present study reports inhibition of fMLP-induced activation of murine peritoneal and P388D-1 macrophage cell line by protein kinase C (PKC) inhibitors, H-7 and chelerythrine chloride. Similarly, tumoricidal activity was also downregulated by protein tyrosine kinase (PTK) inhibitors genestein and lavendustin A. Further, fMLP increased tyrosine phosphorylation of several proteins in murine macrophages, which were inhibited in presence of genestein and lavendustin A. The production of phytase and associated feed enzymes (phosphatase, xylanase, CMCase, alpha-amylase and beta-glucosidase) was determined in a thermotolerant fungus Mucor indicus MTCC 6333, isolated from composting soil. Solid-substrate culturing on wheat bran and optimizing other culture conditions (C and N sources, level of N, temperature, pH, culture age, inoculum level), increased the yield of phytase from 266 +/- 0.2 to 513 +/- 0.4 nkat/g substrate dry mass. The culture extract also contained 112, 194, 171, 396, and 333 nkat/g substrate of phosphatase, xylanase, CMCase, beta-glucosidase and alpha-amylase activities, respectively. Simple 2-step purification employing anion exchange and gel filtration chromatography resulted in 21.9-fold purified phytase. The optimum pH and temperature were pH 6.0 and 70 degrees C, respectively. The phytase was thermostable under acidic conditions, showing 82% residual activity after exposure to 60 degrees C at pH 3.0 and 5.0 for 2 h, and displayed broad substrate specificity. The Km was 200 nmol/L and v(lim) of 113 nmol/s per mg protein with dodecasodium phytate as substrate. In vitro feed trial with feed enzyme resulted in the release of 1.68 g inorganic P/kg of feed after 6 h of incubation at 37 degrees C. 相似文献

1. The purpose of the present study was to determine the effect of zinc on the status of various neurotransmitters as well
as behavioral patterns of lithium-treated rats. The study was designed with a view to find out whether zinc affords protection
to brain against lithium toxicity.
2. Animals were segregated into four different groups. Animals in group I were fed with standard laboratory feed and water
ad libitum and served as normal controls. Animals in group II and IV were given lithium in the form of lithium carbonate in their diet
at a dose level of 1.1 g/Kg diet. Animals in group III and IV were given zinc treatment in the form of zinc sulfate at a dose
level of 227 mg/L mixed in drinking water of animals.
3. The effects of all the treatments were studied for a duration of 1, 2, and 4 months with regard to the parameters, which
included estimation of serotonin and dopamine concentrations as well as the activity of acetylcholinesterase in cerebral cortex
of rat brain. Further, passive avoidance, active avoidance, and behavior despair tests were conducted to assess the short-term
memory, cognitive behavior, and psychomotor dysfunction of the animals, respectively.
4. Initially, a decrease in the acetylcholinesterase activity was reported in cerebral cortex followed by an increase in the
enzyme activity after 2 and 4 months of lithium treatment. Serotonin concentration significantly decreased after 2 and 4 months
of lithium treatment, whereas dopamine concentration increased significantly after 4 months of lithium treatment. Zinc administration
to the lithium-treated group significantly improved the acetylcholinesterase activity as well as the concentration of dopamine
and serotonin. Further, lithium-treated rats showed an increase in depression time as compared to normal controls both after
1 and 4 months of treatment. Short-term memory significantly improved in lithium-treated rats in all treatment groups. However,
no change in the cognitive behavior of the animals was reported after lithium treatment. Zinc co-administration with lithium
significantly improved the short-term memory and cognitive functions of the animals. From the above results it can be concluded
that zinc proved beneficial in altering the status of neurotransmitters as well as the behavior patters of the animals treated

This study reports the production of xylanolytic and cellulolytic enzymes by a thermophilic fungal isolate Myceliophthora sp. using a cheap medium containing rice straw and chemically defined basal medium under solid-state culture. A combination of one factor at a time approach followed by response surface methodology using Box-Behnken design of experiments resulted in 2.5, 1.25, 1.28 and 4.23 fold increase in xylanase, endoglucanase, beta-glucosidase and FPase activity, respectively. The zymograms developed against IEF gels showed that multiple isoforms of xylanase (5), endoglucanase (4) and beta-glucosidase (2) were produced under optimized culture conditions. This study reports the purification and characterization of endoglucanases (EG I and EG II) from a newly isolated thermophilic fungus, Melanocarpus sp. MTCC 3922. The molecular weight of EG I and EG II as with SDS-PAGE and pI were approximately 40 and 50 kDa, and approximately 4.0 and 3.6, respectively. EG I and EG II were optimally active at 50 and 70 degrees C, and pH 6.0 and 5.0, respectively. EG I was active over a broad range of pH (5.0-7.0), whereas, loss of activity was observed as the temperature was increased from 50 to 80 degrees C. However, EG II was active over pH 4.0-6.0 and temperature 40-80 degrees C. The presence of mercaptoethanol and SDS inhibited the EG I activity but showed no negative effect on EG II. Both the endoglucanases showed higher activity against barley-beta-glucan as compared to CMC. Km values of EG I and EG II for barley-beta-glucan were lower than CMC. Turn over number (K(cat)) and catalytic efficiency (K(cat)/Km) values of both the endoglucanases were higher with barley-beta-glucan as substrate than CMC. EG I showed affinity for Avicel indicating the presence of cellulose binding domains (CBD) whereas, EG II was found to lack CBD. 相似文献

Posttranslational modification by small ubiquitin-like modifiers (SUMOs), known as SUMOylation, is a key regulatory event in many eukaryotic cellular processes in which SUMOs interact with a large number of target proteins. SUMO binding motifs (SBMs) are small peptides derived from these target proteins that interact noncovalently with SUMOs and induce conformational changes. To determine the effect of SBMs on the mechanical properties of SUMO1 (the first member of the human SUMO family), we performed single-molecule force spectroscopy experiments on SUMO1/SBM complexes. The unfolding force of SUMO1 (at a pulling speed of 400 nm/s) increased from ∼130 pN to ∼170 pN upon binding to SBMs, indicating mechanical stabilization upon complexation. Pulling-speed-dependent experiments and Monte Carlo simulations measured a large decrease in distance to the unfolding transition state for SUMO1 upon SBM binding, which is by far the largest change measured for any ligand binding protein. The stiffness of SUMO1 (measured as a spring constant for the deformation response along the line joining the N- and C-termini) increased upon SBM binding from ∼1 N/m to ∼3.5 N/m. The relatively higher flexibility of ligand-free SUMO1 might play a role in accessing various conformations before binding to a target. 相似文献

This study was aimed to investigate the effect of inoculation on three salt-tolerant, plant-growth-promoting rhizobacteria (PGPR) STR2 (Bacillus pumilus), STR8 (Halomonas desiderata) and STR36 (Exiguobacterium oxidotolerans), for their growth-promoting potential and efficacy in augmenting salt tolerance in Mentha arvensis, an essential oil-bearing crop and natural source of l-menthol, under varying levels of NaCl stress (0, 100, 300 and 500 mM) imposed through irrigating water. Increase in the levels of salt concentration led to a decrease in the growth, fresh weight, leaf–stem ratio, oil content and yield. However, the negative effects of salinity were observed to be convalesced in the PGPR inoculated plants. At salinity levels of 100 and 300 mM NaCl, H. desiderata inoculated plants recorded the highest herb yield, whereas at 500 mM NaCl, the plants inoculated with E. oxidotolerans yielded maximum herb. The oil content in non-inoculated, salt-stressed plants was observed to be 0.46, 0.42 and 0.35 % at 100, 300 and 500 mM NaCl, respectively, whereas the plants inoculated with H. desiderata recorded the oil content of 0.71 and 0.60 and 0.48 % at similar levels of NaCl stress, respectively. The halotolerant PGPR minimized the deleterious effects of salt toxicity producing at par or higher yields at lower and medium salinity levels (100, 300 mM NaCl) than the un-inoculated non-salt-stressed plants through improved foliar nutrient uptake and enhanced antioxidant machinery. Based on the results of the experiments reported herein, the use of salt-tolerant, plant-growth-promoting bacteria may provide an effective means of facilitating M. arvensis growth in salt-stressed environments. 相似文献
