Using a sensitive, economical, and reproducible microassay, the relationship of toxoplasma inhibiting factor to interleukin 2 has been examined. The assay developed took advantage of the observation that (1) Toxoplasma gondii tachyzoites replicated efficiently in the murine monocytic cell line, RAW 264; (2) treatment of RAW 264 cells with toxoplasma inhibiting factor prevented intracellular replication of the parasite to an extent similar to that observed with identical treatment of freshly isolated murine peritoneal exudate cells; and (3) [3H]uracil incorporation was an efficacious means to quantify replication (or inhibition of replication) of tachyzoites within the cell line. Although toxoplasma inhibiting factor and interleukin 2 were both present in the same lectin -and antigen-stimulated splenocyte supernatant fluids, results from microassays strongly suggested that the molecules were two distinct entities. 相似文献

The Cyt f and P700 contents in leaves of three Sorghum, varietieswere measured, in relation to their carbon assimilation, underdifferent light intensities during growth. At the maximum irradiationused (1,800 µE m2 s1) the ratio of P700to Cyt f was close to unity, whereas under low irradiation (450µE m2 s1) the ratio of P700 to Cyt f rangedfrom two to three. A strikingly positive correlation existedbetween the P700 contents of the leaves and their rates of carbondioxide fixation, dry matter production and Cyt f contents,only when the plants were grown under high light intensities.The P700 content of the leaves in plants grown under low irradiationwas unrelated to the contents of Cyt f. Thus, at a high lightintensity there is a close relationship between the Cyt f andP700 levels, but at low intensities the amounts of electroncarriers and the reaction centre are independent. The binding ofRicinus communis agglutinin andAbrus agglutinin to 4-methylumbelliferyl β-D-galactopyranoside was studied by equilibrium dialysis, fluo-rescence quenching and fluorescence polarization. The number of binding sites and the association constant value obtained by fluorescence polarization for bothRicinus communis agglutinin andAbrus agglutinin are in close agreement with those obtained by the other methods. Lipid peroxidation, glutathione level and activity of glutathione-S-transferase were studied in liver and brain of rats 4 and 3 h after a single i.p. administration of 0, 25, 75, 100 mg/kg acrylamide or 0, 50, 100, 200, 600 mg/kg styrene, respectively. In liver both acrylamide and styrene caused an increase in lipid peroxidation and decrease in glutathione contents and activity of glutathione-S-transferase in a dose dependent manner, while in brain only acrylamide produced a decrease in glutathione content. The decrease in glutathione content was not always associated with increase of lipid peroxidation. The enhancement of lipid peroxidation occurred only when glutathione contents were depleted to certain critical levels. No effect of acrylamide or styrene was seen on lipid peroxidation under in vitro conditions. The addition of glutathione in the incubation mixture significantly inhibited the rate of lipid peroxidation of liver homogenates of acrylamide and styrene treated animals.The results suggest that enhancement of lipid peroxidation in liver on exposure to acrylamide or styrene is a consequence of depletion of glutathione to certain critical levels. The inhibition of glutathione-S-transferase activity by acrylamide and styrene suggests that detoxication of these neurotoxic compounds could be suppressed following acute exposure. 相似文献

The contents of phosphoinositides, ATP, glucose and lactate in leg and claw nerves of the lobster were determined. Nerves were also analysed after cyanide poisoning, after electrical stimulation, and 1 h after removing the leg from the lobster. Cyanide poisoning decreased the levels of ATP and glucose and increased the content of lactate but did not alter the levels of phosphoinositides. Nerves left in situ for 1 h after disconnection from the central nervous system exhibited a decrease in the content of tri-phosphoinositides (TPI) of 50 per cent, without changes in ATP, glucose or lactate. The TPI change was reversed after incubation for 1 h in oxygenated seawater. Nerves labelled in vivo with 32P were removed and stimulated at 50 Hz for 5 min. The turnover of TPI phosphorus increased on stimulation in both normal and cyanide-poisoned nerves. In contrast, turnover of ATP increased after stimulation in normal nerves but not in cyanide-treated nerves. We sought to determine whether polyphosphoinositides play a greater role in resting metabolism of the nerve or in the conducting mechanisms. Summary In the present study attempts have been made to elucidate the composition and physiological actions of root exudates from
CB-1 Rupsail and the mixed population of these two varieties. The chemical nature of the different components of root exudates
was studied by means of paper chromatography, bioassay and other biochemical procedures. It was noted that exudates from CB-1
and Rupsail seedlings inhibited root/shoot growth of test seedlings of both varieties. But when these two varieties were grown
together root/shoot growth of both varieties were markedly promoted. Both qualitative and quantitative differences in amino
