Recent studies have shown that phagosome maturation depends on the balance between pro-inflammatory and anti-inflammatory cytokines, indicating that cytokine modulates phagosome maturation. However, the mechanism of cytokine-mediated modulation of intracellular trafficking remains to be elucidated. Here, we have shown that treatment of macrophages with IL-6 specifically induce the expression of Rab5 through the activation of extracellular signal-regulated kinase, whereas IL-12 exclusively upregulate the expression of Rab7 through the activation of p38 MAPK. We have cloned the 5'-flanking regions of the rab5c or rab7 into the promoterless reporter vector. Our results have shown that cells transfected with rab5c chimera are transactivated by IL-6, and IL-12 specifically transactivates cells containing rab7 chimera. Moreover, our results also show that IL-12 induces lysosomal transport, whereas IL-6 stimulates the fusion between early compartments in macrophages and accordingly modulates Salmonella trafficking and survival in macrophages. This is the first demonstration showing that cytokine differentially regulates endocytic trafficking by controlling the expression of appropriate Rab GTPase, and provides insight into the mechanism of cytokine-mediated regulation of intracellular trafficking. 相似文献

It is not known why some women develop breast cancer-related lymphedema (BCRL) of the arm, whereas others having similar treatment do not. We speculated that increased uptake of protein into local blood may protect against BCRL. Sixteen women were given bilateral subcutaneous hand webspace injections of polyclonal immunoglobulin (HIgG), (99m)Tc-HIgG on one side and (111)In-HIgG on the other, before and 3 mo after axillary clearance surgery. The rates of clearance of activity from the depot (k) and accumulation in central blood (b(contra)) were measured using a scintillation probe and bilateral antecubital vein blood sampling, respectively. Activity accumulating in blood ipsilateral to the injected side, in excess of central blood activity (b(ipsi)) was also calculated as a measure of local vascular uptake. The k correlated with b(contra), but neither changed in response to surgery. However, b(ipsi) for injections of (99m)Tc-HIgG into the affected arm increased in all seven patients in whom data were available (0.018 +/- 0.006 to 0.038 +/- 0.007%/min; P < 0.05); indeed, in five of these seven, b(ipsi) paradoxically exceeded b(contra), and none developed BCRL at 3-yr follow-up. We conclude that uptake of protein into local blood and/or proteolysis increases after axillary surgery and may protect against BCRL. 相似文献

Morphology plays a vital role on the performance of organic photovoltaics. However, our understanding of the morphology‐performance relationships for organic photovoltaics remains lacking. Specifically, it is still an open question why some bulk‐heterojunction blends exhibit electric field dependent J–V curves, while others do not. Through detailed fs‐μs transient absorption spectroscopy and morphology studies on the representative bulk‐heterojunction type small molecule (SM) donor system, a picture of different J–V behaviors from morphology aspects and excited dynamics is revealed. Our findings reveal that amorphous morphology in the lack of percolated pathways leads to the formation of strongly bound charge transfer states (CTSs), which accounts for about one third of the photoexcited species. Therefore, field‐dependent J–V curves are obtained as these CTSs mainly undergo geminate recombination or function as interfacial traps for nongeminate recombination. On the other hand, the CTSs are totally suppressed after post‐treatment owning to the formation of bicontinuous morphology, which results in very high efficiencies from exciton generation, diffusion, dissociation to charge extraction, thus contributes to field‐independent J–V characteristics. The insights gained in this work provide the effective guidelines to further optimize the performance of bulk‐heterojunction type SM‐organic photovoltaics through judicious morphology control and engineering. 相似文献

The effect of temperatures and exogenous supply of different carbon and nitrogen sources on the conidial germination and appresoria formation inC. capsici has been studied. 25 °C was observed to be the best temperature for conidial germination. At 18 °C, conidia germinated only in hanging drops and did not germinate on 2 % agar. Amongst carbon sources, 1-rhamnose supported maximum conidial germination and appresoria formation. Potassium nitrate supported very good conidial germination and appresoria formation. Ammonium nitrate, ammonium sulphate and ammonium chloride were found to be inhibitory. A few aminoacids stimulated conidial germination but dl-alanine, dl-methionine, tyrosine and l-glutamine were found to be inhibitory. 相似文献

Accumulated evidence indicates that there is a circulating monoclonal Ig protein related to the leukemic cell-associated Ig in the majority of patients with B cell chronic lymphocytic leukemia (CLL) despite the failure to demonstrate such a protein by conventional serum electrophoresis. Methodology has been developed to reveal these hidden monoclonal bands and to show that they are related to the leukemia-associated membrane Ig (mIg). Of nine CLL cases with stainable mIgM and without discernable plasma Ig bands, marked hypogammaglobulinemia was evident in six. In the other three, a significant amount of protein was present in the gamma region. IgM was isolated from the plasma of these patients by affinity chromatography with Sepharose-4B, conjugated with affinity purified anti-human IgM antibodies. One to 3 mg were isolated from 20 to 40 ml of plasma. Agarose electrophoresis revealed a monoclonal Ig band in the isolated IgM in all cases. Eight of these IgM proteins were analyzed by high-pressure liquid chromatography. Five were found to be pentameric IgM. In the remaining three, various amounts of monomeric IgM were detected. Attempts to make anti-idiotypic antibodies to the isolated proteins have been successful. Thus far, a rabbit anti-idiotypic antiserum was obtained in one case and two mouse monoclonal anti-idiotypic antibodies in two additional cases. Immunofluorescence analysis revealed that plasma IgM and mIgM shared similar idiotypic determinants. One other monoclonal antibody was shown to be specific for a V region marker of a minor Ig population. These findings indicate that B leukemic lymphocytes do secrete a small amount of IgM and lend further support to the thesis that the maturation defect in CLL is incomplete. It is also feasible to isolate the secreted IgM and to produce anti-idiotypic antibodies to them. In view of the potential therapeutic effect of anti-idiotypic antibodies, this may offer an alternative and efficient approach to generate a large panel of anti-idiotypic antibodies for clinical trials. The possibility also exists that this approach is applicable to other B cell proliferative disorders such as the non-Hodgkin B cell lymphomas. 相似文献

Phosphorylation (activation) and dephosphorylation (deactivation) of the slit diaphragm proteins NEPHRIN and NEPH1 are critical for maintaining the kidney epithelial podocyte actin cytoskeleton and, therefore, proper glomerular filtration. However, the mechanisms underlying these events remain largely unknown. Here we show that NEPHRIN and NEPH1 are novel receptor proteins for hepatocyte growth factor (HGF) and can be phosphorylated independently of the mesenchymal epithelial transition receptor in a ligand-dependent fashion through engagement of their extracellular domains by HGF. Furthermore, we demonstrate SH2 domain–containing protein tyrosine phosphatase-2–dependent dephosphorylation of these proteins. To establish HGF as a ligand, purified baculovirus-expressed NEPHRIN and NEPH1 recombinant proteins were used in surface plasma resonance binding experiments. We report high-affinity interactions of NEPHRIN and NEPH1 with HGF, although NEPHRIN binding was 20-fold higher than that of NEPH1. In addition, using molecular modeling we constructed peptides that were used to map specific HGF-binding regions in the extracellular domains of NEPHRIN and NEPH1. Finally, using an in vitro model of cultured podocytes and an ex vivo model of Drosophila nephrocytes, as well as chemically induced injury models, we demonstrated that HGF-induced phosphorylation of NEPHRIN and NEPH1 is centrally involved in podocyte repair. Taken together, this is the first study demonstrating a receptor-based function for NEPHRIN and NEPH1. This has important biological and clinical implications for the repair of injured podocytes and the maintenance of podocyte integrity. 相似文献

We studied reproductive behavior of free-ranging capped langurs (Trachypithecus pileatus) in the Pakhui Wildlife Sanctuary, Arunachal Pradesh, India. Four species of primates —Trachypithecus pileatus, Macaca mulatta, M. assamensis, and Nycticebus bengalensis— live there. We studied the mating seasons, mating frequency, copulatory attempts, time spent in copulation, and interval
between 2 successive copulations, gestation length, and interbirth interval of 4 groups of capped langurs during 2001–2003.
We observed 2 mating seasons in a year. The first was larger, comprising 5 months (September–January), and the second was
short, April and May. Mating was intensive in the morning session (0600–1000 h); 57% of total mating events occurred then.
The average gestation period was 200 d. November was the most favorable month for breeding. In a year, 107 mating events occurred
involving 5 adult females. Average time per mounting attempt is 12 s. Duration of mounting was the maximum in November. Interbirth
interval was 23 months and 10 d. The birth season was 129 days, December–April; 53% of births occurred in February and March.
Average birth rate is 0.386 birth/female/yr. 相似文献
