Increasing air pollution has resulted in ozone depletion, which has led to an increase in the amount of ultraviolet (UV) radiation reaching the surface of earth posing hazardous effects on the plant yield. The chlorophyll a fluorescence transients were recorded in vivo using plant efficiency analyzer and analyzed according to JIP test, which can monitor photosystem II (PSII) behavior. This study aims to evaluate the sensitivity of different components of PSII to UV radiations and the extent of damage caused when wheat plants were exposed to UV radiations for 2 and 4 h. It was observed that functional disconnection of light harvesting complexes from PSII complex, accumulation of inactive reaction center, inactivation of oxygen evolving complex, and thus the linear electron transport process (ET0/CS), were drastically affected by UV radiations. This study will contribute to understanding of the basic photosynthetic mechanisms affected in crop plants due to increased UV radiations. The knowledge obtained will be relevant for environmental and plant scientists to plan strategies to cope with stresses. 相似文献

Endothelial dysfunction is associated with increase in oxidative stress and low NO bioavailability. The endothelial NO synthase (eNOS) uncoupling is considered an important factor in endothelial cell oxidative stress. Under increased oxidative stress, the eNOS cofactor tetrahydrobiopterin (BH4) is oxidized to dihydrobiopterin, which competes with BH4 for binding to eNOS, resulting in eNOS uncoupling and reduction in NO production. The importance of the ratio of BH4 to oxidized biopterins versus absolute levels of total biopterin in determining the extent of eNOS uncoupling remains to be determined. We have developed a computational model to simulate the kinetics of the biochemical pathways of eNOS for both NO and O2•− production to understand the roles of BH4 availability and total biopterin (TBP) concentration in eNOS uncoupling. The downstream reactions of NO, O2•−, ONOO−, O2, CO2, and BH4 were also modeled. The model predicted that a lower [BH4]/[TBP] ratio decreased NO production but increased O2•− production from eNOS. The NO and O2•− production rates were independent above 1.5 μM [TBP]. The results indicate that eNOS uncoupling is a result of a decrease in [BH4]/[TBP] ratio, and a supplementation of BH4 might be effective only when the [BH4]/[TBP] ratio increases. The results from this study will help us understand the mechanism of endothelial dysfunction. 相似文献

The identification of a potent, selective, and orally available MK2 inhibitor series is described. The initial absence of oral bioavailability was successfully tackled by moving the basic nitrogen of the spiro-4-piperidyl moiety towards the electron-deficient pyrrolepyridinedione core, thereby reducing the pKa and improving Caco-2 permeability. The resulting racemic spiro-3-piperidyl analogues were separated by chiral preparative HPLC, and the activity towards MK2 inhibition was shown to reside mostly in the first eluting stereoisomer. This led to the identification of new MK2 inhibitors, such as (S)-23, with low nanomolar biochemical inhibition (EC50 7.4 nM) and submicromolar cellular target engagement activity (EC50 0.5 μM). 相似文献

Cellulose esters containing adipates and other ester groups are synthesized by the reaction of commercially available cellulose esters in solution with the benzyl monoester of adipoyl chloride. The products, cellulose adipate esters in which the distal end of the adipate moiety is a benzyl ester, were easily converted to cellulose adipate derivatives by Pd-catalyzed hydrogenation. Mutations in isocitrate dehydrogenases (IDHs) have a gain-of-function effect leading to R(−)-2-hydroxyglutarate (R-2HG) accumulation. By using biochemical, structural and cellular assays, we show that either or both R- and S-2HG inhibit 2-oxoglutarate (2OG)-dependent oxygenases with varying potencies. Half-maximal inhibitory concentration (IC50) values for the R-form of 2HG varied from approximately 25 μM for the histone Nɛ-lysine demethylase JMJD2A to more than 5 mM for the hypoxia-inducible factor (HIF) prolyl hydroxylase. The results indicate that candidate oncogenic pathways in IDH-associated malignancy should include those that are regulated by other 2OG oxygenases than HIF hydroxylases, in particular those involving the regulation of histone methylation. 相似文献

Clostridium perfringens is the causative agent of clostridial myonecrosis or gas gangrene and produces many different extracellular toxins and enzymes, including the cysteine protease α-clostripain. Mutation of the α-clostripain structural gene, ccp, alters the turnover of secreted extracellular proteins in C. perfringens, but the role of α-clostripain in disease pathogenesis is not known. We insertionally inactivated the ccp gene C. perfringens strain 13 using TargeTron technology, constructing a strain that was no longer proteolytic on skim milk agar. Quantitative protease assays confirmed the absence of extracellular protease activity, which was restored by complementation with the wild-type ccp gene. The role of α-clostripain in virulence was assessed by analysing the isogenic wild-type, mutant and complemented strains in a mouse myonecrosis model. The results showed that although α-clostripain was the major extracellular protease, mutation of the ccp gene did not alter either the progression or the development of disease. These results do not rule out the possibility that this extracellular enzyme may still have a role in the early stages of the disease process. 相似文献
