Fibroblast growth factor 21 is a novel hormonal regulator with the potential to treat a broad variety of metabolic abnormalities, such as type 2 diabetes, obesity, hepatic steatosis, and cardiovascular disease. Human recombinant wild type FGF21 (FGF21) has been shown to ameliorate metabolic disorders in rodents and non-human primates. However, development of FGF21 as a drug is challenging and requires re-engineering of its amino acid sequence to improve protein expression and formulation stability. Here we report the design and characterization of a novel FGF21 variant, LY2405319. To enable the development of a potential drug product with a once-daily dosing profile, in a preserved, multi-use formulation, an additional disulfide bond was introduced in FGF21 through Leu118Cys and Ala134Cys mutations. FGF21 was further optimized by deleting the four N-terminal amino acids, His-Pro-Ile-Pro (HPIP), which was subject to proteolytic cleavage. In addition, to eliminate an O-linked glycosylation site in yeast a Ser167Ala mutation was introduced, thus allowing large-scale, homogenous protein production in Pichia pastoris. Altogether re-engineering of FGF21 led to significant improvements in its biopharmaceutical properties. The impact of these changes was assessed in a panel of in vitro and in vivo assays, which confirmed that biological properties of LY2405319 were essentially identical to FGF21. Specifically, subcutaneous administration of LY2405319 in ob/ob and diet-induced obese (DIO) mice over 7–14 days resulted in a 25–50% lowering of plasma glucose coupled with a 10–30% reduction in body weight. Thus, LY2405319 exhibited all the biopharmaceutical and biological properties required for initiation of a clinical program designed to test the hypothesis that administration of exogenous FGF21 would result in effects on disease-related metabolic parameters in humans. 相似文献

Stability and function of eukaryotic genomes are closely linked to chromatin structure and organization. During cell division the entire genome must be accurately replicated and the chromatin landscape reproduced on new DNA. Chromatin and nuclear structure influence where and when DNA replication initiates, whereas the replication process itself disrupts chromatin and challenges established patterns of genome regulation. Specialized replication-coupled mechanisms assemble new DNA into chromatin, but epigenome maintenance is a continuous process taking place throughout the cell cycle. The newly described piscine reovirus (PRV) appears to be associated with the development of heart and skeletal muscle inflammation (HSMI) in farmed Atlantic salmon Salmo salar L. PRV seems to be ubiquitous among fish in Norwegian salmon farms, but high viral loads and tissue distribution support a causal relationship between virus and disease. In order to improve understanding of the distribution of PRV in the salmon production line, we quantified PRV by using real-time PCR on heart samples collected at different points in the life cycle from pre-smolts to fish ready for slaughter. PRV positive pre-smolts were found in about 36% of the freshwater cohorts and a general increase in viral load was observed after their transfer to seawater. A reduction in viral loads was recorded when fish approached slaughter (18 mo in sea cages). Sequencing of positive samples did not support the hypothesis that outbreaks are caused by the spreading of a particular (virulent) strain of PRV. 相似文献

Earthworms and arbuscular mycorrhizal fungi (AMF) have profound impacts on plant performance. However, it is largely unknown
if and how earthworms and AMF may affect plant succession. We planted mesocosms with an early-mid successional and a mid-late
successional grassland plant community and added endogeic earthworms and commercial AMF in a full-factorial way to natural
background soil. Earthworms had a positive effect on the total root and shoot biomass of both plant communities, with the
effect on the shoots being slightly enhanced by co-inoculation with AMF. Surprisingly, the earthworm effect on the mid successional
plant species depended on the successional stage of the plant community. Earthworms had a positive effect on the mid successional
plant species when they were growing in the mid-late successional plant community, but no effect when the same plant species
were growing in the early-mid successional plant community. Addition of AMF alone tended to reduce the shoot biomass of the
early successional plant species, while the addition of earthworms in the presence or absence of AMF increased their shoot
biomass. We conclude that the impacts of earthworms on plant species may depend on the successional stage of the plant community,
while the effect of AMF addition depends on the successional stage of the plant community and may be changed by the presence
of earthworms. Earthworms and AMF addition affect plants and plant communities of different successional stages differently

RAB5 GTPases are important regulators of endosomal membrane traffic. Among them Arabidopsis thaliana ARA7/RABF2b is highly conserved and homologues are present in fungal, animal and plant kingdoms. In land plants ARA7 and its homologues are involved in endocytosis and transport towards the vacuole. Here we report on the isolation of an ARA7 homologue (CaARA7/CaRABF2) in the highly evolved characean green alga Chara australis. It encodes a polypeptide of 202 amino acids with a calculated molecular mass of 22.2 kDa and intrinsic GTPase activity. Immunolabelling of internodal cells with a specific antibody reveals CaARA7 epitopes at multivesicular endosomes (MVEs) and at MVE‐containing wortmannin (WM) compartments. When transiently expressed in epidermal cells of Nicotiana benthamiana leaves, fluorescently tagged CaARA7 localizes to small organelles (putative MVEs) and WM compartments, and partially colocalizes with AtARA7 and CaARA6, a plant specific RABF1 GTPase. Mutations in membrane anchoring and GTP binding sites alter localization of CaARA7 and affect GTPase activity, respectively. This first detailed study of a conventional RAB5 GTPase in green algae demonstrates that CaARA7 is similar to RAB5 GTPases from land plants and other organisms and shows conserved structure and localization. 相似文献

Extracellular nicotinamide adenine dinucleotide (NAD+) is known to increase the intracellular calcium concentration [Ca2+]i in different cell types and by various mechanisms. Here we show that NAD+ triggers a transient rise in [Ca2+]i in human monocytes activated with lipopolysaccharide (LPS), which is caused by a release of Ca2+ from IP3-responsive intracellular stores and an influx of extracellular Ca2+. By the use of P2 receptor-selective agonists and antagonists we demonstrate that P2 receptors play a role in the NAD+-induced calcium response in activated monocytes. Of the two subclasses of P2 receptors (P2X and P2Y) the P2Y receptors were considered the most likely candidates, since they share calcium signaling properties with NAD+. The identification of P2Y1 and P2Y11 as receptor subtypes responsible for the NAD+-triggered increase in [Ca2+]i was supported by several lines of evidence. First, specific P2Y1 and P2Y11 receptor antagonists inhibited the NAD+-induced increase in [Ca2+]i. Second, NAD+ was shown to potently induce calcium signals in cells transfected with either subtype, whereas untransfected cells were unresponsive. Third, NAD+ caused an increase in [cAMP]i, prevented by the P2Y11 receptor-specific antagonist NF157. 相似文献
