There is a strong bias concerning the regions of the globe where research on biological invasions is conducted, with notably lower representation of developing countries. However, in developing countries, effective management strategies to control invasions could be more beneficial in conserving global biodiversity since these countries tend to have larger, highly diverse natural habitats. Lower levels of development are seen as an obstacle to tackling biological invasions, but little thought is given to the advantages of developing countries in dealing with invasive species. We analyzed differences between developed and developing countries regarding the problem of invasive species and their historical and current patterns of international trade, disturbance levels and land use, research and monitoring, control and mitigation, and social awareness. Developed nations have some advantages, especially in levels of social awareness and means for controlling and studying exotics, but developing nations also enjoy important advantages given their lower levels of international trade and the availability of low-cost labor. Also, there is evidence that the process of economic development, which results in more efficient ways to transform landscapes and increases international trade, is strongly associated with increasing rates of biological invasion. Differences in data quality and availability between developed and developing countries make comparative analyses of biological invasions a difficult task. Thus, these differences creates a challenge in forming global strategies to deal with invasions. There have been calls for creating international plans to deal with invasive species, but we believe that it is important first to acknowledge the challenges and understand both the advantages and disadvantages of developing countries.  相似文献

BACKGROUND: Cytochrome c release from mitochondria to cytosol is a hallmark of apoptosis and is used to characterize the mitochondria-dependent pathway of this type of cell death. Techniques currently used to measure cytochrome c release, Western blot and fluorescence microscopy of immunolabeled cells, are time-consuming and inaccurate, and the latter is still limited by sample size. METHODS: We developed a rapid and reliable technique to detect cytochrome c release during drug-induced apoptosis, using flow cytometry. Plasma membrane of apoptotic HL-60 cells and thymocytes, treated with staurosporine and dexamethasone, respectively, were selectively permeabilized by digitonin at a low concentration. The released cytochrome c was quickly washed out from cells and that which remained in the mitochondria was immunolabeled after fixing the cells. RESULTS: The fraction of cells that retained their mitochondrial cytochrome c, or the highly fluorescent cells, gradually decreased so that after 4-8 h of drug treatment almost all the cells lost their cytochrome c and emerged as a population of low fluorescent cells. This was confirmed by parallel fluorescence microscopy of cells immunolabeled for cytochrome c. CONCLUSIONS: This technique allows the analysis of cytochrome c release from mitochondria of a large number of apoptotic cells in a short period of time and is proposed as an alternative to the methods currently used for this same purpose.  相似文献

Non-native (alien, exotic) plant invasions are affecting ecological processes and threatening biodiversity worldwide. Patterns of plant invasions, and the ecological processes which generate these patterns, vary across spatial scales. Thus, consideration of spatial scale may help to illuminate the mechanisms driving biological invasions, and offer insight into potential management strategies. We review the processes driving movement of non-native plants to new locations, and the patterns and processes at the new locations, as they are variously affected by spatial scale. Dispersal is greatly influenced by scale, with different mechanisms controlling global, regional and local dispersal. Patterns of invasion are rarely documented across multiple spatial scales, but research using multi-scale approaches has generated interesting new insights into the invasion process. The ecological effects of plant invasions are also scale-dependent, ranging from altered local community diversity and homogenization of the global flora, to modified biogeochemical cycles and disturbance regimes at regional or global scales. Therefore, the study and control of invasions would benefit from documenting invasion processes at multiple scales.  相似文献

The mitochondrial effects of submicromolar concentrations of six triarylmethane dyes, withpotential applications in antioncotic photodynamic therapy, were studied. All dyes promotedan inhibition of glutamate or succinate-supported respiration in uncoupled mitochondria, in amanner stimulated photodynamically. No inhibition of N,N,N,N-tetramethyl-p-phenylenediamine(TMPD) supported respiration was observed, indicating that these dyes do not affectmitochondrial complex IV. When mitochondria were energized with TMPD in the absence ofan uncoupler, treatment with victoria blue R, B, or BO, promoted a dissipation of mitochondrialmembrane potential and increase of respiratory rates, compatible with mitochondrialuncoupling. This effect was observed even in the dark, and was not prevented by EGTA, Mg2+ orcyclosporin A, suggesting that it is promoted by a direct effect of the dye on inner mitochondrialmembrane permeability to protons. Indeed, victoria blue R, B, and BO promoted swellingof valinomycin-treated mitochondria incubated in a hyposmotic K+-acetate-based medium,confirming that these dyes act as classic protonophores such as FCCP. On the other hand, ethylviolet, crystal violet, and malachite green promoted a dissipation of mitochondrial membranepotential, accompanied by mitochondrial swelling, which was prevented by EGTA, Mg2+, andcyclosporin A, demonstrating that these drugs induce mitochondrial permeability transition.This mitochondrial permeabilization was followed by respiratory inhibition, attributable tocytochrome c release, and was caused by the oxidation of NAD(P)H promoted by these drugs.  相似文献

Ca2+ and inorganic phosphate-induced mitochondrial swelling and membrane protein thiol oxidation, which are associated with mitochondrial permeability transition, are inhibited by progressively decreasing the incubation medium pH between 7.2 and 6.0. Nevertheless, the detection of mitochondrial H2O2 production under these conditions is increased. Permeability transition induced by phenylarsine oxide, which promotes membrane protein thiol cross-linkage in a process independent of Ca2+ or reactive oxygen species, is also strongly inhibited in acidic incubation media. In addition, we observed that the decreased protein thiol reactivity with phenylarsine oxide or phenylarsine oxide-induced swelling at pH 6.0 is reversed by diethyl pyrocarbonate, in a hydroxylamine-sensitive manner. These results provide evidence that the inhibition of mitrochondrial permeability transition observed at lower incubation medium pH is mediated by a decrease in membrane protein thiol reactivity, related to the protonation of protein histidyl residues.  相似文献

In this report we study the effect of Fe(III) on lipid peroxidation induced by Fe(II)citrate in mitochondrial membranes, as assessed by the production of thiobarbituric acid-reactive substances and antimycin A-insensitive oxygen uptake. The presence of Fe(III) stimulates initiation of lipid peroxidation when low citrate:Fe(II) ratios are used ( 4:1). For a citrate:total iron ratio of 1:1 the maximal stimulation of lipid peroxidation by Fe(III) was observed when the Fe(II):Fe(III) ratio was in the range of 1:1 to 1:2. The lag phase that accompanies oxygen uptake was greatly diminished by increasing concentrations of Fe(III) when the citrate:total iron ratio was 1:1, but not when this ratio was higher. It is concluded that the increase of lipid peroxidation by Fe(III) is observed only when low citrate:Fe(II) ratios were used. Similar results were obtained using ATP as a ligand of iron. Monitoring the rate of spontaneous Fe(II) oxidation by measuring oxygen uptake in buffered medium, in the absence of mitochondria, Fe(III)-stimulated oxygen consumption was observed only when a low citrate:Fe(II) ratio was used. This result suggests that Fe(III) may facilitate the initiation and/or propagation of lipid peroxidation by increasing the rate of Fe(II)citrate-generated reactive oxygen species.  相似文献

The CO2 evolution of intact potato tubers (Solanum tuberosum, L., var. Bintje) was analyzed during a 10-day period of their warm (25 ± 2°C) or cold (5 ± 1°C) storage, to evaluate cold-stress effects on expression and activities of plant uncoupling mitochondrial protein (PUMP) and alternative oxidase (AOX). CO2 evolution rates were analyzed at 20°C, to reflect their possible capacities. The 20°C CO2 production declined from 13 to 8 mg kg–1 h–1 after 2 days of warm storage and then (after 3 to 7 days) decreased from 8 to 6.5 mg kg–1 h–1. In contrast, 20°C CO2 evolution did not change after the first day of cold storage, increased up to 14.5 mg kg–1 h–1 after 2 days, and decreased to about 12 mg kg–1 h–1 after 3 to 7 days of cold storage. Cold storage increased PUMP expression as detected by Western blots and led to elevated capacities of both PUMP (44%) and CN-resistant AOX (10 times), but not the cytochrome pathway. Since we found that cold storage led to about the same mitochondrial respiration of 40 nmol O2 min–1 mg–1 attributable to each of the respective proteins, we conclude that both AOX and PUMP equally contribute to adaptation of potato tubers to cold.  相似文献

Five hundred fifty-six samples representing 24 species of small mammals (two species of marsupials and 22 rodents) were collected in Panama between February 2000 and July 2002. The samples were examined for antibodies to hantaviruses by means of enzyme-linked immunosorbent assay or immunoblot assays. The serologic results indicated that several rodent species might act as hantaviral reservoirs in Panama: Costa Rican pygmy rice rat (Oligoryzomys fulvescens costaricensis), four positive of 72 tested (5.6%); Cherrie's cane rat (Zygodontomys brevicauda cherriei), five of 108 (4.6%); Mexican deer mouse (Peromyscus mexicanus), one of 22 (5%); Mexican harvest mouse (Reithrodontomys mexicanus), one of seven (14%); Chiriquí harvest mouse (Reithrodontomys creper), one of two (50%); and Sumichrast's harvest mouse (Reithrodontomys sumichrasti), three of four (75%). Hantavirus infection in Peromyscus mexicanus and the three species of Reithrodontomys was caused by Rio Segundo hantavirus, a species of virus not previously reported from Panama. At least three hantaviruses, therefore, are known to infect populations of wild rodents in the country. However, given the total number of animals tested, the role of these rodent species in the epidemiology and epizootiology of hantavirus infections remains unclear.  相似文献

We examined the effects of human cytomegalovirus (HCMV)infection on theNa+-K+-Clcotransporter (NKCC) in a human fibroblast cell line. Using the Cl-sensitive dye MQAE, weshowed that the mock-infected MRC-5 cells express a functional NKCC.1) IntracellularCl concentration([Cl]i)was significantly reduced from 53.4 ± 3.4 mM to 35.1 ± 3.6 mMfollowing bumetanide treatment. 2)Net Cl efflux caused byreplacement of external Clwith gluconate was bumetanide sensitive.3) InCl-depleted mock-infectedcells, the Cl reuptake rate(in HCO3-free media) was reduced inthe absence of external Na+ and bytreatment with bumetanide. After HCMV infection, we found that although[Cl]iincreased progressively [24 h postexposure (PE), 65.2 ± 4.5 mM; 72 h PE, 80.4 ± 5.0 mM], the bumetanide andNa+ sensitivities of[Cl]iand net Cl uptake and losswere reduced by 24 h PE and abolished by 72 h PE. Western blots usingthe NKCC-specific monoclonal antibody T4 showed an approximatelyninefold decrease in the amount of NKCC protein after 72 h ofinfection. Thus HCMV infection resulted in the abolition of NKCCfunction coincident with the severe reduction in the amount of NKCCprotein expressed.

Invasive species can increase fire frequency and intensity, generating favorable conditions for their self-perpetuation. Mediterranean south-central Chile may be especially prone to the effects of invasive species on fire regimes because it is less adapted to fire and it contains a highly endemic flora. Teline monspessulana (L.) K. Koch (syn. Cytisus monspessulanus L.; Genista monspessulana (L.) L.A.S. Johnson) is an introduced shrub that forms monotypic stands or is present as an understory species in native forests as well as in forestry plantations. Dense T. monspessulana stands are completely destroyed by fire, generating the conditions for it seeds to germinate and establish an abundant regeneration, with up to 900 plants/m2. We report key evidence on abundance and biomass in adult stands, and patterns of seed bank and regeneration after fire in stands of T. monspessulana around the city of Concepción, Chile. We estimated living biomass in pure stands and underneath Eucalyptus plantations. In burned areas, we assessed T. monspessulana seed bank and studied regeneration patterns. We found that T. monspessulana densities reaches 52,778 plants/ha and 8.92 ton/ha in pure stands and 34,223 plants/ha and 2.31 ton/ha underneath Eucalyptus plantations. T. monspessulana generates small caliper fuel and acts as a ladder-fuel. Large soil seed banks allow for abundant regeneration after fire, with mean densities of 877,111 plants/ha, but an overall mortality of 37.2% in the first year after the fire. The high values of regeneration compared to final densities in adult stands suggest that density-dependent mortality. Our results indicate that T. monspessulana regeneration is not only favored by fires, but also that the species creates favorable conditions for intense and continuous fires, both under pure conditions, but also associated to exotic tree plantations. To understand the implications of positive feedbacks between invaders and fire, we recommend focusing in the mechanisms by which they increases fuel accumulation and fuel flammability, and how higher fire frequency and intensity favors invasive species recruitment over native species. Comprehension of this dynamics will allow for better management and control of these invasions which have major ecological, economical and social implications.  相似文献
