It has been reported that MHC class I-restricted Ag presentation persists for only a short period following infection with certain pathogens, declining in parallel with the emergence of specific CTL activity. We have examined this issue in the case of murine infection with HSV-1. We found that the period of Ag presentation capable of priming naive CD8(+) T cells is comparatively prolonged, persisting for at least 7 days after infection, and continuing despite the appearance of localized CTL activity. Ag presentation was abbreviated to 3 or 4 days postinfection by surgical excision of the inoculation site early after infection. This intervention attenuated the size of the primary CTL response, implying that prolonged presentation is necessary to drive maximal CTL expansion. Combined, these data show that, in some types of infection, CTL priming can extend well beyond the first 24-48 h after primary inoculation.  相似文献

The identity of pro- and anti-inflammatory cytokines in the neuropathogenesis of prion diseases remains undefined. Here we have investigated the role of anti-inflammatory cytokines on the progression of prion disease through the use of mice that lack interleukin-4 (IL-4), IL-10, IL-13, or both IL-4 and IL-13. Collectively our data show that among these anti-inflammatory cytokines, IL-10 plays a prominent role in the regulation of prion disease. Mice deficient in IL-10 are highly susceptible to the development of prion disease and show a markedly shortened incubation time. In addition, we have correlated cytokine gene expression in prion-inoculated IL-10(-/-) mice to wild-type-inoculated animals. Our experiments show that in the absence of IL-10 there is an early expression of tumor necrosis factor alpha (TNF-alpha). In wild-type prion-inoculated mice, the expression of TNF-alpha mRNA occurs at a later time point that correlates with the extended incubation time for terminal disease development in these animals compared to those that lack IL-10. Elevated levels of IL-13 mRNA are found at early time points in the central nervous system of prion-inoculated IL-10(-/-) mice. At terminal disease, the brains of wild-type mice inoculated with RML or ME7 are characterized by elevated levels of mRNA for the proinflammatory cytokines TNF-alpha and IL-1beta, together with the anti-inflammatory cytokines IL-10, IL-13, and transforming growth factor beta. Our data are consistent with a role for proinflammatory cytokines in the initiation of pathology during prion disease and an attempt by anti-inflammatory cytokines to regulate the ensuing, invariably fatal pathology.  相似文献

Fast- and slow-rising AMPA receptor-mediated EPSCs occur at central synapses. Fast-rising EPSCs are thought to be mediated by rapid local release of glutamate. However, two controversial mechanisms have been proposed to underlie slow-rising EPSCs: prolonged local release of transmitter via a fusion pore, and spillover of transmitter released rapidly from distant sites. We have investigated the mechanism underlying slow-rising EPSCs and the diffusion coefficient of glutamate in the synaptic cleft (Dglut) at cerebellar mossy fiber-granule cell synapses using a combination of diffusion modeling and patch-clamp recording. Simulations show that modulating Dglut has different effects on the peak amplitudes and time courses of EPSCs mediated by these two mechanisms. Slowing diffusion with the macromolecule dextran slowed slow-rising EPSCs and had little effect on their amplitude, indicating that glutamate spillover underlies these currents. Our results also suggest that under control conditions Dglut is approximately 3-fold lower than in free solution.  相似文献

Diversity of locust gut bacteria protects against pathogen invasion   总被引:2,自引:1,他引:2  
Diversity–invasibility relationships were explored in the novel context of the colonization resistance provided by gut bacteria of the desert locust Schistocerca gregaria against pathogenic bacteria. Germ‐free insects were associated with various combinations of one to three species of locust gut bacteria and then fed an inoculum of the pathogenic bacterium Serratia marcescens. There was a significant negative relationship between the resulting density of Serratia marcescens and the number of symbiotic gut bacterial species present. Likewise there was a significant inverse relationship between community diversity and the proportion of locusts that harboured Serratia. Host mortality was not negatively correlated with resistance to gut‐invasion by Serratia marcescens, although there were significantly more deaths among pathogen fed germ‐free insects than tri‐associated gnotobiotes. The outcome is consistent with the predictions of community ecology theory that species‐rich communities are more resistant to invasion than species‐poor communities.  相似文献

Open‐cut mining severely disrupts landforms and soils, preventing or impeding the restoration of preexisting or functional ecosystems because essential properties of the original soils cannot immediately or easily be reinstated. We examined the soil physicochemical and bacterial characteristics of 21 coal‐mined sites in subtropical Queensland, Australia, 3–23 years after establishment of native plant species relative to nonmined analogue sites. Soil disturbance significantly decreased total nitrogen, nitrate nitrogen, and especially total carbon (TC). The TC is projected to take 36 years to recover. Bacterial communities assessed by 16S ribosomal RNA sequencing showed greater species richness and evenness in rehabilitated as compared with nonmined soils, regardless of rehabilitation age. However, bacterial species composition was associated significantly with soil electrical conductivity, the plant density, and total stem cross‐sectional area of woody vegetation. The bacterial communities on rehabilitated sites became progressively more similar to those of nonmined analogue sites over time. This work demonstrates that if topsoils are conserved carefully during mining and supplemented by inorganic fertilizer addition, vigorous plant growth and changes in bacterial community composition can occur soon after plant establishment. This will mitigate the effects of soil disturbance and accelerate the return to the chemical and biological attributes of nonmined analogue soils.  相似文献

Cooperation can be maintained if cooperative behaviours are preferentially directed towards other cooperative individuals. Tag‐based cooperation (greenbeards) – where cooperation benefits individuals with the same tag as the actor – is one way to achieve this. Tag‐based cooperation can be exploited by individuals who maintain the specific tag but do not cooperate, and selection to escape this exploitation can result in the evolution of tag diversity. We tested key predictions crucial for the evolution of cheat‐mediated tag diversity using the production of iron‐scavenging pyoverdine by the opportunistic pathogen, Pseduomonas aeruginosa as a model system. Using two strains that produce different pyoverdine types and their respective cheats, we show that cheats outcompete their homologous pyoverdine producer, but are outcompeted by the heterologous producer in well‐mixed environments. As a consequence, co‐inoculating two types of pyoverdine producer and one type of pyoverdine cheat resulted in the pyoverdine type whose cheat was not present having a large fitness advantage. Theory suggests that in such interactions, cheats can maintain tag diversity in spatially structured environments, but that tag‐based cooperation will be lost in well‐mixed populations, regardless of tag diversity. We saw that when all pyoverdine producers and cheats were co‐inoculated in well‐mixed environments, both types of pyoverdine producers were outcompeted, whereas spatial structure (agar plates and compost microcosms), rather than maintaining diversity, resulted in the domination of one pyoverdine producer. These results suggest cheats may play a more limited role in the evolution of pyoverdine diversity than predicted.  相似文献

Capsule: Over 50% of saltmarsh breeding Common Redshank have been lost since 1985, with current conservation management having only limited success at halting these declines.

Aims: To update population size and trend estimates for saltmarsh-breeding Redshank in Britain, and to determine whether conservation management implemented since 1996 has been successful in influencing grazing intensity and Redshank population trends.

Methods: A repeat national survey of British saltmarsh was conducted in 2011 at sites previously visited in 1985 and 1996. Redshank breeding density and grazing pressure were recorded at all sites; the presence of conservation management was additionally recorded for English sites. Results from all three national surveys were used to update population size and trend estimates, and to investigate changes in grazing pressure and breeding density on sites with and without conservation management.

Results: Of the 21 431 pairs breeding on saltmarsh in 1985, 11 946 pairs remained in 2011, with the highest proportion of this population found in East Anglia. From 1985, British breeding densities declined at a rate of 1 pair km?2 year?1, representing a loss of 52.8% of breeding pairs over 26 years, although regional trends varied across different time periods. Grazing pressures did not change markedly with conservation management. Redshank declines were less severe on conservation-managed sites in East Anglia and the South of England where grazing pressures remained low, though were more severe on conservation-managed sites in the North West where heavy grazing persisted.

Conclusion: Saltmarsh-breeding Redshank declines continue and are likely to be driven by a lack of suitable nesting habitat. Conservation management schemes and site protection implemented since 1996 appear not to be delivering the grazing pressures and associated habitat conditions required by this species, particularly in the North West of England, though habitat changes may not be linked to unsuitable grazing management in all regions. An in-depth understanding of grazing practices, how conservation management guidelines could be improved, and the likely success of more long-term management solutions is needed urgently.  相似文献

Ca2+/calmodulin-dependent protein kinase II (CaMKII) forms a major component of the postsynaptic density where its functions in synaptic plasticity are well established, but its presynaptic actions are poorly defined. Here we show that CaMKII binds directly to the C-terminal domain of CaV2.1 channels. Binding is enhanced by autophosphorylation, and the kinase-channel signaling complex persists after dephosphorylation and removal of the Ca2+/CaM stimulus. Autophosphorylated CaMKII can bind the CaV2.1 channel and synapsin-1 simultaneously. CaMKII binding to CaV2.1 channels induces Ca2+-independent activity of the kinase, which phosphorylates the enzyme itself as well as the neuronal substrate synapsin-1. Facilitation and inactivation of CaV2.1 channels by binding of Ca2+/CaM mediates short term synaptic plasticity in transfected superior cervical ganglion neurons, and these regulatory effects are prevented by a competing peptide and the endogenous brain inhibitor CaMKIIN, which blocks binding of CaMKII to CaV2.1 channels. These results define the functional properties of a signaling complex of CaMKII and CaV2.1 channels in which both binding partners are persistently activated by their association, and they further suggest that this complex is important in presynaptic terminals in regulating protein phosphorylation and short term synaptic plasticity.  相似文献

The checklist comprises all species of six families of Iranian aquatic Polyphaga (Coleoptera). In total, 43 species/subspecies within the families, Georissidae (one species), Helophoridae (25 species and two subspecies), Hydrochidae (three species), Spercheidae (one species), Curculionidae (nine species) and Erirhinidae (two species) are listed for the fauna of Iran.
